Wuguchong Bioactive Peptides Promote Wound Healing via Hedgehog Pathway‑Mediated Fibroblast Modulation.

The skin undertakes multiple critical physiological functions, including barrier protection and thermoregulation. However, chronic nonhealing ulcers have emerged as a major clinical challenge. In wound repair, skin healing is a highly coordinated, time-sequential process wherein fibroblasts-key dermal cells-play irreplaceable roles in proliferation, migration, differentiation, and extracellular matrix (ECM) synthesis. When external environmental factors induce fibroblast dysfunction, this can lead to impaired epithelialization, thereby resulting in persistent wound nonclosure, which may repres

Regulatory time-kill sequencing maps antibiotic-specific transcriptional control of Escherichia coli persistence at genome scale.

Bacterial persistence enables a phenotypically tolerant subpopulation to survive lethal antibiotic exposure, contributing to the recalcitrance of chronic infections and facilitating the evolution of heritable resistance. Despite its importance, a systems-level, quantitative understanding of how transcriptional regulation shapes persistence kinetics has remained limited, largely because conventional time-kill assays are inherently low-throughput. Here, we develop Regulatory Time-Kill Sequencing (RTK-seq), a high-throughput functional genomics strategy that combines a pooled CRISPR interference

Potential roles of metabolomic alterations in neuropathic pain: Evidence from In Vivo to clinical studies.

Neuropathic pain is a chronic condition often associated with damage to the somatosensory nervous system. The exact mechanistic understanding of neuropathic pain remains elusive. Metabolomics, an analytical approach used to assess metabolic alterations in various diseases, is a promising technique that can offer mechanistic insights into neuropathic pain. It may lead to the identification of novel therapeutic targets and biomarkers. Evidence from animal studies indicates that alterations in multiple metabolic pathways, including those involving amino acids, fatty acids, glycolysis intermediate

Mediterranean Dietary Pattern in Type 2 Diabetes Management: Pathways and Clinical Evidence.

The Mediterranean diet (MedDiet) has emerged as a promising dietary strategy for the prevention and management of type 2 diabetes mellitus (T2DM). This narrative review provides a comprehensive synthesis linking the biological pathways of the MedDiet with established clinical evidence. Adherence to this traditional dietary pattern-characterized by a high intake of fiber, complex carbohydrates, antioxidants, and healthy fats-has demonstrated significant benefits in terms of glycemic control, enhanced insulin sensitivity, and overall metabolic health. Mechanistically, the review explains how the

Wolfberry glycopeptides alleviate levodopa-induced dyskinesia.

Levodopa-induced dyskinesia is a prevalent and almost inevitable complication of long-term levodopa therapy in Parkinson's disease, significantly impairing patients' quality of life. Despite its clinical significance, levodopa-induced dyskinesia remains insufficiently understood, and effective therapeutic strategies are still limited. Wolfberry glycopeptides, bioactive components extracted from Lycium barbarum L. have recently been widely reported to possess neuroimmune activities. In this study, levodopa-induced dyskinesia was induced through chronic levodopa administration in multiple Parkin

Blood eosinophil count and clinical characteristics according to etiology in chronic obstructive pulmonary disease.

BACKGROUND: Chronic obstructive pulmonary disease (COPD) is a heterogeneous condition with multiple etiologies. We evaluated whether clinical characteristics differ according to COPD etiology. METHODS: Patients with infection as the sole etiology were classified as the infection-related group (COPD-I, Group I), whereas those with any smoking-related etiology were assigned to the smoking-related group (COPD-C, Group C). RESULTS: Of 873 patients included, 41 were classified as Group I and 832 as Group C. A significantly higher proportion of patients in Group I had a blood eosinophil count < 10

Seeing is believing-on the utility of CT in phenotyping chronic obstructive pulmonary disease.

Chronic obstructive pulmonary disease (COPD) is a heterogeneous condition with complicated structural and functional impairments. For decades now, chest CT has been used to quantify various abnormalities related to COPD. More recently, with the newer data-driven approaches, biomarker development and validation have evolved rapidly. Studies now target multiple anatomical structures including lung parenchyma, the airways, the vasculature, and the fissures to better characterize COPD. This review explores the evolution of chest CT biomarkers in COPD, beginning with traditional thresholding approa

Mind the Gut: Cognitive Decline, Microbiota, and Nutrition-Related Modulators in Older Adults with Chronic Kidney Disease.

Chronic kidney disease (CKD) is a progressive condition characterized by persistent kidney abnormalities with systemic consequences. Beyond its metabolic and cardiovascular complications, CKD has been associated with structural and functional brain alterations that are particularly evident in advanced stages and in patients undergoing hemodialysis (HD). Deficits across multiple cognitive domains are frequently observed and may compromise treatment adherence, clinical management, and quality of life, yet remain largely underrecognized in clinical practice. Older adults are particularly vulnerab

IgG4-Related Disease in Childhood: Clinical Presentation, Management, and Diagnostic Challenges.

Immunoglobulin G4-related disease (IgG4-RD) is an immune-mediated fibroinflammatory disorder primarily affecting adults. The disease in pediatric age is unusual and preferentially affects adolescents. In contrast to adults, who commonly exhibit the involvement of multiple organs simultaneously or sequentially over time, young patients tend to present with a localized disease, typically affecting the orbits. Proptosis, ptosis, diplopia, and restricted eye movement may be observed in these patients. Symptoms are proteiform, and the disease is chronic and indolent with a relapsing-remitting cours

Renal tubular epithelial IGFBP7 interacts with PKM2 to drive renal lipid accumulation and fibrosis.

Renal fibrosis serves as a critical pathological mechanism driving the progression of chronic kidney disease (CKD). However, the pathogenesis and therapeutic targets involved in this process remain unclear. Interestingly, we currently found that IGFBP7 is highly expressed in tubular epithelial cells (TECs) from the fibrotic kidneys of human patients and animal models. However, their functional roles in abnormal kidney repair and renal fibrosis remain unclear. Here, we report that IGFBP7 knockout (KO) or TEC conditional KO (cKO) attenuated renal fibrosis in multiple mouse models, whereas IGFBP7

Histone deacetylases in Duchenne muscular dystrophy: a role in the mechanism of disease and a target for inhibition.

Aberrant activity of histone deacetylases (HDACs) is a pathological phenomenon in several diseases, including Duchenne muscular dystrophy (DMD). In DMD, the upregulation of HDACs is driven by the disassembly of the dystrophin-associated protein complex (DAPC), which, under normal physiological conditions, provides mechanical stability to muscle fibres and acts as a signalling hub anchoring signalling proteins and molecules to their functional sites. In dystrophic muscle, DAPC disassembly causes delocalisation of signalling proteins and, therefore, disrupts signalling pathways. Displacement of

Umbilical cord mesenchymal stromal cells in sarcopenia: benefits and strategies for enhancing efficacy.

Sarcopenia is a progressive age-related condition characterized by the decline of skeletal muscle mass and function. Its development involves multiple cellular and molecular mechanisms, including chronic inflammation, apoptosis, mitochondrial dysfunction, and impaired muscle regeneration. Human umbilical cord mesenchymal stromal cells (HUC-MSCs) have emerged as a promising therapeutic strategy due to their potent immunomodulatory properties, paracrine effects, and ability to promote tissue repair. The regenerative potential of HUC-MSCs is largely mediated by the secretion of bioactive factors

Extra-intestinal manifestations in inflammatory bowel disease: a comparative review of pediatric and adult-onset disease.

INTRODUCTION: Inflammatory Bowel Diseases (IBD) are chronic immune-mediated disorders diagnosed in both adult and pediatric populations. In recent years, there has been a significant global increase in pediatric-onset IBD, not only in Western countries but also in newly industrialized regions. Age of onset plays a crucial role in disease behavior and prognosis, with pediatric onset often associated with a more extensive and aggressive disease. As a systemic inflammatory condition, IBD can involve multiple organ systems beyond the gastrointestinal tract, thus resulting in extraintestinal manife

Actinomycosis of the Sigmoid Colon Mimicking Malignancy and Presented with Bowel Obstruction.

BACKGROUND: Actinomycosis is a chronic inflammatory condition caused by Actinomyces israelii or other Actinomycetes species, with primary abdominopelvic involvement being the most important form of the disease, especially when there is splanchnic infiltration. In cases of colonic involvement, the disease is often misdiagnosed as a neoplasm by imaging studies, and surgery is undertaken. CASE PRESENTATION: We report the case of a 48-year-old male living in a rural area in whom a computed tomography (CT) scan showed a large, irregular abdominopelvic mass infiltrating the urinary bladder, the lef
