Reversal of Calcium Dysregulation as Potential Approach for Treating Alzheimer's Disease.

Despite decades of research and effort, there is still no effective disease-modifying treatment for Alzheimer's Disease (AD). Most of the recent AD clinical trials were targeting amyloid pathway, but all these trials failed. Although amyloid pathology is a hallmark and defining feature of AD, targeting the amyloid pathway has been very challenging due to low efficacy and serious side effects. Alternative approaches or mechanisms for our understanding of the major cause of memory loss in AD need to be considered as potential therapeutic targets. Increasing studies suggest that Ca2+ dysregulatio

Suppression of hippocampal synaptic transmission by the spider toxin omega-agatoxin-IV-A.

Effects of omega-agatoxin-IV-A (AGTX) on synaptic transmissions were examined in thin transverse sections of the guinea pig hippocampus. AGTX suppressed, in a dose-dependent manner, field potentials elicited by mossy fiber stimulation in CA3 pyramidal cell layer. AGTX also suppressed field potentials elicited by perforant fiber stimulation in granular layer without marked changes in antidromic responses. These results suggest that Ca2+ entry into presynaptic terminals via P-like calcium channels is at least partly responsible for release of glutamate.

Exercise training rescues impaired H2O2-mediated vasodilation in porcine collateral-dependent coronary arterioles through enhanced K+ channel activation.

We previously reported that exercise training drives enhanced agonist-stimulated hydrogen peroxide (H2O2) levels and restores endothelium-dependent dilation via an increased reliance on H2O2 in arterioles isolated from ischemic porcine hearts. In this study, we tested the hypothesis that exercise training would correct impaired H2O2-mediated dilation in coronary arterioles isolated from ischemic myocardium through increases in protein kinase G (PKG) and protein kinase A (PKA) activation and subsequent colocalization with sarcolemmal K+ channels. Female adult Yucatan miniature swine were surgic

A K+ single channel and whole-cell clamp study on the effects of levocromakalim in guinea pig portal vein cells.

Single channel cell-attached patch and whole-cell clamp experiments on the mode of action of the K+ channel opener (KCO), levcromakalim, were performed in guinea pig isolated portal vein cells. At +20 mV (135/23 mM K+ in bath/pipette), 10 microM levcromakalim activated K+ channels with a chord conductance of 23.2 pS (K(KCO)), which were sensitive to the blocker of ATP-dependent K+ channels (K(ATP)), glibenclamide. Voltage steps from -80 mV to +20 mV activated 4-aminopyridine-sensitive K+ channels of 6.5 pS with properties of delayed rectifier K+ channels (Kv). In patches which upon a previous

Synthesis and biological evaluation of aconitine-type derivatives as potential cardiotonic agents.

Eighteen C19 aconitine-type derivatives were designed and syn-thesized with the modification of core active and strong toxicity components, such as yunaconitine (YA), mesaconitine (MA), and hypaconitine (HA) in Fuzi through hydrolysis and demethylation reactions. Their structures were confirmed by MS,1H NMR and 13C NMR. Isolated frog heart perfusion assay demonstrated that compound 18 exhibited excellent anti-heart failure activity with a cardiotonic rate of 103.14%. Its cardiotonic effect could be completely antagonized by nonselective calcium channel blockers, suggesting that the pharmacolog

Metachrony drives effective mucociliary transport via a calcium-dependent mechanism.

The mucociliary transport apparatus is critical for maintaining lung health via the coordinated movement of cilia to clear mucus and particulates. A metachronal wave propagates across the epithelium when cilia on adjacent multiciliated cells beat slightly out of phase along the proximal-distal axis of the airways in alignment with anatomically directed mucociliary clearance. We hypothesized that metachrony optimizes mucociliary transport (MCT) and that disruptions of calcium signaling would abolish metachrony and decrease MCT. We imaged bronchi from human explants and ferret tracheae using mic

Activation of PIEZO1 Attenuates Kidney Cystogenesis In Vitro and Ex Vivo.

KEY POINTS: PIEZO1 activation reduces cystogenesis: Yoda1 activates PIEZO1, raising calcium and lowering cAMP, reducing cyst growth in autosomal dominant polycystic kidney disease models. Context-dependent role of PIEZO1: PIEZO1 knockout in mice with or without polycystic kidneys does not affect cyst formation, suggesting redundancy in mechanosensitive pathways. Therapeutic potential: Findings support PIEZO1 activation as a part of combination therapy to slow cyst growth in autosomal dominant polycystic kidney disease, needing more in vivo studies. BACKGROUND: The disruption of calcium signal

Extraction and Prioritization of a Gene-Cancer-By-Survival Network Involved in Homeostasis of Intracellular Calcium Concentrations Using TCGA PANCAN Data.

Regulation of intracellular calcium concentrations, [Ca++]i is important in maintaining the viability of normal as well as cancer cells and can be mediated by tumor microenvironment. Calcium release-activated calcium channel protein (ORAI) calcium channels on the plasma membrane (PM) become physically connected by stromal interaction molecules (STIMs) to the endoplasmic reticulum (ER), on which paralogous receptors of inositol phosphate and of ryanodine are also present along with ATP2A/SERCA (sarco/endoplasmic reticulum calcium ATPases) subunits (also known as PM-ER geneset). Control of cardiac arrhythmias by calcium antagonism.

In recent years calcium channel blockers have emerged as a new class of antiarrhythmic agents for the control of certain supraventricular and ventricular arrhythmias. Electrophysiologically, they are heterogeneous but their main action is mediated through a depressant effect on the slow calcium channel in cardiac muscle. In isolated muscle, their actions are modulated by their reflex actions and by their interaction with the autonomic nervous system due to the nonocompetitive adrenergic blocking actions that some of the compounds exhibit. The major agents exerting antiarrhythmic actions are ve

Dysregulation of Neuronal Ca2+ Channel Linked to Heightened Sympathetic Phenotype in Prohypertensive States.

UNLABELLED: Hypertension is associated with impaired nitric oxide (NO)-cyclic nucleotide (CN)-coupled intracellular calcium (Ca(2+)) homeost … e hypothesized that impaired CN regulation provides a direct link to abnormal signaling of neuronal calcium channels in the SHR and that targeting cGMP can restore the channel phenotype. We found significantly larger whole-cell Ca(2+) currents from diseased neurons that were largely mediated by the N-type Ca(2+) channel (Cav2.2). Elevating cGMP restored the SHR Ca …

Therapeutic effects of TRPC4/5 antagonism in a rat model of bipolar disorder.

Bipolar disorder, a chronic neuropsychiatric condition, is characterized by alternating episodes of mania and depression. Although acute manic episodes have become increasingly manageable with the advent of third-generation neuroleptics, the primary therapeutic challenge remains the prevention of recurrences and the effective management of often refractory depressive phases. Emerging evidence implicates transient receptor potential (TRP) channels in mood regulation through their roles in calcium signaling, synaptic plasticity, and neuroinflammation. Among these, TRPC5 has gained attention as a

Mechanisms involved in corticotropin-releasing factor-induced excitation of entorhinal neurons and facilitation of glutamate release at the perforant pathway.

The entorhinal cortex (EC) is closely related to emotional control, consolidation and recall of memories, Alzheimer's disease, schizophrenia, and temporal lobe epilepsy. Corticotropin-releasing factor (CRF) is also implicated in these physiological functions and pathological disorders. Although both CRF peptide and CRF2 receptors are expressed in the EC, their roles and underlying cellular and molecular mechanisms in the EC have not been determined. Here, we found that activation of CRF2 receptors excited layer II principal neurons in the medial EC of rats and mice aged 18 to 28 days via eleva

Kv2 channels contribute to neuronal activity within the vagal afferent-nTS reflex arc.

Diversity in the functional expression of ion channels contributes to the unique patterns of activity generated in visceral sensory A-type myelinated neurons versus C-type unmyelinated neurons in response to their natural stimuli. In the present study, Kv2 channels were identified as underlying a previously uncharacterized delayed rectifying potassium current expressed in both A- and C-type nodose ganglion neurons. Kv2.1 and 2.2 appear confined to the soma and initial segment of these sensory neurons; however, neither was identified in their central presynaptic terminals projecting onto relay

RYR2 Channel Inhibition Is the Principal Mechanism of Flecainide Action in CPVT.

RATIONALE: The class Ic antiarrhythmic drug flecainide prevents ventricular tachyarrhythmia in patients with catecholaminergic polymorphic ventricular tachycardia (CPVT), a disease caused by hyperactive RyR2 (cardiac ryanodine receptor) mediated calcium (Ca) release. Although flecainide inhibits single RyR2 channels in vitro, reports have claimed that RyR2 inhibition by flecainide is not relevant for its mechanism of antiarrhythmic action and concluded that sodium channel block alone is responsible for flecainide's efficacy in CPVT. OBJECTIVE: To determine whether RyR2 block independently con

Maxi K+ channels co-localised with CFTR in the apical membrane of an exocrine gland acinus: possible involvement in secretion.

The primary secretion formed in various exocrine glands has a [K+] 2-5 times that of plasma. In this study we measured the transepithelial flux of 36Cl-, 22Na+ and 42K+ across the frog skin and applied the single-channel patch-clamp technique to the apical membrane of frog skin gland acini to investigate the pathway taken by K+ secreted by the glands. Transepithelial K+ secretion was active and was driven by a larger force than the secretion of Na+. When driving Na+ through the epithelium by clamping the transepithelial potential to 100 mV (apical solution reference), blockers of cellular secr

Prevalence of coronary calcification in relation to age, gender and risk factor profile in the insight population.

Calcium controls numerous events within the vessel wall. Permeability of the endothelium is calcium dependent. Calcium is also essential for penetration of low-density lipoprotein particles through the endothelium as well as the monocytes that travel through the subendothelial space. Other calcium-dependent processes include platelet activation and adhesion, vascular smooth muscle proliferation and migration, and synthesis of fibrous connective tissue in the subendothelial space. Current evidence indicates that calcium channel blockers retard the development of atherosclerosis in monkeys and r

Effect of protein kinase C activation on sarcoplasmic reticulum function and apparent myofibrillar Ca2+ sensitivity in intact and skinned muscles from normal and diseased human myocardium.

Protein kinase C regulates the activity of a diverse group of cellular proteins including membrane ion channel proteins. Although protein kinase C and its substrate protein have been identified in both membrane and cytosolic fractions in the heart, the physiological role of this kinase in the regulation of cardiac function remains unknown. We examined the physiological role of protein kinase C by stimulating its activity with 12-deoxyphorbol 13 isobutyrate 20 acetate (DPBA) in human trabeculae carneae. This resulted in decreased peak isometric twitch force and peak intracellular sarcoplasmic r
