The relationship between early renal status, and the resolution of vesico-ureteric reflux and bladder function at 16 months.

OBJECTIVE: To examine, in infants presenting with vesico-ureteric reflux (VUR), the relationship between the presence of initial renal abnormalities with the outcome of VUR and bladder function at 16 months of age. PATIENTS AND METHODS: The study group comprised 40 infants (32 boys) presenting consecutively (29 after prenatal detection) with VUR grade III or greater (bilateral in 29) on the initial micturating cystogram (median age 8 weeks). The initial presence of abnormal kidneys was determined from isotopic renography and/or ultrasonography. These data were correlated with the outcome of V

Comparative Bioassay-Guided Fractionation of Citrus Species: Phytochemical Characterization and Nanoformulation of a Polyphenol-Rich Leaf Fraction from Citrus aurantifolia for Skin Anti-Aging Applications.

BACKGROUND: Skin aging is driven by oxidative stress and ultraviolet (UV) exposure, leading to extracellular matrix degradation and loss of skin elasticity. This study aimed to identify the most biologically active Citrus species using a bioassay-guided approach and evaluate its potential for dermal applications. METHODS: Hydroalcohol extracts and ethyl acetate fractions of Citrus sinensis, Citrus aurantifolia, and Citrus reticulata leaves were screened for antioxidant, enzyme-inhibitory, and polyphenol content. The most active fraction was characterized by UPLC-PDA and LC-MS/MS, formulated i

Intracranial mapping identifies high-frequency cortical dynamics underlying visual discomfort.

High-contrast striped patterns near 3 cycles per degree are well established as a cause of visual discomfort and perceptual distortions, which in clinical populations can manifest as symptoms such as migraines or seizures. This sensitivity has been linked to cortical hyperexcitability, characterized by abnormally increased neural responses to visual input. Although gamma-band oscillations in visual cortex are known to reflect excitatory-inhibitory dynamics associated with pattern sensitivity, the contribution of higher-frequency neural activity and large-scale network interactions has received

Reconstructing the incidence rate and immune fraction of the population via a single snapshot survey: A case study of COVID-19 in Japan.

While the global health burden of COVID-19 continues, multifaceted epidemiological surveillance is required to monitor the epidemic's dynamics and its population-wide risk. By collecting information that is used in conventional vaccine effectiveness studies through questionnaire surveys, we proposed a simple framework using a population-wide snapshot questionnaire survey to estimate the incidence and protective effect of immunity by natural infection or vaccination against the SARS-CoV-2 JN.1 variant. Our results revealed that in Japan in February 2024, the personal risk of diagnosed infection

A predictive multiscale framework for post-translational modification-dependent peptide-MHC class I binding.

The specific binding of antigenic peptides to major histocompatibility complex class I (MHC-I) molecules is a pivotal step in adaptive immune responses. Post-translational modifications (PTMs) have been shown to profoundly regulate this process and thereby modulate T-cell recognition; however, their atomic-level mechanisms remain insufficiently understood. To address this gap, we employed all-atom molecular dynamics simulations combined with multidimensional energetic and dynamic analyses to systematically dissect PTM-dependent regulatory mechanisms across diverse antigen peptide-MHC-I (pMHC)

Epimutations in migraine and in medication overuse headache: a longitudinal study after acute medication withdrawal.

BACKGROUND: Headache disorders are among the leading causes of neurological disability worldwide, affecting over half of the global population. Recent data indicate that headache prevalence among young people increased by 23.7% between 1990 and 2021. Migraine, a highly disabling primary headache, significantly impacts daily life, yet timely and accurate diagnosis remains challenging. Many patients self-manage with analgesics, contributing to medication-overuse headache (MOH), which affects 1–2% of the general population. Emerging evidence suggests that epigenetic mechanisms, particularly DNA m

The role of actin dynamics in vesicle formation during clathrin-mediated endocytosis.

Clathrin-mediated endocytosis (CME) is an important internalization route for macromolecules, lipids, and membrane receptors in eukaryotic cells. During CME, the plasma membrane invaginates and pinches off to form clathrin-coated vesicles (CCVs). This rapid, nanoscale process involves significant changes to plasma membrane shape. We previously found heterogeneity in CCV formation: some vesicles form with simultaneous membrane bending and clathrin assembly (constant curvature), whereas others form with membrane bending after the accumulation of flat clathrin lattices (flat to curved). These arc

Continuous glucose monitoring of postprandial and 24-hour responses in individuals with spinal cord injury.

CONTEXT/OBJECTIVE: Spinal cord injury (SCI) is associated with autonomic and metabolic alterations that may impair glucose regulation. However, postprandial and 24-h glycemic dynamics in this population remain poorly understood. This study compared 5-h controlled postprandial and 24-h continuous glucose monitoring (CGM)-derived glycemic responses, including free-living monitoring, in individuals with and without SCI. DESIGN: Comparative acute experimental study. SETTING: University-based laboratory setting. PARTICIPANTS: Twenty-nine individuals with SCI and 20 individuals without SCI were i

Coenzyme Q10 ameliorates obesity by promoting white adipose tissue browning and preserving mitochondrial dynamics in ovariectomized rats fed a high-fat diet.

Estrogen deficiency caused by menopause leads to obesity in women. In obesity, excessive visceral fat accumulation induces a chronic, low-grade inflammatory response, thereby increasing the risk of cardiovascular disease, insulin resistance, and type 2 diabetes mellitus. Browning of white adipose tissue (WAT) has emerged as a promising strategy to counteract obesity and related metabolic disorders. Coenzyme Q10 (CoQ10) has been reported to reduce oxidative stress, enhance mitochondria function and improve metabolic syndrome in obese and diabetic animals and patients. In this study, we evaluate

Innate and T-cellular immune responses to sequential vaccination with chimeric hemagglutinin split influenza virus vaccines in mice.

Chimeric hemagglutinin (cHA)-based vaccines represent a promising strategy toward the development of a broadly protective universal influenza virus vaccine. Previous studies show that sequential administration of cHA split influenza vaccines stimulates broadly cross-reactive antibodies targeting the HA stalk and induces systemic and localized CD4+ T-cell responses to the HA, neuraminidase (NA), and nucleoprotein (NP). However, the exact role of the T-cellular immune response in the protection after vaccination with cHA constructs remains underinvestigated. Here we provide an in-depth character

Genomic and biogeographic patterns of endemic geckos in the Galapagos Islands reveal population structure and species delimitation on inhabited islands.

Islands offer invaluable opportunities for studying evolutionary processes due to their isolation and distinct environmental conditions. The Galapagos Islands, renowned for their rich biodiversity, host several endemic gecko species of the genus Phyllodactylus (Gekkota: Phyllodactylidae). Despite their importance derived from their specialized adaptations and their crucial role in maintaining ecosystem balance, few studies have been conducted on these geckos. This highlights the need for comprehensive genomic research to understand their evolutionary patterns and population dynamics. This stud

"Is she pregnant with Jesus?" exploring sociocultural obstacles to following medical advice in the context of stillbirth prevention in Nigeria.

BACKGROUND: Each year 182,000 babies are stillborn in Nigeria, representing nearly 10% of the annual global stillbirth burden. Imo state in south-eastern Nigeria has one of the highest levels of maternal health service access in Nigeria, yet this has not translated into good pregnancy outcomes. Many stillbirth prevention initiatives in Nigeria focus on maternal health education but empirical evidence suggests that sociocultural factors impact healthcare choices and outcomes. This study aims to explore women's and health workers' perspectives of the sociocultural barriers to following medical a

Total and Free Zinc Dynamics as Biomarkers for Neurological Impairment in Traumatic Spinal Cord Injury.

Background/Objectives: Traumatic spinal cord injury (TSCI) profoundly impacts patients by precipitating a loss of motor and sensory capabilities, largely due to oxidative stress and inflammation during the secondary injury phase. Methods: This investigation explores the diagnostic potential of zinc (Zn) and free zinc (fZn) as biomarkers by analyzing their serum concentration dynamics in 48 TSCI individuals with TSCI, with the aim of correlating these levels with neurological impairment. Serum samples collected at admission, 4 h, 9 h, 12 h, 24 h, and 3 days post-injury were analyzed for total s

Exploring barriers to accessing treatment for Lymphatic Filariasis through a socio-ecological lens in Buvuma and Napak districts, Uganda.

BACKGROUND: The World Health Organization launched the Global Programme to Eliminate Lymphatic Filariasis in 2000, which aimed at eradicating the disease by 2030. This goal depends on community mass drug administration and essential care. Despite these efforts, many rural communities still face untreated lymphatic filariasis and lack access to treatment and self-management. Using a socioecological model, this study examined barriers to treatment and support for people living with lymphatic filariasis in Uganda's Buvuma and Napak districts. METHODS: We conducted 12 key informant interviews wit

Relationships Between Lateral Ventricle Size, Cerebrospinal Fluid Dynamics, and Aqueductal Resistance in Young Healthy Adults.

BACKGROUND: Ventricular enlargement and abnormal cerebrospinal fluid (CSF) circulation are closely associated in communicating hydrocephalus (NPH), yet their causal relationship remains unclear. Studying healthy populations may help clarify these mechanisms. Existing metrics for CSF dynamics and ventricular morphology are limited by physiological variability such as heart rate and brain size, and aqueductal resistance has been little studied in healthy cohorts. PURPOSE: To quantify aqueductal resistance and two ratio-based indices-the CSF stroke volume ratio of the aqueduct to the cervical re

Evaluation of the Anti-Inflammatory Activity of Iodo Benzofuran Derivatives: In Vivo and In Silico Studies.

This study explores the anti-inflammatory potential of iodobenzofuran derivatives using both in vivo and in silico approaches. The five derivatives tested (2a-e) revealed high activity, particularly compounds 2b and 2c, which had vigorous anti-inflammatory activity, surpassing the reference drug, diclofenac (DCF). In silico analyses supported the study, including ADMET (absorption, distribution, metabolism, excretion, and toxicity) profiling, molecular docking, molecular dynamics simulations, and density functional theory (DFT). These computational results aligned with experimental outcomes, p

Review of evidence linking exposure to environmental stressors and associated alterations in the dynamics of immunosenescence (ISC) with the global increase in multiple sclerosis (MS).

Historical survey confirms that, over the latter part of the 20th century, autoimmune-based diseases, including multiple sclerosis (MS), have shown a worldwide increase in incidence and prevalence. Analytical population studies have established that the exponential rise in MS is not solely due to improvements in diagnosis and healthcare but relates to an increase in autoimmune risk factors. Harmful environmental exposures, including non-communicable social determinants of health, anthropogens and indigenous or transmissible microbes, constitute a group of causal determinants that have been clo
