The immunological profile of RC17 hESC-derived dopaminergic neural progenitor cells in vitro: Implications for the STEM-PD clinical trial.

Parkinson's disease involves the progressive loss of dopaminergic neurons, prompting clinical trials replacing cell loss with neural grafts. This includes the transplantation of pluripotent stem cell-derived mesencephalic dopaminergic neural progenitors (mesDAp), including the RC17 human embryonic stem cell (hESC)-derived cells currently under investigation in the European STEM-PD trial (NCT05635409). To assess potential immune rejection risk, we characterized RC17-mesDAp immunogenicity in vitro, comparing them to human fetal ventral mesencephalic tissue (hfVM), as successfully used in similar

Parthenogenote-derived brain unveils the critical role of paternal genome in neural development.

Genomic imprinting, an epigenetic mechanism that governs parent-of-origin-specific gene expression, is essential for mammalian development, yet its role in late-stage development remains unclear due to the lethality of parthenogenetic (Pg) embryos. Here, we establish cell replacement with parthenogenote-derived cells (CReP), a blastocyst complementation strategy that enables survival and tissue-specific contribution of Pg-derived cells. By creating tissue-specific niches in recipient embryos, CReP allows targeted incorporation of Pg-derived cells into late-stage tissues. Brain-targeted CReP sh

A malignant symbiosis: The neuro-metabolic symphony rewires the tumor microenvironment.

Emerging evidence in cancer neuroscience has established a reciprocal bidirectional loop between neural signaling and tumor metabolism as a central driver of malignant progression. This review critically evaluates bidirectional neuro-metabolic symbiosis and its profound implications for overcoming treatment refractoriness. We first analyzed the anterograde mechanism, in which peripheral nerve terminals and systemic endocrine factors impose distinct metabolic dependencies on malignant and stromal cell populations. By forcing alterations in nutrient utilization pathways, these neural cues create

Neophobia and motivation in birds: A gene expression study informed by reinforcement sensitivity theory.

Neophobia, fear of new stimuli, exhibits stable individual variation and has been suggested to have genetic underpinnings linked to specific neural pathways. In this study, we explored the relationship between neophobic responses and gene expression in Japanese quails (Coturnix japonica) within the context of Reinforcement Sensitivity Theory, which delineates the neural mechanisms governing approach and avoidance behaviors. Using a novel object task, we demonstrated that individual differences in approach motivation toward food rewards were significantly associated with the expression of the d…

Developmental regulation of cytokinetic abscission in animal stem cells.

Recent studies have revealed that cytokinetic abscission is developmentally regulated in a wide range of stem cell systems. Rather than a constitutive endpoint of cell division, abscission can be delayed, accelerated, or arrested altogether depending on developmental context. In this review, we highlight recent advances from Drosophila and mammalian models showing how stem cells modulate their link with daughter cells to regulate cell fate decisions and tissue development. Work in Drosophila germline and neural stem cells has uncovered how conserved regulators of cytokinesis, including endosom

Molecular Mechanisms and Therapeutic Targets for Pain Following Osteoporotic Vertebral Fractures.

Osteoporotic vertebral fractures (OVF) are among the most common fractures in older adults. They are strongly linked to severe pain, disability, and a reduced quality of life. Pain from OVFs often becomes chronic and differs from pain caused by fractures in normal bone. This review systematically summarizes the molecular mechanisms of OVF-related pain. It focuses on how changes in bone metabolism, inflammation, non-coding RNA regulation, and neural pathways affect each other. Major factors that drive the onset and persistence of OVF pain include increased osteoclast activity, abnormal Wnt/β-ca…

Adaptive Charge Modulation Enables Focal, Selective Spinal Cord Stimulation.

Clinical neuromodulation primarily employs near-field low frequency electrical stimulation to activate neurons in the immediate vicinity of the electrode. We introduce Adaptive Charge Modulation (ACM), a spatiotemporal, charge-balanced stimulation strategy that focuses activation at deep tissue sites distant from the stimulating contacts. We apply ACM to stimulate deep regions of the spinal cord, distant from dorsal root entry zones which are preferentially activated during low frequency stimulation (LFS). ACM applies multipolar, biphasic rectangular pulses to exceed activation thresholds in d

Theanine: a phytochemical candidate for neural protection.

Theanine, which accumulates in Camellia sinensis (L.) O. Kuntze, has demonstrated strong neuroprotective effects and other health benefits that have attracted attention. This paper reviews relevant literature published during the past 5 years, analyzing and summarizing studies focusing on the neuroprotective functions of theanine. By alleviating neuroinflammation, theanine exerts therapeutic effects on mental disorders caused by cumulative stress, ranging from mild sleep disturbances to depression induced by sleep deprivation. It has also demonstrated beneficial effects in delaying disease pro

Defective neuronal differentiation in Lowe syndrome is associated with mitochondrial dysfunction and impaired cilia-related Sonic Hedgehog signaling.

Human brain development requires tight coordination of metabolic and signaling pathways. Lowe syndrome (LS) is a recessive X-linked disorder characterized by proximal tubular renal disease, congenital cataracts, glaucoma, and neurodevelopmental delays. While LS results from mutations in the OCRL gene, which encodes an inositol polyphosphate 5-phosphatase, the cellular mechanisms driving neuronal dysfunction remain poorly understood. In this study, using patient-derived iPSC neurons, an Ocrl knockout mouse model, and an independent zebrafish OCRL-deficient model, we identified mitochondrial dys

The neuro-immune axis in cancer: from mechanisms to therapeutic opportunities.

In recent years, neural mechanisms involved in tumor initiation, progression, and immune regulation have garnered growing attention, fueling the rapid advancement of the burgeoning interdisciplinary domain of neuro-oncology. As a central component of physiological homeostasis, the neurophysiological system also exerts significant influence on the development and dynamics of the tumor microenvironment via complex neuro-immune signaling. Studies have shown that neurotransmitters, neuropeptides, and their mediated signaling pathways can directly or indirectly affect the functional state of tumor-

Circuit Reconstruction after Traumatic Spinal Cord Injury by Cellular and Pharmacological Approaches.

Extensive neuronal loss, progressive neurodegeneration, and circuit impairment are the key pathological features of spinal cord injury (SCI). The intrinsic capacity of the injured spinal cord to replenish damaged neurons and reassemble the disrupted spinal circuitry is restricted. Transplantation of exogenous neural precursor cells (NPCs) has shown promise to structurally repair the injured spinal network. However, proper maturation and integration of newly generated neurons from NPC graft within the zhost spinal circuit has remained challenging. Here, in adult female Sprague Dawley rats, we d

Insulin and epidermal signals independently shape sexually dimorphic neurite branching in C. elegans.

Sexual dimorphism in neural wiring and behavior arises from both intrinsic genetic programs and environmental cues, yet how these factors interact to shape neuronal morphogenesis remains unclear. Here, we investigate sexually dimorphic collateral branching in PVP cholinergic interneurons of Caenorhabditis elegans. In hermaphrodites, PVP branches form near the vulva and exhibit dynamic morphologies enriched with synaptic proteins for dense core vesicles but not synaptic vesicles, suggesting a role in selective neuropeptide transmission. We find that sex identity is necessary but not sufficient

Hypoxia, Psychedelics, and Terminal Lucidity: A Perspective on Neuroplasticity and Neuropsychiatric Disorders.

Hypoxia and psychedelics, despite their distinct origins, both induce altered states of consciousness and promote neuroplasticity, suggesting a shared underlying mechanism relevant to neuropsychiatric treatment and neurological recovery. Terminal lucidity, the transient resurgence of cognitive function in late-stage dementia, highlights the brain's latent capacity for rapid reorganization, a phenomenon that may be driven by transient hypoxia. Similarly, acute intermittent hypoxia and pharmacological agents like HypoxyStat, which modulate oxygen availability, have emerged as potential strategie

Updated Toolbox for Assessing Neuronal Network Reconstruction after Cell Therapy.

Cell therapy has proven to be a promising treatment for a range of neurological disorders, including Parkinson Disease, drug-resistant epilepsy, and stroke, by restoring function after brain damage. Nevertheless, evaluating the true effectiveness of these therapeutic interventions requires a deep understanding of the functional integration of grafted cells into existing neural networks. This review explores a powerful arsenal of molecular techniques revolutionizing our ability to unveil functional integration of grafted cells within the host brain. From precise manipulation of neuronal activit

Cholinergic-Induced Specific Oscillations in the Medial Prefrontal Cortex to Reverse Propofol Anesthesia.

General anesthesia is a drug-induced reversible state comprised of altered states of consciousness, amnesia, analgesia, and immobility. The medial frontal cortex (mPFC) has been discovered to modulate the level of consciousness through cholinergic and glutamatergic pathways. The optogenetic tools combined with in vivo electrophysiological recording were used to study the neural oscillatory modulation mechanisms in mPFC underlying the loss of consciousness (LOC) and emergence. We found that optogenetic activation of both cholinergic and glutamatergic neurons in the basal forebrain (BF) reversed

Optic nerve regeneration screen identifies multiple genes restricting adult neural repair.

Adult mammalian central nervous system (CNS) trauma interrupts neural networks and, because axonal regeneration is minimal, neurological deficits persist. Repair via axonal growth is limited by extracellular inhibitors and cell-autonomous factors. Based on results from a screen in vitro, we evaluate nearly 400 genes through a large-scale in vivo regeneration screen. Suppression of 40 genes using viral-driven short hairpin RNAs (shRNAs) promotes retinal ganglion cell (RGC) axon regeneration after optic nerve crush (ONC), and most are validated by separate CRISPR-Cas9 editing experiments. Cortical organoids model early brain development disrupted by 16p11.2 copy number variants in autism.

Reciprocal deletion and duplication of the 16p11.2 region is the most common copy number variation (CNV) associated with autism spectrum disorders. We generated cortical organoids from skin fibroblasts of patients with 16p11.2 CNV to investigate impacted neurodevelopmental processes. We show that organoid size recapitulates macrocephaly and microcephaly phenotypes observed in the patients with 16p11.2 deletions and duplications. The CNV dosage affects neuronal maturation, proliferation, and synapse number, in addition to its effect on organoid size. We demonstrate that 16p11.2 CNV alters the r

Thalamic Volume Is Related to Increased Anterior Thalamic Radiations in Children with Reactive Attachment Disorder.

Reactive attachment disorder (RAD) is associated with childhood maltreatment and affects approximately 1% of the general population. Recent data suggest that childhood maltreatment is associated with brain alterations in white and gray matter. However, the neural mechanisms of RAD-related brain alterations remain unknown. Herein, we evaluated the white matter pathways and gray matter volumes in 31 and 41 age-matched children with RAD and typical development (TD), respectively, by analyzing T1- and diffusion-weighted images. An increased fractional anisotropy (FA) and axial diffusivity in the a

Mitochondrial dysfunction is implicated in retinoic acid-induced spina bifida aperta in rat fetuses.

Neural tube defects (NTDs) are the most common and severe congenital malformations, which result from failure of the neural tube to close during embryonic development. The etiology of NTDs is complex, caused by interactions between genetic defects and environmental factors, but the exact mechanisms of this disease are still not fully understood. We herein employ a Seahorse Bioscience microplate-based extracellular flux (XF) analyzer to determine mitochondrial function and quantify respiratory coupling to various bioenergetic functions using specific pharmacological inhibitors of bioenergetic p
