A simple multiplex polymerase chain reaction to determine ABO blood types of rhesus macaques (Macaca mulatta).

Rhesus macaques are the most common nonhuman primate model organism used in biomedical research. Their increasingly frequent use as subjects in studies involving transplantation requires that blood and other tissue antigens of donors and recipients be compatible. We report here an easy and rapid multiplex polymerase chain reaction (PCR) to determine the ABO blood group phenotypes of rhesus macaques that can be performed with only small amounts of DNA. We phenotyped 78 individuals and found this species to exhibit the A, B and AB phenotypes in frequencies that vary by geographic region. The pro

No Lewy pathology in monkeys with over 10 years of severe MPTP Parkinsonism.

The recent knowledge that 10 years after transplantation surviving human fetal neurons adopt the histopathology of Parkinson's disease suggests that Lewy body formation takes a decade to achieve. To determine whether similar histopathology occurs in 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-primate models over a similar timeframe, the brains of two adult monkeys made parkinsonian in their youth with intermittent injections of MPTP were studied. Despite substantial nigral degeneration and increased alpha-synuclein immunoreactivity within surviving neurons, there was no evidence of Lew

The role of upstream U3 sequences in HIV-1 replication and CD4+ T cell depletion in human lymphoid tissue ex vivo.

The LTRs of all primate lentiviruses contain long U3 regions overlapping the nef gene. To assess the relevance of the modulatory U3 region for HIV-1 replication, we inactivated the T-rich region, the Polypurine tract and attachment (att) sequences in nef by silent mutations and inserted intact cis-regulatory elements just upstream of the core enhancer. These modifications severely truncated the U3 region and eliminated the nef overlap. The resulting HIV-1 mutants expressed functional Nef, replicated efficiently and caused CD4+ T cell depletion in ex vivo-infected lymphoid tissue suggesting tha

Current approaches to etiology and pathophysiology of specific phobia.

Specific phobia is a common, heterogeneous disorder whose central feature is persistent, unreasonable fear of a circumscribed object or situation. This article reviews current etiological theories and empirical data that seem likely to be important in investigating the pathophysiology of this disorder. These include conditioning, modified conditioning, and nonassociative models of phobia development, physiological response to the phobic stimulus, neuroimaging, primate, and biological challenge studies. Pathophysiological hypotheses suggested by recent research on the neurocircuitry of conditio

Antinociceptive effects of SC-39566, an opioid dipeptide arylalkylamide, in the Rhesus monkey.

This study evaluated the antinociceptive activity of an opioid dipeptide arylalkylamide, SC-39566 (2,6-dimethyl-L-tyrosinyl-D-alanyl-(3-phenyl-l-propyl)- amide), in the Rhesus monkey using the discrete trial, intermittent shock titration paradigm. I.m. administration of doses ranging from 12 to 24 mg/kg produced a dose-dependent increase in escape threshold and a dose-dependent rightward shift in the frequency distribution of responses to suprathreshold electrocutaneous stimuli. Although equivalent with respect to efficacy, SC-39566 was only one-fourth to one-sixth as potent as morphine in thi

Efficient reversion of simian sarcoma virus-transformation and inhibition of growth factor-induced mitogenesis by suramin.

Simian sarcoma virus, an acutely transforming primate retrovirus with capacity to induce gliomas and sarcomas in experimental animals, has acquired its transforming properties by transducing the cellular gene sequences that encode one of the constituent chains of platelet-derived growth factor. Suramin, a drug used in the treatment of trypanosomiasis and onchocerciasis, has previously been reported to inhibit the interaction of platelet-derived growth factor with its cell surface receptor. We show here that suramin efficiently reverts the simian sarcoma virus-induced transformed phenotype in h

Induction of AIDS-like disease in macaque monkeys with T-cell tropic retrovirus STLV-III.

The T-cell tropic retrovirus of macaque monkeys STLV-III has morphologic, growth, and antigenic properties indicating that it is related to HTLV-III/LAV, the etiologic agent of the acquired immune deficiency syndrome (AIDS) in humans. Four of six rhesus monkeys died within 160 days of STLV-III inoculation with a wasting syndrome, opportunistic infections, a primary retroviral encephalitis, and immunologic abnormalities including a decrease in T4+ peripheral blood lymphocytes. These data show that an immunodeficiency syndrome can be produced experimentally in a nonhuman primate by an agent from

Serotonergic innervation on the motoneurons in the mammalian brainstem. Light and electron microscopic immunohistochemistry.

A comparative study of serotonergic innervation on motoneurons in the brainstem of various mammals (mouse, rat, guinea pig, dog, cat and monkey) was carried out using a sensitive immunohistochemical method. Except for the extraocular muscle nuclei, the motor nuclei of the cranial nerves received rich inputs from serotonin neurons, in all species examined--rodent, carnivore and primate. The motoneurons of the monkey were innervated by varicose serotonin fibers, in a manner different from that of other species, i.e. their cell bodies and proximal dendrites were tightly encircled by a large numbe

Unique effects of infectious or inflammatory stress on fat metabolism in rats.

Infectious or inflammatory stress in the rat causes very typical functional and metabolic alterations. Among the most typical are elevation in body temperature, insulin, and glucagon and depression in the concentrations of plasma ketones and free fatty acids. These changes occur only with infectious or inflammatory stress and not with noninflammatory stresses such as femoral fracture, screen restraint, or exercise. It appears that the depression in plasma ketone bodies during infection or inflammation is closely related to the rise in plasma insulin. During infection imposed on experimentally

Chronic methanol exposure induces cognitive impairment and Alzheimer's-like pathology in rhesus monkeys.

BACKGROUND: The absence of effective animal models for sporadic Alzheimer's disease (AD) remains a pivotal barrier to therapy development. Because methanol metabolism produces endogenous formaldehyde, a neurotoxic agent linked to cognitive decline, this study investigated whether chronic, low-dose methanol exposure could recapitulate AD-like pathology and cognitive deficits in rhesus monkey, thereby establishing a nonhuman primate animal model driven by this environmental-metabolic insult. METHODS: Adult rhesus monkeys received low-concentration methanol for 9 months. Behavioral tests for cog

Kyasanur Forest Disease: Advances in Experimental Animal Models, Immune Assays, and Challenges in Vaccine Development.

Kyasanur Forest Disease (KFD) is a tick-borne viral haemorrhagic fever endemic to forested regions of South India, with case fatality rates ranging from 3% to 10% and seasonal outbreaks causing recurrent illness among at-risk populations such as forest workers and villagers. The historically used formalin-inactivated KFD vaccine showed limited effectiveness in field studies, providing only about 62% protection. This moderate level of protection highlights the need for improved vaccines that offer higher efficacy and longer-lasting immunity for populations living in endemic areas. A major chall

Laparoscopic-assisted ovariohysterectomy for pyometra with concurrent endometriosis in a captive cheetah (Acinonyx jubatus): A case report.

BACKGROUND: Reproductive tract disorders are increasingly recognized in aging captive cheetahs (Acinonyx jubatus), a species listed as vulnerable by the International Union for Conservation of Nature. Pyometra is commonly reported in felids under human care; however, open ovariohysterectomy has traditionally been used for surgical treatment. CASE DESCRIPTION: We describe the first documented case of laparoscopic-assisted ovariohysterectomy for pyometra in a cheetah, with the incidental histopathological identification of endometriosis-a condition rarely reported in non-primate species and nev

History of Health at Cayo Santiago-An Investigation of Environmental and Genetic Influences on the Skeletal Remains of the Introduced Rhesus Macaque (Macaca mulatta) Colony.

The Cayo Santiago rhesus macaque colony is a renowned primate population that has experienced significant natural and anthropogenic ecological variation in their 85-year history. Demographic and familial information is also tracked and collated for the majority of monkeys. Thus, the health history of rhesus macaques at Cayo Santiago should reflect the impacts of both environmental and genetic factors. In this study, we utilized a sample of skeletal remains comprised of 2787 individuals (1571 females, 1091 males), born between 1938 and 2017 from the derived skeletal collection of the primate co

Selective PET imaging of CXCR4 using the Al18F-labeled antagonist LY2510924.

BACKGROUND: [68Ga]PentixaFor detects C-X-C chemokine receptor type 4 (CXCR4) overexpression in various malignancies, such as multiple myeloma and non-Hodgkin lymphomas, as well as in endocrine and inflammatory disorders. This study aimed to develop an Al18F-labeled radiotracer derived from LY2510924 for CXCR4-targeted imaging, leveraging the physical and logistical advantages of fluorine-18. METHODS: We designed a CXCR4-specific radioprobe, [18F]AlF-NOTA-SC, based on LY2510924 by incorporating a triglutamate linker and NOTA chelator to enable Al18F-labeling. The in vitro CXCR4 affinity was as

Early-life behavioral features are associated with chronic emesis in rhesus macaques (Macaca mulatta).

Chronic emesis (CE) is a poorly understood condition in human and nonhuman primates that negatively impacts the quality of life. Early identification of risk factors for the development of CE is likely to improve the ability to manage CE cases successfully and is, therefore, desirable. Using a case-control study, we reviewed the necropsy records of the California National Primate Research Center and identified 24 animals with recorded CE, defined as five or more incidents of emesis in 1 month. A group of 89 healthy rhesus macaques (Macaca mulatta), comparable in age and percent time housed ind…
