Hypertension is an established risk factor for subsequent cardiovascular diseases, with Ang II (angiotensin II) playing a major role in mediating thrombotic and inflammatory abnormalities. Although T cells and IL-6 (interleukin-6) play an important role in adaptive immune responses, little is known about their role(s) in the thromboinflammatory responses associated with Ang II. Here we show using intravital microscopy coupled with the light/dye injury model that Rag-1 deficient (Rag-1-/-) and IL-6 deficient (IL-6-/-) mice are afforded protection against Ang II-induced thrombosis. Blocking IL-6

BACKGROUND: The genetic basis of muscle fat deposition in pigs is not well known. So far, we have only identified a limited number of genes involved in the absorption, transport, storage and catabolism of lipids. Such information is crucial to interpret, from a biological perspective, the results of genome-wide association analyses for intramuscular fat content and composition traits. Herewith, we have investigated how the ingestion of food changes gene expression in the gluteus medius muscle of Duroc pigs.
RESULTS: By comparing the muscle mRNA expression of fasted pigs (T0) with that of pigs

Amyloid fibrils playing a critical role in disease expression, have recently been found to exhibit the excellent mechanical properties such as elastic modulus in the order of 10 GPa, which is comparable to that of other mechanical proteins such as microtubule, actin filament, and spider silk. These remarkable mechanical properties of amyloid fibrils are correlated with their functional role in disease expression. This suggests the importance in understanding how these excellent mechanical properties are originated through self-assembly process that may depend on the amino acid sequence. Howeve

Etiopathogenesis of coronary artery ectasia (CAE), which is defined as abnormal dilatation of a segment of the coronary artery to 1.5 times of an adjacent normal coronary artery segment, is unclear. However, it is speculated that CAE develops in the atherosclerosis process through degeneration of coronary artery media layer. Our objective in this study is to compare levels of adiponectin between cases with CAE and normal coronary anatomy, and to examine whether adiponectin plays a role in CAE etiopathogenesis. The study registered a total of 66 cases, consisting of CAE cases (group 1, n = 36)

BACKGROUND: Mature spermatozoa contain thousands of mRNA transcripts. It has been recently shown that human sperm can deliver RNA into the oocyte, suggesting that mRNAs might have a role before or after fertilization. Human embryos express PSG1 (pregnancy-specific beta-1-glycoprotein 1) and HLA-E (human leukocyte antigen-E), molecules playing a role in implantation and early development. We compared PSG1 and HLA-E sperm mRNA levels in fertile and infertile men and we tested the hypothesis that these transcripts are selectively retained in the newly formed zygote.
METHODS: Real-time RT-PCR was

Therapeutic approach in osteogenic sarcoma until 1982 was connected mainly with surgery. Introduction of chemotherapy to the therapeutic protocols of osteogenic sarcoma improved the 5 years survival of patients with osteogenic sarcoma from 20% to 60-70%. The approach to surgical treatment was also changed. The principle of this treatment is usually to perform an operation to spare the limb with an intraoperative frozen section examination of bone marrow. In our Institute during the last 14 years about 300 cases were diagnosed, out of which about 200 were treated surgically, among these about 5

The trace metals represent an extremely small part of the total mass of the organism. In spite of this, these elements appear to play major roles in metabolism. At the level of the hematopoietic system, specific roles can be identified for iron, copper, zinc, and cobalt. Other trace metals appear to play less clearly defined roles in hematopoiesis but clearly are involved by virtue of a number of interactions at the level of absorption, intermediary metabolism, or modulation of toxicity. Still other trace metals, such as lithium, appear to have a therapeutic role in altering production of gran

BACKGROUND: Oral squamous cell carcinoma (OSCC) has a poor prognosis due to late-stage diagnosis, early metastasis, and resistance to treatment. OSCC is heavily influenced by its tumor microenvironment (TME), especially the stromal cell populations, which support tumor growth and contribute to recurrence and treatment resistance. Despite the tumor-stroma ratio being linked to disease progression, it is not yet part of standard staging. Moreover, it is not directly addressed by current treatments, including surgery, radiation, and chemotherapy. MAIN BODY: Among the diverse stromal components, s

Heart failure is a significant disease that threatens human life and health globally, yet its pathogenesis remains incompletely understood, posing challenges for targeted treatment. The pleiotropic transcription factor c-Myc participates in the regulation of various biological processes and has been extensively studied in the field of oncology. Recent studies have increasingly indicated that c-Myc is also associated with the development of non-oncological diseases such as heart failure, playing a role in regulating key biological processes including cardiac hypertrophy, cardiac fibrosis, energ

Parkinson's disease (PD) is an age-related neurodegenerative disorder characterized by the progressive degeneration of dopaminergic neurons, with mitochondrial dysfunction playing a critical role in its pathogenesis. As a critical organelle in eukaryotic cells, mitochondria not only serve as the central hub for energy metabolism but also play a pivotal role in regulating inflammation and cell apoptosis. However, mitochondrial damage leads to the accumulation of reactive oxygen species (ROS), oxidative stress, and abnormal aggregation of α-synuclein (α-Syn), which collectively contribute to neu

BACKGROUND: Parkinson's disease (PD) is the second most common neurodegenerative condition after Alzheimer's disease, with no cure for either. The major hurdle in identifying a therapeutic agent for such progressive disorders is their complex pathology. The current understanding is still in the cradle stage due to multifactorial protein and pathway participation, with neuroinflammation playing a major role. In the quest to link neuroinflammatory processes with cell death, pyroptosis was found to play a key role in various pathologies, including PD.
METHODOLOGY: To identify the pathological me

BACKGROUND: Ischemic stroke (IS) is a leading global cause of morbidity and mortality, with neuronal autophagy playing a pivotal role in ischemia/reperfusion (I/R) injury following stroke. Repulsive guidance molecule A (RGMa), known for its involvement in axon guidance and neuronal repair, has recently been linked to I/R injury. This study aimed to explore RGMa's role in neuronal autophagy and its therapeutic potential in mitigating I/R-induced damage using a rat model.
METHODS: A middle cerebral artery occlusion (MCAO) model was used in 108 male Sprague-Dawley rats, divided into six groups (

Immunoglobulin A (IgA) is the most abundantly produced antibody in mammals, with its secretory dimeric form playing a central role in maintaining intestinal homeostasis. Although the J chain is known to be essential for IgA transcytosis, its precise role in IgA biosynthesis and secretion is not fully understood. Here, using CRISPR/Cas9-edited J558 plasmacytoma cells, a mouse line that secretes IgA, we demonstrate that MZB1 and the J chain act sequentially to ensure proper IgA assembly. MZB1 stabilizes α-heavy chain-light chain complexes (HL complexes), thereby enabling their efficient associat

OBJECTIVE: Cellular ageing of the immune system has been suggested to play a role in the pathogenesis of rheumatoid arthritis (RA). RA onset is preceded by a preclinical phase of systemic autoimmunity without overt tissue inflammation, allowing the investigation of predisease cellular dysfunction in at-risk individuals. Lymph nodes (LNs) are crucial for (auto)antibody development, with LN fibroblasts playing a key role in regulating immune homeostasis. Here, we investigated age-associated alterations in LN fibroblast differentiation capacity during the at-risk and established stages of RA.

The connection between hypoxia and inflammation is well established at the local level, with HIF (hypoxia-inducible factor) playing a central role, which controls both the cellular response to oxygen deficiency and immune cell functioning. The literature contains extensive data regarding the roles of the transcription factors HIF, NF-κB (a key regulator of inflammatory processes) and NRF2 (a master regulator of antioxidant defense), as well as the roles of oxidative stress level and mitochondrial functional features, in the response to oxygen deficiency and the development of inflammation. As a member of the ABC transporter superfamily, ABCG2 is a half transporter that mediates the translocation of various xenobiotic substrates across cell membranes, playing an essential role in cellular detoxification. With the aim of developing a reliable in vivo model to study the role of ABCG2 and its interaction with drugs and environmental contaminants, in this study we used the CRISPR/Cas9 gene-editing technology to develop a zebrafish (Danio rerio) Abcg2a mutant line. The generated Abcg2a mutants developed normally to adulthood with no visible phenotype changes, abcg2a gene expression wa
