Palladium-Catalyzed Cross-Coupling of Aryl Bromides and Chlorides with Trimethylsilylalkynes under Mild Conditions.

Herein, we disclose a palladium-catalyzed cross-coupling of aryl bromides and chlorides with trimethylsilylalkynes under mild reaction conditions. This method utilizes commercially available and air stable palladium precatalysts and avoids the use of copper cocatalysts. Moreover, it allows for the synthesis of a wide range of disubstituted alkynes in high yields with excellent functional group tolerance. The utility of the developed method was further demonstrated via the late-stage alkynylation of pharmaceuticals and natural bioactive compounds.

Modular Synthesis of Alkenyl Sulfamates and β-Ketosulfonamides via Sulfur(VI) Fluoride Exchange (SuFEx) Click Chemistry and Photomediated 1,3-Rearrangement.

Herein, we report a synthesis of medicinally relevant β-ketosulfonamides via a photomediated 1,3-rearrangement of alkenyl sulfamates. This protocol tolerates a wide array of sensitive functional groups including alkenes, alkynes, and nitrogen-based heterocycles. Additionally, this work provides a general approach toward alkenyl sulfamates via a two-step Sulfur(VI) Fluoride Exchange (SuFEx) sequence capitalizing on SO2F2 as a linchpin to efficiently couple readily available ketones and amines without a large excess of either partner.

Synthesis of Acenes by Mechanochemical Reductive Aromatization.

A Sn-mediated mechanochemical reductive aromatization of 1,4-dihydroxy- and 1,4-dimethoxy-cyclohexadienes has been used to form 12 acenes, including derivatives of benzene, anthracene, tetracene, pentacene, and anthradithiophene. This method has been developed to overcome issues faced during the formation of electron-deficient 1,4-dihydroxy- or 1,4-dimethoxy-cyclohexadienes under homogeneous conditions. While this method does not tolerate trimethylsilyl protecting groups for alkynes, it can provide a fast and easily set up alternative to homogeneous reductive aromatization.

Synthesis of Spirocyclohexadienones via Palladium-Catalyzed Dearomatization of Dibenzoxaborins.

The development of Pd(II)-catalyzed dearomatization transformation of dibenzoxaborins with alkynes triggered by transmetalation from boron to palladium has been achieved, leading to the synthesis of spirocyclohexadienones, an important skeleton demonstrating potential biomedical utility. The [3 + 2] spiroannulation exhibits remarkable regioselectivity and broad substrate scope under mild reaction conditions. This methodology employs dibenzoxaborin as a substrate to establish the formal dearomatization of 2-phenylphenol, which poses a formidable energy barrier to the destruction of aromaticity.

Synthesis and evaluation of N-arylsulfonylated succinimides as activity-based probes.

Activity-based protein profiling (ABPP) technology has served as a powerful platform for studying proteins for more than two decades. However, the further growth of this field depends on the development of new probe structures to expand the proportion of the proteome that can be studied using these methods. Inspired by previous reports of succinimide-containing covalent inhibitors for proteases, we synthesized a panel of potential probe structures with a succinimide reactive group and a terminal alkyne tag suitable for subsequent azide-alkyne click chemistry. Members of this panel with an N-ar

Synthesis and Light-Mediated Structural Disruption of an Azobenzene-Containing Helical Poly(isocyanide).

Helical poly(isocyanide)s are an important class of synthetic polymers possessing a static helical structure. Since their initial discovery, numerous examples of these helices have been fabricated. In this contribution, the synthesis of a chiral, azobenzene (azo)-containing isocyanide monomer is reported. Upon polymerization with nickel(II) catalysts, a well-defined circular dichroism (CD) trace is obtained, corresponding to the formation of a right-handed polymeric helix. The helical polymer, dissolved in chloroform and irradiated with UV light (365 nm), undergoes a cis to trans isomerization

Microwave-assisted synthesis, anti-inflammatory and anti-proliferative activities of new maslinic acid derivatives bearing 1,5- and 1,4-disubstituted triazoles.

In this work, 40 analogs with a natural maslinic acid core (from Olea europaea L.) and various aromatic azides were synthesized. A regiospecific, facile and practical synthesis of 1,5-triazolyl derivatives by Ru(II)-catalyzed azide-alkyne cycloaddition (RuAAC), and mono-, bis- and tri-1,4-triazolyl derivatives by Cu(I)-catalyzed azide-alkyne cycloaddition (CuAAC) was described. All the reactions were assisted by microwave irradiation avoiding toxic reagents and solvents. The new products were obtained from the reaction mixture by simple purification in almost quantitative yields and the reacti

A Biocompatible Cu(I)-Catalyzed Click Chemistry Framework for Long-Term Cell Tracking.

Cellular immunotherapies have transformed oncologic treatment in recent years, yet the inability to monitor cell migration post-transfusion remains a significant barrier to their development. Here, we explore the feasibility of a non-invasive cell-tracking method utilizing Cu(I)-catalyzed click chemistry in which cells are first labeled with alkynes and, after 1-2 weeks, are selectively targeted via an azide-conjugated fluorescent probe. We developed Click1, a novel tris-Cu-chelating azide capable of a rapid, robust click reaction directly on live cell surfaces with minimal cytotoxicity. When …

Bis-MPA Janus dendrimers-prednisone conjugates against human breast cancer cells.

In this paper, we report the synthesis of Janus dendrimers using 2,2-bis(hydroxymethyl)propionic acid (bis-MPA) polyester dendrons with an azide and an alkyne group. The Janus dendrimers were obtained via azide-alkyne click reaction. The dendron and Janus dendrimers are shown to be selective against K-562 and MFC-7 cell lines. We found that the cytotoxic activity and selectivity against cancer cells of Janus prednisone-dendrimers depend on the functional groups in the dendritic arms. The conjugated dendrimer with OH as terminal groups was lower in activity against K-562 (IC50 38.98 ± 0.1  μM)

2'-Modified Thymidines with Bioorthogonal Cyclopropene or Sydnone as Building Blocks for Copper-Free Postsynthetic Functionalization of Chemically Synthesized Oligonucleotides.

The development of facile methods for conjugating relevant probes, ligands, or delivery agents onto oligonucleotides (ONs) is highly desirable both for fundamental studies in chemical biology and for improving the pharmacology of ONs in medicinal chemistry. Numerous efforts have been focused on the introduction of bioorthogonal groups onto phosphoramidite building blocks, allowing the controlled chemical synthesis of reactive ONs for postsynthetic modifications. Among these building blocks, alkyne, cyclooctynes, trans-cyclooctene, and norbornene have been proved to be compatible with automated

From Bench to Cell: A Roadmap for Assessing the Bioorthogonal "Click" Reactivity of Magnetic Nanoparticles for Cell Surface Engineering.

In this work, we report the use of bioorthogonal chemistry, specifically the strain-promoted click azide-alkyne cycloaddition (SPAAC) for the covalent attachment of magnetic nanoparticles (MNPs) on living cell membranes. Four types of MNPs were prepared, functionalized with two different stabilizing/passivation agents (a polyethylene glycol derivative and a glucopyranoside derivative, respectively) and two types of strained alkynes with different reactivities: a cyclooctyne (CO) derivative and a dibenzocyclooctyne (DBCO) derivative. The MNPs were extensively characterized in terms of physicoch…

The copper-catalyzed synthesis of (Z)-2H-naphtho[1,8-bc]thiophenes with solid emission.

In recent years, one of the main research interests of our group has been the construction of 6/5 π-systems through the regioselective conversion of parallel alkynes in naphthalene with the aim of exploiting potential fluorescent materials. Herein, the copper-catalyzed synthesis of polysubstituted (Z)-2H-naphtho[1,8-bc]thiophenes from 8-halo-1-ethynylnaphthalenes using potassium ethylxanthate as the sulfur source is reported. In this protocol, a series of thiophene-fused 6/5 π-system compounds was synthesized via copper-catalyzed Ullmann-type C(aryl)-S bond formation and the α-addition of an a…

Synthesis and Application of Ruthenium(II) Alkenyl Complexes with Perylene Fluorophores for the Detection of Toxic Vapours and Gases.

A series of new ruthenium(II) vinyl complexes has been prepared incorporating perylenemonoimide (PMI) units. This fluorogenic moiety was functionalised with terminal alkyne or pyridyl groups, allowing attachment to the metal either as a vinyl ligand or through the pyridyl nitrogen. The inherent low solubility of the perylene compounds was improved through the design of poly-PEGylated (PEG=polyethylene glycol) units bearing a terminal alkyne or a pyridyl group. By absorbing the compounds on silica, vapours and gases could be detected in the solid state. The reaction of the complexes [Ru(CH=CH-P

N-Phenylhydroxylamine-Induced Radical Relay for the Synthesis of α-Trifluoromethyl Ketones.

Herein, we report a previously unexplored N-phenylhydroxylamine-induced radical relay strategy for the synthesis of α-trifluoromethyl ketones from readily available alkynes and Togni's reagent. This protocol features a broad substrate scope, high functional group tolerance, operational simplicity, and scalability. Mechanistic studies reveal that N-phenylhydroxylamine plays a key role in facilitating the radical relay process. Furthermore, the late-stage functionalization of biologically relevant molecules highlights the synthetic utility of this method for the preparation of structurally diver…

Divergent Access to Polycyclic Fluorenofuran Systems via Palladium-Catalyzed Cascade Furan-Participated Diels-Alder Cycloaddition Reactions.

The Pd-catalyzed reaction of 1-(2-iodophenyl)-3-arylprop-2-yn-1-ones with benzene-tethered furan-ynes produces novel fused hexacyclic indenone-furan scaffolds, while the reaction with propargyl furfuryl ethers affords the pentacyclic indenone-spirodifuran system. The thiophene-containing alkynes also proceed by similar transformations. A mechanism involving the Pd-catalyzed coupling and propargylic Alder-ene reaction to generate the indenone-allene intermediate, followed by an intramolecular furan/thiophene-participated inverse electron-demand Diels-Alder reaction, is proposed.
