Isolation and molecular characterization of a new Muscovy duck parvovirus from Muscovy ducks in the USA.

Between 1997 and 1999 several cases of a new disease in Muscovy ducks were reported in Pennsylvania, USA. The cases were characterized by locomotor dysfunction, weakness, recumbency, 40 to 60% morbidity and 10 to 40% mortality. The most characteristic microscopic lesions were moderate to severe degenerative rhabodomyopathy. In order to characterize the aetiological agent, virus isolation was attempted from the spleen, liver, heart, skeletal muscle and intestine by inoculation of 14-day-old Muscovy duck embryos with tissue homogenates. Deaths occurred on the second egg passage and parvoviruses

Public health importance of Brachiola algerae (Microsporidia)--an emerging pathogen of humans.

Brachiola algerae, a parasite of Anopheles mosquitoes, has also been isolated from a human cornea, a cutaneous nodule and deep muscle tissue. All three human isolates of B. algerae are morphologically, serologically, and genetically similar to the mosquito-derived isolates including the original isolate of Vavra and Undeen. All of these isolates grew well in mammalian cell cultures at 37 degrees C and produced spores. Transmission electron microscopy revealed that all developmental stages including meronts, sporoblasts and spores were diplokaryotic and developed in direct contact with the host

Netherton syndrome: report of two Taiwanese siblings with staphylococcal scalded skin syndrome and mutation of SPINK5.

Netherton syndrome (NS) is a severe autosomal recessive ichthyosis. It is characterized by congenital ichthyosiform erythroderma, trichorrhexis invaginata, ichthyosis linearis circumflexa, atopic diathesis and frequent bacterial infections. Pathogenic mutations in SPINK5 have recently been identified in NS. SPINK5 encodes lymphoepithelial Kazal-type-related inhibitor (LEKTI), a new type of serine protease inhibitor involved in the regulation of skin barrier formation and immunity. We report two Taiwanese brothers with NS. The patients had typical manifestations of NS with an atopic diathesis a

Homozygous nonsense mutation in helix 2 of K14 causes severe recessive epidermolysis bullosa simplex.

We have studied a consanguineous family containing two children with severe, generalized epidermolysis bullosa simplex (EBS). Electron microscopy of skin biopsies from the affected individuals showed that basal keratinocytes were devoid of tonofilament bundles, although some single intermediate filament were visible. Genetic linkage analysis with the microsatellite probe D12S96 excluded the type II keratin gene cluster in this family. However, homozygosity by descent was observed with the polymorphic probes KRT9, KRT10 Ava II, and D17S1787 in both affected children, consistent with a recessive

Direct detection of ototoxicant-induced reactive oxygen species generation in cochlear explants.

The proposal that free-radical generation contributes to the ototoxicities of several chemical agents was studied utilizing electron paramagnetic resonance (EPR) spectrometry to detect directly ototoxicant-induced reactive oxygen species formation in cochlear tissue. Guinea pig cochlear explants in chelexed artificial perilymph (AP: 200 microliters) were exposed to an ototoxicant or AP for 10 min. Ototoxic agents included gentamicin sulfate (4.0 mM), kanamycin monosulfate (4.0 mM), ethacrynic acid (0.5 mM), furosemide (0.3 mM), cisplatin (0.1 mM), trimethyltin chloride (0.1 mM), and quinine HC

CD30-positive cutaneous large cell lymphomas. A comparative study of clinicopathologic and molecular features of 16 cases.

The authors have analyzed and compared the clinicopathologic and molecular features of 16 cases of large cell cutaneous lymphomas expressing CD30 antigen. Three main clinical groups were defined: (1) a group of localized skin disease (7 cases); (2) a group of multicentric skin disease (5 cases); and (3) a group of concomitant skin and extracutaneous disease. Good prognosis was associated with localized skin disease and no history of lymphoma. Interestingly, a majority of Reed Sternberg-like cells was only observed in this group (5 of 6 cases). The two other groups did not show distinctive evol

Outbreaks of gastroenteritis in elderly nursing homes and retirement facilities associated with human caliciviruses.

Eleven outbreaks of acute gastroenteritis, eight of which were in nursing homes or retirement facilities, were reported in virginia during the winter of 1993-1994. Serum samples (four outbreaks) and stool samples (two outbreaks) from involved people were tested for human calicivirus (HuCV) infection by enzyme immune assays (EIAs) using recombinant Norwalk virus (rNV) and Mexico virus (rMX) capsid antigens and reverse transcription-polymerase chain reaction (RT-PCR). Of the 31 pairs of acute and convalescent serum specimens tested, 24 had a fourfold or more titer increase to rMX and 4 responded

Flexible real-time skin dosimeter based on a thin-film copper indium gallium selenide solar cell for electron radiation therapy.

PURPOSE: Various dosimeters have been proposed for skin dosimetry in electron radiotherapy. However, one main drawback of these skin dosimeters is their lack of flexibility, which could make accurate dose measurements challenging due to air gaps between a curved patient surface and dosimeter. Therefore, the purpose of this study is to suggest a novel flexible skin dosimeter based on a thin-film copper indium gallium selenide (CIGS) solar cell, and to evaluate its dosimetric characteristics. METHODS: The CIGS solar cell dosimeter consisted of (a) a customized thin-film CIGS solar cell and (b)

Photothermal-enhanced bisphenol A degradation in iron doped alginate biochar bead activated peroxymonosulfate Fenton-like system with amplified singlet oxygen contribution.

Herein, iron-doped biochar microspheres ((AB + Fe)-C) were prepared from alginate biomass precursor, which was as a Fenton-like catalyst for peroxymonosulfate (PMS) activation to degrade bisphenol A (BPA) in water with the assistance of photothermal effect. The results indicated that under simulated sunlight irradiation, (AB + Fe)-C/PMS system exhibited remarkable enhanced BPA degradation efficiency, in which the pseudo-first-order rate constant was 2.4-fold higher in photothermal-assisted system (0.17 min-1) than photothermal-absent system (0.072 min-1). Further investigation revealed that ph

7-(Diethylamino)coumarin-3-carboxylic acid functionalized aptamers induced photoelectrochemical/fluorescence dual-mode microfluidic sensor for integrated detection of three liver cancer biomarkers.

Single-mode biosensor is prone to external interference, and detection of a single biomarker is insufficient for accurate diagnosis of liver cancer. Herein, we report a 7-(Diethylamino)coumarin-3-carboxylic acid (7DCCA) functionalized aptamers induced photoelectrochemical(PEC)/fluorescence(FL) dual-mode aptasensor integrated into a three-channel microfluidic chip for multiplexed detection of three liver cancer biomarkers: alpha-fetoprotein (AFP), carcinoembryonic antigen (CEA), and Golgi protein 73 (GP73). Piezoreflectance Spectroscopy of Optical Transitions in van der Waals Layered Crystals.

The goal of this protocol is to enable precise identification of direct optical transitions in van der Waals semiconductors by piezoreflectance spectroscopy. The method applies a small periodic strain modulation to a sample mounted on a piezoelectric transducer and detects the reflectance change with a lock-in amplifier, yielding derivative-like spectra in which a background signal is eliminated, and band-edge features become prominent. The workflow details assembly of a single-path, broadband optical setup (halogen source, monochromator, microscope optics, silicon photodiode), integration of

Mesoporous PU/PEDOT:PSS Electrodes Reveal Population-Level Stochastic Bioelectrical Dynamics in Marine Diatoms.

Understanding how microbial populations generate and modulate electrical signals remains a major technical challenge because extracellular ionic processes are inherently weak, spatially distributed and stochastic. Here, we introduce a mesoporous ultra-low-impedance PEDOT:PSS electrode that exploits volumetric ionic-electronic coupling to probe stochastic electrochemical dynamics in axenic populations of the marine model diatom Phaeodactylum tricornutum. The three-dimensional porous architecture provides a large electrochemically accessible surface area with high capacitance and low impedance,

Functional matrix engineering enables aggregate-state photophysics in tumor-responsive organic NIR-II phototheranostics.

Organic NIR-II phototheranostic nanoparticles function as molecular assemblies, making their optical performance fundamentally dependent on aggregate-state photophysics governed by intermolecular organization. However, exploiting the matrix to actively program intermolecular organization during nanoparticle assembly remains largely unexplored. Here, we establish functional matrix engineering as a practical implementation of assembly engineering, in which tumor-microenvironment-responsive prodrugs serve as multifunctional matrix components that reorganize intermolecular packing to enhance aggre

Temperature-Responsive Xylene Isomer Separation via Biomimetic Window Confinement and Defect-Engineered Nanospace in Metal-Organic Frameworks.

Biological channels achieve remarkable selectivity by amplifying subtle molecular differences through nanoscale confinement and steric gating. Inspired by this principle, we investigate the zirconium fumarate framework MOF-801, whose triangular pore windows act as biomimetic steric filters that preferentially accommodate para-xylene (PX) over its meta- (MX) and ortho- (OX) isomers. To further enhance the separation performance, we introduced structural defects that enlarge the internal nanospace while preserving the selective gating effect of the windows. The resulting material, MOF-801-132AA,

Bio-heterojunction-engineered recombinant collagen hydrogel orchestrates multimodal sterilization and immunomodulation for MRSA-infected wound healing.

Multidrug-resistant (MDR) bacterial infections, notably methicillin-resistant Staphylococcus aureus (MRSA), necessitate innovative antibiotic-free wound therapies. Here, a bio-heterojunction-integrated recombinant collagen hydrogel (CAP@MXene/CuTCPP) is designed that synergistically combines photothermal therapy (PTT), photodynamic therapy (PDT), and peroxidase-like (POD-like) activity for multimodal antibacterial action. The borate-bonded dynamically crosslinked hydrogel is composed of polyvinyl alcohol (PVA), 3-aminophenylboronic acid (APBA)-modified recombinant collagen (CF-1552), and MXene

Teaching clinical vigilance in technology-mediated care: Rethinking deterioration recognition in nursing education.

Recognizing patient deterioration remains a central concern in nursing education and patient safety. Students are commonly taught to identify abnormal observations, interpret cues, apply early warning tools, and escalate according to protocol. These approaches are necessary, but they can unintentionally present deterioration as a sequence of discrete cues waiting to be detected by an individual learner. This Contemporary Issues article argues that deterioration recognition should also be taught as a technology-mediated educational problem. The paper develops technology-mediated clinical vigila

Mitochondrial Complex I hyperactivation drives PET microplastic-induced intestinal bioenergetic collapse.

Polyethylene terephthalate (PET) microplastics (MPs) are pervasive environmental contaminants with documented human exposure, transporting across intestinal epithelium into the circulatory system and accumulating in multiple organs. This study demonstrates that digestive transformation confers a surface modification on PET MPs, altering their bio-identity and initiating a multilevel pathogenic cascade leading to intestinal dysfunction. Using physiologically relevant particles derived from commercial bottles and a simulated gastrointestinal tract, digested PET MPs inhibit the GLUT2 causing intr

Artificial Intelligence in Neurocritical Care : Multimodal Biosignal Analysis for Prognosis, Monitoring, and Future Pediatric Applications.

Neurocritical care relies on continuous assessment of neurological function and physiology under time pressure, yet bedside teams must interpret high-frequency, multimodal data that include hemodynamic waveforms, intracranial pressure (ICP), electroencephalography (EEG), near-infrared spectroscopy, neuroimaging, and electronic health record (EHR) context. Artificial intelligence (AI) and machine learning are increasingly used to fuse these biosignals, reduce interobserver variability, and generate dynamic risk estimates that can support monitoring, early warning, and prognostication. A dual signal ratio electrochemical sensor based on DNA tetrahedrons and bimetallic nanocomposites for AFB1 analysis.

Given the hepatotoxicity and widespread contamination of Aflatoxin B1 (AFB1), developing ultra-sensitive, anti-interference analytical platforms is paramount for public health. Herein, a dual signal ratio electrochemical sensing platform for accurate AFB1 analysis was constructed integrating DNA tetrahedrons (TNDA), a dynamic DNA walker, and nitrogen-doped graphene oxide-supported hollow silver-platinum bimetallic nanospheres (NGR-HP-AgPt). The NGR-HP-AgPt cavity provides a uniform microenvironment, synergistically promoting catalytic effects to significantly accelerate interfacial electron tr

Graphdiyne-PtCu hollow nanocubes-cooperated dual-engine hairpin assembler toward amplifiable electrochemical biosensing.

Developing graphdiyne-PtCu hollow nanocubes (GPtCu)-cooperated dual-engine hairpin assembler (dEHA) for amplifying electrochemical response of 17β-estradiol (E2) might be intriguing and desirable, particularly in the area of environmental analysis. Herein, we propose the first design of GPtCu-coupled dEHA amplification for constructing electrochemical biosensor sensitively responsive to E2. The modification of GPtCu and AuNPs in the electrode surface aims to enhance electronic conductivity, increase electroactive surface area, and provide more accessible Au-S binding sites for immobilizing ele
