Ocular anaphylaxis: induction by local injection of antigen.

A model of local ocular anaphylaxis has been developed in the rat. Erythema, oedema, and enhanced retention of radioiodinated rat serum albumin ([125I]-RSA) were noted in ocular adnexal tissues of immunized rats within 5 min of injection of antigen; these changes reached a maximum 15 min after antigen injection. Erythema, oedema, and retention of [125I]-RSA subsided to baseline levels 1--6 hr after challenge. A significant increase in weight of ocular adnexal tissues was seen within 15 min after challenge. The weight increase reached a maximum at 45 min and persisted through 6 hr. Weight appro…

Exercise-induced anaphylaxis as a manifestation of cholinergic urticaria.

Two patients presented with a history of exercise-induced hypotension associated with severe pruritus and either generalized urticaria or facial angioedema. Each patient was exercised under controlled conditions with use of bicycle ergometer exerciser (900 KPM/min) for 20 to 30 min at 23 degrees C. Each patients complained of generalized pruritus and then erupted in lesions typical of cholinergic urticaria. In one patient the lesions became confluent about the face and were followed by eyelid edema, lip swelling, and transient hypotension. Plasma histamine levels were elevated in each patient

Intestinal anaphylaxis in the rat as a model of food allergy.

An animal model of food allergy has been developed in which some aspects of the allergic response could be quantified and the effects of various drugs evaluated. The change in permeability of the intestinal tract of actively sensitized rats, after oral challenge with the sensitizing antigen, was the parameter measured. Rats were sensitized by injection of egg albumin and B. pertussis vaccine to induce reaginic antibody to egg albumin. Two weeks after sensitization, 125I-labelled bovine serum albumin (125I-labelled BSA) was injected intravenously, followed by oral challenge with egg albumin. Pi

Inhibition of human cutaneous anaphylaxis by cytochalasin B.

The effect of cytochalasin B (which selectively inhibits the function of cytoplasmic contractile microfilaments) on IgE-mediated hypersensitivity in human skin was studied. Cytochalasin B inhibited the antigen-induced histamine release from human skin slices sensitized passively in vitro with human reaginic serum. The magnitude of inhibition of histamine release was related to the duration of preincubation of sensitized skin slices with cytochalasin B. The effect of cytochalasin B on antigen-induced histamine release from sensitized skin was reversible, indicating that the inhibition of histam

In vitro studies of reversed anaphylaxis with rat cells.

Rabbit anti-rat γ globulin (anti-RGG) was used to bring about reversed anaphylaxis in vitro using rat mesentery, peritoneal cells, or suspensions of isolated peritoneal mast cells. Histamine and 5-hydroxytryptamine were released but not slow-reacting substance. The effect of various inhibitors on the reversed anaphylactic reaction was studied quantitatively. The reaction required calcium and was inhibited by diisopropylfluorophosphate (DFP), indole or salicylaldoxime. Magnesium dependence could not be demonstrated. Succinate, caproate and large changes in the sodium chloride concentration of t…

BLOOD CALCIUM DISTRIBUTION IN ANAPHYLAXIS IN THE GUINEA PIG.

The results above reported for total calcium and the membrane-diffusible fraction in the serum of the guinea pig, taken at various intervals during anaphylactic shock, confirm the findings of previous workers that the total calcium is essentially unchanged. There is, however, the further finding that the diffusible fraction is considerably increased over that found for the animal similarly treated but not manifesting characteristic symptoms.

STUDIES IN IMMUNITY AND ANAPHYLAXIS : THE PROTEINS OF THE KIDNEY AND LIVER.

1. The sera of rabbits injected repeatedly with the nucleoproteins, globulins, and albumins of the liver and kidney of the dog give no evidence in vitro or in vivo experiments of organ affinity. The precipitin test offers no proof of the specificity of these sera for the proteins employed as antigens. 2. The anaphylaxis reaction applied to the same proteins indicates a slight relative organ affinity but no specificity as far as the respective protein fractions are concerned. The relative organ affinity resides, rather, in the globulin and albumin fractions than in the nucleoprotein fraction. D…

Idiopathic anaphylaxis and histamine dysregulation: Revisiting pathophysiologic assumptions.

Idiopathic anaphylaxis (IA) is a diagnosis of exclusion, and the etiology remains elusive. Research on immunoglobulin E (IgE)-mediated anaphylaxis has identified histamine as the immune system mediator, but dietary histamine's potential role in IA is often overlooked. For some individuals, excessive histamine may cause IA and mimic signs and symptoms of IgE-mediated anaphylaxis.1 This review provides an overview of IA and the potential role of histamine in symptom manifestation. It aims to enhance awareness and provide nurse practitioners with insights into the complexities of managing IA, emp…

Anaphylaxis Clinical Care Pathway: Incorporating Intranasal Epinephrine (Adrenaline).

Health care providers across settings must be trained to manage anaphylaxis by (1) removing the offending allergen if still present; (2) positioning the patient with legs elevated, and if respiratory distress is present, allow the patient to sit upright with legs extended and elevated; (3) immediately administering epinephrine (adrenaline) intramuscularly (IM) or intranasally (IN) every 5 to 15 minutes for persistent anaphylaxis; and (4) optimizing airway, breathing, and cardiovascular (ABC) resuscitation. This may include supplemental oxygen, noninvasive or invasive positive-pressure ventilat…
