Effects of microgravity on muscular explosive power of the lower limbs in humans.

The maximal explosive power of the lower limbs of one astronaut has been measured before launch, and 2, 6 and 11 days after re-entry from 31 days on the MIR Station (EUROMIR '94). The subject, sitting on the carriage-seat of a Multipurpose Ergometer-Dynamometer (MED) constructed ad hoc in our laboratory, pushed maximally with both feet on two force platforms (knees angle 110 degrees). The carriage was free to move backwards on two rails inclined 20 degrees upwards. The force (F) of the lower limbs and the speed of the carriage (v) were recorded and the instantaneous mechanical power (w) was ca

Label-free blood cell separation for space health monitoring using a portable blast cell biochip.

We demonstrate a novel biomedical application of a commercial spiral microfluidic chip (Fluidic 382), originally developed for particle sorting, by repurposing it for label-free, size-based isolation of pathological blood cells, including leukemic blasts from acute myeloid leukemia (AML) samples. For the first time, we establish and validate a streamlined protocol for cell separation using Dean-driven hydrodynamic forces in a chip not originally designed for blood analysis. Using a 9-turn, 6-outlet spiral channel configuration, we achieved high-efficiency sorting of red and white blood cells f

Head-Down Tilt Bed Rest Studies as a Terrestrial Analog for Spaceflight Associated Neuro-Ocular Syndrome.

Astronauts who undergo prolonged periods of spaceflight may develop a unique constellation of neuro-ocular findings termed Spaceflight Associated Neuro-Ocular Syndrome (SANS). SANS is a disorder that is unique to spaceflight and has no terrestrial equivalent. The prevalence of SANS increases with increasing spaceflight duration and although there have been residual, structural, ocular changes noted, no irreversible or permanent visual loss has occurred after SANS, with the longest spaceflight to date being 14 months. These microgravity-induced findings are being actively investigated by the Un…

Biomarkers from automatic milking system as an indicator of subclinical acidosis and subclinical ketosis in fresh dairy cows.

The objective of this study was to determine the association between subclinical acidosis (SARA) and subclinical ketosis (SCK) with biomarkers from an automatic milking system (AMS) measuring in relation to rumination time (RT), milk yield (MY), bodyweight (BW), milk temperature, the milk fat-to-protein ratio, and the electrical conductivity of milk at the udder quarters-level which can be read in fresh dairy cows. During the course of the study, all of the fresh dairy cows (n=711) were examined according to a general clinical investigation plan. The cows were selected for 1-30 days of milk (D

Motion sickness: part III--a clinical study based on surgery of posterior fossa tumors.

Three patients who had large, benign cerebellar tumors were operated upon in the sitting position and developed symptoms referable to the temporoparieto-occipital region of the brain 24-48 h postoperatively. They consisted of dizziness, nausea, vomiting, formed and unformed hallucinations, and inversion of image or disorientation in space, some of which were experienced by some of the astronauts and cosmonauts during space flight. Such findings are not due to stimulation of the cerebellum, the site of the lesion, but must come from the cerebral hemisphere. The symptoms were believed to be caus…

Ambient Temperature Preservation Supports Recovery of Beating Capacity in Human Cardiac Organoids After Exposure to Simulated Space Stressors.

Human space exploration exposes astronauts to environmental stressors including microgravity (µg) and ionizing radiation, which may adversely affect cardiovascular health. Human induced pluripotent stem cell (hiPSC)-derived cardiac organoids, also referred to as cardioids, provide a physiologically relevant model to investigate these effects, but preservation of complex 3D tissues remains challenging. Current methods rely mainly on cryopreservation, creating variability and logistical constraints, particularly for spaceflight research. Alternative preservation strategies suitable for both grou…

Autoantibody reactome profiling reveals distinct humoral immune signatures induced by simulated microgravity, radiation and combined exposure in mice.

Microgravity (μG) and space radiation are major environmental stressors leading to immune dysfunction in astronauts, but the mechanisms governing humoral immunity and effective interventions remain unclear. In this study, we used a high-throughput autoantigen microarray derived from the AAgAtlas 1.0 database to profile humoral immune responses in mice exposed to μG and proton irradiation. We generated 70,490 autoantibody reactivity measurements and observed combined effects of radiation and μG on humoral immunity. Specifically, 38 IgM and 57 IgG autoantibody candidates showed dose-associated c…

In search of a mouse model of Spaceflight Associated Neuro-ocular Syndrome using one-carbon genetics.

Spaceflight Associated Neuro-ocular Syndrome (SANS) represents a group of ocular signs and symptoms affecting some astronauts, with genetic and metabolic factors associated with its development. This study investigated whether genetic disruption of one-carbon metabolism in a murine model, specifically via Shmt1 knockout (KO) and folate restriction (FR), could induce retinal changes mimicking SANS. Wild-type (WT) and Shmt1 KO mice were maintained on either a folate-sufficient (FS) or FR diet, and retinal thickness was assessed via optical coherence tomography at multiple time points. Red blood …

The effects of elevated CO2 on brain and ocular signal intensity with intravenous contrast MRI.

Up to 70% of astronauts develop spaceflight-associated neuro-ocular syndrome (SANS), a constellation of ocular structural changes, in microgravity; the mechanisms are unknown but have hypothesized to stem from elevated CO2, glymphatic dysfunction, and other processes. In this preliminary study, we examined whether brain and ocular tissue signal enhancement on MRI at delay following intravenous gadolinium differs when participants breathe elevated CO2 versus ambient air across two counterbalanced sessions. We scanned 12 participants at baseline using structural MRI and at 90 and 360 min followi…

A scalable, hyperstable intelligent fibre velocimeter for dynamic digitization of resistance training.

Real-time accuracy and continuous dynamic monitoring capability of devices are crucial for the scientific configuration and dynamic modulation of resistance training, such as strength training, rehabilitation and in-orbit training for astronauts. However, developing monitoring devices capable of providing real-time, accurate and dynamic quantification for high-velocity resistance training remains a notable challenge. Here, we present a scalable and hyperstable intelligent fibre velocimeter designed for the digital, real-time and dynamic monitoring of resistance training. By incorporating the f…

Differential Functional Changes in Visual Performance during Acute Exposure to Microgravity Analogue and Their Potential Links with Spaceflight-Associated Neuro-Ocular Syndrome.

BACKGROUND: Spaceflight-Associated Neuro-Ocular Syndrome (SANS) is a complex pathology threatening the health of astronauts, with incompletely understood causes and no current specific functional diagnostic or screening test. We investigated the use of the differential performance of the visual system (central vs. perimacular visual function) as a candidate marker of SANS-related pathology in a ground-based microgravity analogue. METHODS: We used a simple reaction time (SRT) task to visual stimuli, presented in the central and perimacular field of view, as a measure of the overall performance…

Impacts of psychological stress on high dose-rate radiation acute effects in a mouse experimental model.

Psychological stress affects health. Radiation workers in the medical field or astronauts living in space have possible risks of exposure to radiation, and psychological stress is considered to be easily induced in them due to activities performed in small areas or stress conditions. The impact of psychological stress on the effects of radiation was evaluated in senescence-accelerated mouse prone 10 (SAMP10) mice and ddY mice using a confrontational housing model, which makes dominant and subordinate mice in a cage live together without severe quarrel. Mice of ddY and SAMP10 have been previous…

Baoyuan jieyu formula ameliorates depression-like behaviour in rats induced by simulated long-term spaceflight composite stress through regulating MAPK and BDNF pathways.

During space flight, astronauts are exposed to various influences of extreme environments and susceptible to develop depression-like behavior. Thus, this study aims to explore the molecular biological mechanism of the cause of depression-like behavior and reveal the effect of Baoyuan Jieyu Formula (BYJYF) on ameliorating depression-like behavior. Here, rats exposed to simulated long-term spaceflight composite stress (LSCS) reduced the sucrose preference rate (P <0.01), and the time of forced swimming immobility and the number of climbing times were also reduced (P < 0.01, P < 0.001). Moreover,…

Influence of Microgravity on Apoptosis in Cells, Tissues, and Other Systems In Vivo and In Vitro.

All life forms have evolved under the constant force of gravity on Earth and developed ways to counterbalance acceleration load. In space, shear forces, buoyance-driven convection, and hydrostatic pressure are nullified or strongly reduced. When subjected to microgravity in space, the equilibrium between cell architecture and the external force is disturbed, resulting in changes at the cellular and sub-cellular levels (e.g., cytoskeleton, signal transduction, membrane permeability, etc.). Cosmic radiation also poses great health risks to astronauts because it has high linear energy transfer va…

A standardized, incremental protocol to increase human tolerance to the cross-coupled illusion.

BACKGROUND: Humans can adapt to the "Coriolis" cross-coupled illusion with repeated exposure, improving the tolerability of faster spin rate … acclimation. CONCLUSIONS: These findings are critical for future missions that may require several astronauts to be acclimated concurrently, due to resource and time constraints. Assessing feasibility of fast spin rate, short-radius centrifugation is crucial for the future of artificial gravity implementation during spaceflight.

Restricted sedation and absence of cognitive impairments after administration of intranasal scopolamine.

INTRODUCTION: Space motion sickness in astronauts during spaceflight causes significant discomfort, which might impede their functionality. Pharmacological treatment has been mainly restricted to promethazine. Transdermal and oral scopolamine have also been used in space; however, their use was reduced due to unpredictable effectiveness and side effects. Recently, intranasal scopolamine administration has gained much interest, since this route ensures fast and reliable absorption with a decreased incidence of undesirable side effects. The aim of this study was to evaluate the effect of intrana…

Dose escalation pharmacokinetics of intranasal scopolamine gel formulation.

Astronauts experience Space Motion Sickness requiring treatment with an anti-motion sickness medication, scopolamine during space missions. Bioavailability after oral administration of scopolamine is low and variable, and absorption form transdermal patch is slow and prolonged. Intranasal administration achieves faster absorption and higher bioavailability of drugs that are subject to extrahepatic, first pass metabolism after oral dosing. We examined pharmacokinetics of 0.1, 0.2, and 0.4 mg doses of the Investigational New Drug formulation of intranasal scopolamine gel (INSCOP) in 12 healthy s…

L-NAME, a nitric oxide synthase inhibitor, as a potential countermeasure to post-suspension hypotension in rats.

A large number of astronauts returning from spaceflight experience orthostatic hypotension. This hypotension may be due to overproduction of vasodilatory mediators, such as nitric oxide (NO) and prostaglandins. To evaluate the role of the NO synthase inhibitor NG-nitro-L-arginine methyl ester (L-NAME) as a countermeasure against the post-suspension reduction in mean arterial pressure (MAP), we assessed the cardiovascular responses and vascular reactivity to 7-day 30 degrees tail-suspension and a subsequent 6 hr post-suspension period in conscious rats. After a pre-suspension reading, direct MA…
