Expanding the potential of NAI-107 for treating serious ESKAPE pathogens: synergistic combinations against Gram-negatives and bactericidal activity against non-dividing cells.

OBJECTIVES: To characterize NAI-107 and related lantibiotics for their in vitro activity against Gram-negative pathogens, alone or in combination with polymyxin, and against non-dividing cells or biofilms of Staphylococcus aureus. NAI-107 was also evaluated for its propensity to select or induce self-resistance in Gram-positive bacteria. METHODS: We used MIC determinations and chequerboard experiments to establish the antibacterial activity of the examined compounds against target microorganisms. Time-kill assays were used to evaluate killing of exponential and stationary-phase cells. The eff

Restoration of bioactive lantibiotic suicin from a remnant lan locus of pathogenic Streptococcus suis serotype 2.

Lantibiotics are ribosomally synthesized, posttranslationally modified antimicrobial peptides. Their biosynthesis genes are usually organized in gene clusters, which are mainly found in Gram-positive bacteria, including pathogenic streptococci. Three highly virulent Streptococcus suis serotype 2 strains (98HAH33, 05ZYH33, and SC84) have been shown to contain an 89K pathogenicity island. Here, on these islands, we unveiled and reannotated a putative lantibiotic locus designated sui which contains a virulence-associated two-component regulator, suiK-suiR. In silico analysis revealed that the put

Evaluating the feasibility of lantibiotics as an alternative therapy against bacterial infections in humans.

Since the commercialization and ubiquitous use of antibiotics in the 20th century, there has been a steady increase in the number of reports on resistant bacteria. In recent years, this situation has become even more dramatic. The relatively slow development of new drugs, especially those with novel modes of action on target bacteria, is not paired with the rapid rate of resistance appearance. Lantibiotics form a group of antimicrobial peptides of bacterial origin with a dual mechanism of action not shared by other therapeutic compounds in use. They have a high potency to inhibit diverse (mult

Molecular studies of E. coli mercuric reductase gene (merA) and its impact on human health.

Bacterial plasmids encode resistance systems for toxic metal ions including Hg2+ functioning by energy-dependent efflux of toxic ions. The inducible mercury resistance (mer) operon encodes both a mercuric ion uptake and a detoxification enzymes. In Gram-negative bacteria especially in E. coli, a periplasmic protein, MerP, an inner- membrane transport protein, MerT, and a cytoplasmic enzyme, mercuric reductase (the MerA protein), are responsible for the transport of mercuric ions into cell and their reduction to elemental mercury, Hg0. Phytoremediation involves the use of plants to extract, det

Comparative genomics of 40 Weissella paramesenteroides strains.

Weissella strains are often detected in spontaneously fermented foods. Because of their abilities to produce lactic acid and functional exopolysaccharides as well as their probiotic traits, Weissella spp. improve not only the sensorial properties but also nutritional values of the fermented food products. However, some Weissella species have been associated with human and animal diseases. In the era of vast genomic sequencing, new genomic/genome data are becoming available to the public on daily pace. Detailed genomic analyses are due to provide a full understanding of individual Weissella spe

Niche specialization and spread of Staphylococcus capitis involved in neonatal sepsis.

The multidrug-resistant Staphylococcus capitis NRCS-A clone is responsible for sepsis in preterm infants in neonatal intensive care units (NICUs) worldwide. Here, to retrace the spread of this clone and to identify drivers of its specific success, we investigated a representative collection of 250 S. capitis isolates from adults and newborns. Bayesian analyses confirmed the spread of the NRCS-A clone and enabled us to date its emergence in the late 1960s and its expansion during the 1980s, coinciding with the establishment of NICUs and the increasing use of vancomycin in these units, respectiv

Pesticins. 3. Expression of coagulase and mechanism of fibrinolysis.

Mutational loss of pesticin I, a bacteriocin-like substance produced by Pasteurella pestis, is known to result in concomitant loss of a coagulase and fibrinolytic factor. No relationship was detected between pesticinogeny and other tested properties either associated with virulence or peculiar to P. pestis. Pesticin I was distinguished from the coagulase and fibrinolytic activities on the basis of anatomical distribution, behavior during gel filtration, and sensitivity to heat. Coagulase and the fibrinolytic factor were not differentiated by these criteria. Spontaneous suppressor mutations cau

Probiotic-Derived Antimicrobial Proteins and Peptides: Molecular Mechanisms, Clinical Applications, and Future Therapeutic Perspectives.

Increased antimicrobial resistance and the shortage of new drugs in the pipeline are driving interest in alternative approaches to treating infectious diseases. Probiotic microorganisms, particularly lactic acid bacteria, produce a variety of antimicrobial proteins and peptides (PAPs), such as bacteriocins and caseinolytic fragments, that act in ways distinct from conventional antimicrobials. This review aimed to synthesise the current evidence on PAP mechanisms, host-modulatory effects, and translational perspectives. PAPs have four distinct direct antimicrobial mechanisms: disruption of the …

Selective Protein and Lipid Cargo Sorting into Extracellular Vesicles of Lacticaseibacillus paracasei subsp. paracasei.

Extracellular vesicles (EVs) are nanoscale lipid bilayer particles that transport bioactive cargo. EVs from lactic acid bacteria show health benefits and drug-delivery potential, yet cargo encapsulation remains understudied. Here, EVs were isolated, purified, and characterized from the bacteriocin-producing strain Lacticaseibacillus paracasei subsp. paracasei YCL29, which was isolated from fermented Yucha. Proteomic and lipidomic analyses were subsequently conducted to compare the EVs with their parental L. paracasei YCL29 cells. Nanoflow cytometry indicated EVs with mean diameter 86.0 ± 18.8
