Gefitinib is a widely used fluorinated anticancer drug primarily metabolized by cytochrome P450 3A4 (CYP3A4). However, its oxidative defluorination can generate toxic quinone imine metabolites. The mechanistic origin of distinct defluorination pathways in enzymatic vs. non-enzymatic systems remains incompletely understood. Herein, we combine static DFT calculations with dynamic conformational analysis to elucidate the competitive oxidation and defluorination pathways of gefitinib. Strong hydrogen bonding stabilizes the initial hydrogen atom abstraction (HAT) pathway in both systems. However, t

The α-hydroxyketone motif is a key structural element in many pharmaceuticals and serves as a versatile intermediate in the synthesis of fine chemicals and natural products. Benzaldehyde lyase (BAL), a thiamine diphosphate (ThDP)-dependent enzyme, has emerged as a prominent biocatalyst for the synthesis of α-hydroxyketones. In recent years, significant research efforts have focused on the engineering and discovery of BAL mutants with unique or enhanced properties. In this chapter, we first review the engineering and applications of BALs in α-hydroxyketone synthesis. Next, we outline strategies

Fatty acid photodecarboxylases (FAPs) utilize blue light to convert fatty acids into Cn-1 hydrocarbons and CO2, providing a sustainable route to photobiocatalytic fuel production. Despite increasing interest, substrate-dependent phenomena-such as binding affinity, initial electron-transfer (ET) dynamics, and the influence of residues near the FAD cofactor-remain insufficiently characterized. Here, we systematically analyzed substrate binding, substrate-to-product conversion, and initial ET dynamics for three fatty acids of varying chain lengths using two cosolvent buffer systems. Laccases are multi-copper oxidase enzymes that oxidize a wide range of aromatic and non-aromatic compounds using molecular oxygen, producing water as the sole byproduct and making them attractive biocatalysts for green chemistry. However, the ability of laccases to oxidize specific substrates depends on a complex interplay of molecular structure, enzyme properties, redox potential, and environmental context, making laccase-substrate compatibility hard to predict. We apply machine learning models to pre-screen laccase-substrate combinations, streamlining experimental workflows. We evaluate four

Heroin is a highly addictive drug that exerts its primary effects through activation of μ-opioid receptors. Its principal active metabolite, 6-monoacetylmorphine (6-MAM), significantly contributes to heroin's neurological effects and acute toxicity. Current pharmacotherapies for heroin use disorder, employing opioid receptor agonist or antagonist, are often limited by risks of dependence, tolerance, and/or adverse side effects. In this context, enzyme-based therapy emerges as a promising alternative by rapidly converting drugs into inactive or less harmful metabolites in the blood. As a macrom

ω-Hydroxy and oxo acids are versatile platform chemicals, accessible via enzymes of the iron-containing alcohol dehydrogenase (FeADH) superfamily. Compared with well-characterized zinc-dependent ADHs, FeADHs producing such compounds remain poorly defined regarding assignment to genetic locus, biochemical and biophysical properties, and metal specificity. Here, we uncover two FeADHs from Clostridium viride: a highly selective 4-hydroxybutyrate dehydrogenase and a broad-spectrum 5-hydroxyvalerate dehydrogenase highly active on C5-C12 substrates, identified by purification from the native organis

Proton transfer is essential in all multi-electron transformations. Identifying the proton donor in such processes is crucial to understanding the catalytic mechanism and optimizing activity through pKa matching, which can enhance proton transfer rates. However, aqueous solutions offer multiple proton sources, which confounds clear identification of the catalytically relevant proton donor. In this work, we utilize a model hydrogenase, nickel-substituted rubredoxin (NiRd), which is capable of electrocatalytic hydrogen evolution. Prior mechanistic studies suggested a proton transfer process was

BACKGROUND: Immobilized enzyme possessing both high activity and good selectivity is important in practice. In this study, Candida antarctica lipase B (CALB) was immobilized onto the macroporous resin ADS-17 for triacylglycerol (TAG) synthesis through esterification of oleic acid and glycerol. The reaction conditions were optimized by single-factor study and orthogonal test, and the reusability of the immobilized CALB (CALB@ADS-17) was evaluated. In addition, the mechanism of lipase immobilization was studied and the catalytic mechanism of CALB@ADS-17 was investigated.
RESULTS: Oleic acid con

This study developed a pH and CO₂/N₂ dual-responsive Pickering emulsions stabilized by natural shellac nanoparticles (SNs), eliminating the need for complex synthetic stabilizers. Dynamic interfacial tension analysis reveals pH-dependent adsorption kinetics and interfacial rearrangement behavior of the SNs. Leveraging pH- or CO₂/N₂-regulated protonation-deprotonation transitions, the system could easily achieve O/W emulsion demulsification or emulsification while maintaining exceptional solvent universality and cycling stability (at least fifty cycles). The dual-responsive emulsion was further

Today, biocatalytic methodologies are well established as useful tools in green organic synthesis, since biocatalysts are mild, sustainable, and environmentally friendly catalysts which provide selectivity to the reactions they catalyse. Albumin, the most abundant protein of mammalian blood, is a versatile and mild biocatalyst in vitro. The aim of this review is to provide a perspective on the synthetic applications of albumin over the last decade. These cover transformations with a diverse chemical basis, such as additions, eliminations, and oxidations, including formation of carbon-carbon an

Carbonyl reductases (EC 1.1.1.148, CRs) make up a group of oxidoreductases that catalyze the asymmetric reduction of prochiral ketones or aldehydes to produce the corresponding chiral alcohols, which are widely used in pharmaceutical and fine chemical industries. However, challenges in improving the enzymatic activity and stability continue to hinder the broader industrial application of the biocatalysts. In this study, a novel strategy was developed to target the transition region between the rigid and flexible domains of EaSDR6 from Exiguobacterium sp. s126. B-factor analysis was used to gui

Isothiocyanates (ITCs) in cruciferous plants are natural active ingredients with cancer-preventive, anti-inflammatory, and antibacterial functions. ITCs are generated through the hydrolysis of glucosinolates by myrosinase (MYR). However, the presence of specifier proteins, especially epithionitrile-specifier proteins (ESPs), significantly impacts the yield, purification, and application of ITCs. Herein, we designed a metal-organic framework to selectively immobilize MYR by constructing a concanavalin A (ConA)-loaded ZIF-8 core. Abnormal activity level of human butyrylcholinesterase (BChE) was detected in patients with cardiovascular disease and neurodegenerative disorders, however, the specific role of BChE in the pathology of these diseases are not known yet. Homocysteine thiolactone (HTL) is a toxic thioester metabolite of homocysteine in conditions of hyperhomocysteinemia (HHcy). Experimental evidences suggest that HTL and resultant N-Hcy proteins that disrupt normal protein function, are associated with the pathology of HHcy-related complications such as cardiovascular diseases. Given the abundance of BChE in the

In this study, food-grade glutamine transaminase (TGase) was utilized for the green-catalyzed preparation of N-butyryl amino acids. For improving the reusability of the enzyme preparation, immobilized TG enzyme (94.23% immobilization rate) was prepared. Furthermore, the yield of N-butyryl phenylalanine (BP) synthesized by TGase was obtained as 20.73% by one-factor experiment. The BP synthesis yield of immobilized TGase was 95.03% of that of TGase and remained above 60% of the initial enzyme activity after five runs. The sensory evaluation and E-tongue results showed that the addition of BP sig

Biocatalytic engineering was carried out by varying monotonically the binary CNTs-silica composition and, accordingly, the physicochemical characteristics of adsorbents developed for immobilization of recombinant T. lanuginosus lipase (rPichia/lip). The adsorbents based on composite carbon-silica materials (CCSMs) were produced by impregnating finely dispersed multi-walled carbon nanotubes with silica hydrosol followed by calcination in argon at 350°C; the mass ratio of the hydrophobic and the hydrophilic components varied over a wide range. The synthesis of carbocyclic-ddA, a potent antiviral agent against hepatitis B, relies significantly on (1R,3R)-3-hydroxycyclopentanemethanol as a key intermediate. To effectively produce this intermediate, our study employed a chemoenzymatic approach. The selection of appropriate biocatalysts was based on substrate similarity, leading us to adopt the CrS enoate reductase derived from Thermus scotoductus SA-01. Additionally, we developed an enzymatic system for NADH regeneration, utilising formate dehydrogenase from Candida boidinii. This system facilitated the efficient catalysis of (S)-4-(hy

Partial acylglycerols are valued for their emulsifying and stabilizing properties, yet precise green synthesis remains challenging due to low yield and selectivity. This study aimed to elucidate the "lipase selectivity-substrate structure-product composition" relationship to enhance the yield of targeted partial acylglycerol. The results showed that lipase exhibited a greater selectivity towards fatty acids with shorter chain lengths and higher unsaturation. Hydroxyl donors also affected the esterification process, with the enzyme-acyl complex exhibiting selectivity towards hydroxyl donors as

The catechol oxidase activity of three copper/bicompartmental salen derivatives has been studied. One mononuclear, [CuL] (1), one homometallic, [Cu2L(NO3)2] (2), and one heterometallic, [CuMnL(NO3)2] (3) complexes were obtained using the ligand H2L = N,N'-bis(3-methoxysalicylidene)-1,3-propanediamine through different synthetic methods (electrochemical, chemical and solid state reaction). The structural data indicate that the metal ion disposition models the active site of type-3 copper enzymes, such as catechol oxidase. In this way, their ability to act as functional models of the enzyme has

DT-diaphorase (DT-D) is known to mainly catalyze the two-electron reduction of quinones and nitro(so) compounds. Detection of Gram-negative bacterial outer membrane vesicles (OMVs) that contain pyrogenic lipopolysaccharides (LPSs, also called endotoxins) is required for evaluating the toxic effects of analytical samples. Here, we report that DT-D has a high dephosphorylation activity: DT-D catalyzes reductive dephosphorylation of a phosphate-containing substrate in the presence of NADH. We also report that sensitive and simple OMV detection is possible with a sandwich-type electrochemical immu

The catalytic cycle of cytochrome P450 involves a change from the resting-state, water-bound, six-coordinated form (1, low-spin state) to a five-coordinated form (2, high-spin state) upon binding of a hydrophobic substrate. Here, we used a heme-thiolate model complex (SR complex) with THF as a model of nonionic H2O to address the question of whether or not coordination of nonionic water is sufficient to induce the low-spin state. Measurements of electronic absorption spectra and magnetic properties confirmed that five-coordinated SR complex has a high-spin state, and THF-bound, six-coordinated
