Exploring Carboxamide Derivatives as Promising Anticancer Agents: Design, In Vitro Evaluation, and Mechanistic Insights.

Carboxamide derivatives are a promising class of compounds in anticancer drug discovery, owing to their ability to interact with multiple oncogenic targets and their favorable pharmacological profiles. In this study, we report the design, synthesis, and biological evaluation of a series of N-substituted 1H-indole-2-carboxamides as potential anticancer agents. The synthesized compounds were assessed for antiproliferative activity using the MTT assay against MCF-7 (breast cancer), K-562 (leukemia), and HCT-116 (colon cancer) cell lines, with normal human dermal fibroblasts included as a non-canc

Novel Therapeutic Approaches and Treatment Targets for Psoriatic Arthritis.

BACKGROUND: Psoriatic Arthritis (PsA) is the most common extracutaneous manifestation of psoriasis. This chronic inflammatory arthritis is burdened with significant morbidity, leading to irreversible joint damage and disability. In recent years, a deeper understating of its pathogenesis has led to the development of several new drugs targeting different pathways. OBJECTIVES: This review aims to highlight the clinical efficacy and safety of the novel agents that have become recently available for the treatment of PsA, as well as new promising therapeutic targets that are being evaluated in cli

Screening of Nonpathogenic Fusarium Species Suppressing Fusarium Crown and Root Rot in Asparagus Using Wheat Bran as an Inoculum Carrier.

Fusarium crown and root rot (FCRR), caused by the fungus Fusarium oxysporum f. sp. asparagi, is one of the most destructive diseases affecting asparagus worldwide. Despite evidence of the suppression of FCRR by several nonpathogenic F. oxysporum sensu lato strains in laboratory experiments, biological control trials in FCRR-infested fields have not consistently demonstrated the effectiveness of these strains. Therefore, we screened nonpathogenic Fusarium spp. strains for the biological control of FCRR with the modified inoculation method considering the colonization to soil of biocontrol agent

Biological Control of Aspergillus parasiticus and Aspergillus ochraceus and Reductions in the Amount of Ochratoxin A and Aflatoxins in Bread by Selected Non-Conventional Yeast.

Aspergillus parasiticus and Aspergillus ochraceus are important pathogenic fungi that pose a serious threat because of their ability to produce mycotoxins, including ochratoxin A (OTA) and aflatoxins (AFs). The main method of reducing these pathogens is the use of chemical fungicides, though recently there has been a focus on finding biological control agents. The obtained results from this study indicate the great potential of two wild yeast strains, Aureobasidium pullulans PP3 and Saitozyma podzolicus D10, in the biological control of A. parasiticus and A. ochraceus and reductions in the amo…

Genome damage in children with classical Ehlers-Danlos syndrome - An in vivo and in vitro study.

Ehlers-Danlos syndrome (EDS) is a heritable connective tissue disorder characterized by skin hyperextensibility, abnormal wound healing, and joint hypermobility with prevalence 1:20 000. Its incidence is probably underestimated due to unknown number of subjects having mild symptoms who may have never been diagnosed through entire life time. Classical EDS is characterized by pathogenic variants of genes encoding type V collagen. The biological effects and health risks of patients with EDS exposure to low doses of ionizing radiation is poorly understood. The aim of this study was to investigate

Methods to select suitable fetal bovine serum for use in quality control assays for the detection of adventitious viruses from biological products.

Production of biological products, especially vaccines, usually requires materials derived from animals, and there are always risks that animal pathogens derived from these materials could contaminate the final products. Detection of adventitious agents is performed by quality control tests. In these biological assays, animal derived materials are also used and another problem arises, as fetal bovine serum (FBS) is used as an ingredient in tissue culture media. FBS contaminated with bovine viral diarrhea virus (BVDV) or other bovine pathogens, as well as antibodies against these pathogens may

Real-time PCR using hybridization probes for the rapid and specific identification of Francisella tularensis subspecies tularensis.

Tularemia is a plague-like infection caused by Francisella (F.) tularensis classified as a biological warfare agent. F. tularensis subsp. tularensis is the most virulent subspecies demanding rapid diagnosis. Typing systems for this fastidious bacterium to the subspecies level are laborious and time consuming. Therefore, the aim of this study was to develop a real-time PCR for the rapid and specific identification of F. tularensis subsp. tularensis. The specificity of the assay was determined using a comprehensive panel of Francisella strains, clinically relevant bacteria, and DNA preparations

Linear (-)-Zampanolide: Flexibility in Conformation-Activity Relationships.

Through an understanding of the conformational preferences of the polyketide natural product (-)-zampanolide, and the structural motifs that control these preferences, we developed a linear zampanolide analogue that exhibits potent cytotoxicity against cancer cell lines. This discovery provides a set of three structural handles for further structure-activity relationship (SAR) studies of this potent microtubule-stabilizing agent. Moreover, it provides additional evidence of the complex relationship between ligand preorganization, conformational flexibility, and biological potency. In contrast

A meta-analysis reveals that dragonflies and damselflies can provide effective biological control of mosquitoes.

Dragonfly/damselfly naiads have the potential to control mosquitoes, and indirectly the diseases they carry, due to their extensive predatio … larval stages. These results provide strong evidence that dragonflies/damselflies can be effective biological control agents of mosquitoes, and environmental planning to promote them could lower the risk of spreading mosquito-borne diseases in an environmentally friendly and cost-effective manner.

Biologics for cardiovascular diseases: from bench to bedside.

The rise of biologics, including recombinant proteins, gene therapies, and cell therapies, is reshaping the landscape of modern therapeutics, offering new strategies to address previously "undruggable" targets. Cardiovascular diseases (CVDs), the leading cause of mortality worldwide, remain inadequately managed by traditional therapies, but biologics offer a paradigm shift from symptom control to disease modification. This review provides a comprehensive analysis of biologics in cardiovascular medicine, focusing on five key biological processes: cardiac regeneration, cardiac reverse remodeling

A complete genome sequence enables efficient genetic mapping of adaptive traits in a phytoseiid predatory mite (Amblyseius andersoni).

Phytoseiid mites are important biological control agents in Integrated Pest Management (IPM), yet the genetic architecture underlying their adaptive traits remains largely unknown due to limited genomic and genetic resources. Here, we establish an integrated genetic framework for Amblyseius andersoni, generating a genome sequence and developing a high-resolution genetic mapping approach. The genome was assembled using long-read sequencing, yielding a highly contiguous assembly of approximately 202 Mb with 18,923 predicted protein-coding genes and high completeness. We then dissected resistance…

Oxidative damage and the pathogenesis of menopause related disturbances and diseases.

The postmenopausal phase of life is frequently associated in women with subjective symptoms (e.g. vasomotor) and real diseases (atherosclerosis with coronary ischemia, osteoporosis, Alzheimer-type neurodegeneration, urogenital dystrophy), which together determine the post-menopausal syndrome. Observations that oxidative damage by reactive oxygen/nitrogen species in experimental models can contribute to the pathogenesis of these disturbances stimulated research on the relationships between menopause, its endocrine deficiency, oxidative balance and the "wellness" in postmenopausal life. The conn

Anti-PD-1 Combinations: Triplets and Beyond.

PURPOSE OF REVIEW: The advent of anti-PD-1 therapy has transformed the treatment paradigm of advanced melanoma, enabling durable responses in a proportion of patients. However, the impact of both primary and acquired resistance remains a significant barrier to sustained disease control. This review explores the biological rationale and clinical progress of anti-PD-1 based combination strategies, with a particular emphasis on emerging triplet regimens and next-generation immunotherapeutic approaches designed to extend survival and overcome resistance. RECENT FINDINGS: While dual checkpoint inh

Modulation of Estrogen Receptor Activity by the Phytoalexin Tuberosin Produced from Elicited Kudzu (Pueraria lobata).

Kudzu's invasive nature has contributed to its classification as a weed, as it frequently outcompetes native plant species, leading to extensive overgrowth. Efforts to control kudzu have proven challenging, with moderate success using physical or biological agents. In this study, we evaluated the effects of two such control agents, ultraviolet C radiation and Myrothecium verrucaria, to significantly increase the production of tuberosin, a phytoalexin isoflavone. Our findings demonstrate that estrogenic activity of tuberosin is cell-type-dependent, displaying antagonist or competitive inhibitio
