BOLD-based cerebrovascular reactivity is influenced by baseline cerebral blood volume and oxygen metabolism in humans.

Cerebrovascular reactivity (CVR) is a key indicator of vascular health. It is typically assessed using a vasoactive stimulus such as hypercapnia while recording perfusion changes using blood oxygenation level dependent (BOLD) magnetic resonance imaging (MRI) data or measuring cerebral blood flow (CBF) directly, for example, with arterial spin labelling (ASL). Commonly used BOLD-CVR estimates, however, may be biased by differences in (baseline) cerebral blood volume (CBV). To investigate this, we acquired data from 50 participants at 3 T, obtaining BOLD-CVR, dynamic CBF-CVR, time-encoded CBF-CV…

Blood Coagulation Analysis in Pediatric Patients on Extracorporeal Life Support with Small-Volume Acoustic Tweezing Thromboelastometry.

Blood coagulation analysis is essential for evaluating bleeding and clotting risks, particularly in extracorporeal life support (ECLS) patients receiving preventative anticoagulant therapy, like heparin or bivalirudin. Therapy management requires continuous monitoring; however, existing methods exhibit high variability and require relatively large blood volumes, limiting their use in neonatal and pediatric care. This study assesses the ability of integrated quasistatic acoustic tweezing thromboelastometry (i-QATT), a novel noncontact technique utilizing small blood samples (6 µl), to monitor a…

Larger versus smaller red blood cell volume per transfusion in hospitalized adults, children and preterm neonates.

RATIONALE: While substantial research has evaluated the safety of hemoglobin thresholds and storage length for red blood cell (RBC) transfusion, there is minimal literature on RBC transfusion volume per transfusion event in hospitalized patients. The Choosing Wisely initiative has made a recommendation to transfuse a single unit of RBC per transfusion event, although this is based on little evidence. No recommendation has been made for pediatrics. This review evaluates the effect of the volume of RBC transfusion, administered when the decision has been made to transfuse a hospitalized patient,

Label-free microfluidic small-volume blood processing for analysis of platelet activation and neutrophil-platelet aggregates.

Platelets and leukocytes are central regulators of thrombo-inflammatory processes, and their activation states and interactions provide important insights into disease progression and therapeutic response. However, conventional blood processing relies on centrifugation-based workflows that are labor-intensive, require large sample volumes, and can induce artefactual platelet activation or disrupt native cell-cell interactions. These limitations reduce translational relevance, particularly for applications requiring small blood volumes or longitudinal sampling. Here, we present a two-stage, lab…

Eight months of marathon school training reduced blood pressure, systemic vascular resistance and extracellular water volume.

The effects of an eight-month marathon school training program on blood pressure (BP) and underlying hemodynamics were examined in 45 participants and 43 controls. Hemodynamics were recorded using whole-body impedance cardiography, radial tonometric pulse wave analysis, and third-finger BP measurements during passive head-up tilt. The mean ages were 40.9 and 42.2 years, and body mass indexes (BMI) 25.1 and 25.8 kg/m2, respectively. Marathon training decreased mean weight (-1.6 kg), fat percentage (-2.7%), and BMI (-0.5 kg/m2) and increased maximal oxygen uptake (+3.2 ml/kg/min) and insulin sen

3D Single Vessel Fractional Moving Blood Volume (3D-svFMBV): Fully Automated Tissue Perfusion Estimation Using Ultrasound.

Power Doppler ultrasound (PD-US) is the ideal modality to assess tissue perfusion as it is cheap, patient-friendly and does not require ionizing radiation. However, meaningful inter-patient comparison only occurs if differences in tissue-attenuation are corrected for. This can be done by standardizing the PD-US signal to a blood vessel assumed to have 100% vascularity. The original method to do this is called fractional moving blood volume (FMBV). We describe a novel, fully-automated method combining image processing, numerical modelling, and deep learning to estimate three-dimensional single …

Phospholipid Nanoparticles: A Novel Colloid for Blood Volume Replacement, Reanimation, and Organ Protection in Hemorrhagic Shock.

Background/Objectives: Exsanguination is a leading cause of preventable death in military and civilian settings due to extensive blood loss and hemorrhagic shock, which trigger systemic effects such as impaired tissue perfusion, hypoxia, inflammation, and multi-organ dysfunction. Standard resuscitation restores blood volume but fails to address critical aspects of hemorrhagic shock, including inflammation, coagulopathy, and reperfusion injury. To address these limitations, novel phospholipid nanoparticle (PNP)-based resuscitative fluids, VBI-S and VBI-1, were developed to modulate nitric oxide…

Quantitative Blood Volume Analysis and Hemodynamic Measures of Vascular Compliance in Patients With Worsening Heart Failure.

BACKGROUND: The role of blood volume (BV) expansion vs a change in vascular compliance in worsening heart failure (HF) remains under debate. We aimed to assess the relationship between BV and resting and stress hemodynamics in worsening HF and to further elucidate the significance of BV in cardiac decompensation. METHODS AND RESULTS: Patients with worsening HF underwent radiolabeled indicator-dilution BV analysis and cardiac catheterization. Intravascular volumes and resting/stress hemodynamics were recorded. Provocative stress maneuvers included change in systolic blood pressure (ΔSBP) from …

Building a patient blood management program in a large-volume tertiary hospital setting: Problems and solutions.

Successful implementation of a patient blood management program necessitates the collaboration of a strong organization and a multidisciplinary approach. We organized a meeting with broad participation in our center to establish a consensus for implementation of a specific patient blood management program. International and domestic experiences were shared, the importance of coordination and execution of different pillars in patient blood management were discussed, and the problems about the blood transfusion system were also investigated with the proposal for solutions. The data obtained from

Enhanced Cerebral Blood Volume under Normobaric Hyperoxia in the J20-hAPP Mouse Model of Alzheimer's Disease.

Early impairments to neurovascular coupling have been proposed to be a key pathogenic factor in the onset and progression of Alzheimer's dis … ling remained preserved under a chronic imaging preparation. Further, under hyperoxia, the baseline blood volume and saturation of all vascular compartments in the brains of J20-hAPP mice were substantially enhanced compared to WT controls, but this effect disappeared under normoxic conditions. This study highlights novel findings not previously seen in the J2 …

Quantitative measurement of blood flow volume in the major intracranial arteries by using 123i-iodoamphetamine SPECT.

PURPOSE: The aim of this study was to establish the novel automatic method to quantify blood flow volumes of the major intracranial arteries by using SPECT. METHODS: We created the vascular templates to cover the territory supplied by the major intracranial arteries. Each blood flow volume was calculated as the regional cerebral blood flow on SPECT using this template × volume size of the template. In this study, we evaluated the volume flows in 22 cerebral hemispheres with normal perfusion and 28 hemispheres with severe stenosis in the internal cerebral artery (ICA) or middle cerebral artery
