Beyond Body Weight: Biological and Clinical Determinants of Factor VIII Pharmacokinetics in Tunisian Patients.

INTRODUCTION: Inter-individual variability in factor VIII (FVIII) pharmacokinetics (PK) is well recognized in people with haemophilia A (PWH-A) and may compromise the effectiveness of prophylaxis. This variability is partially explained by clinical and biological determinants, including age, anthropometric parameters and von Willebrand factor antigen (VWF:Ag) levels. AIM: To identify the determinants of FVIII-PK in Tunisian PWH-A and assess their implications for PK-guided prophylaxis. METHODS: A prospective PK study was conducted in 25 people with severe or moderate haemophilia A enrolled b

Beyond Tobacco Prevention: The Effects of Tobacco 21 Laws on Young Adults' Body Weight.

Tobacco 21 (T21) laws effectively reduce youth tobacco use by preventing initiation. This study examines their impact on body weight among young adults aged 18-20. Using 2009-2019 Behavioral Risk Factor Surveillance System data and a two-way fixed-effects difference-in-differences (DID) design, we find limited evidence of broad weight changes in either direction across the BMI distribution. Obesity declines due to modest weight reductions concentrated near the upper BMI threshold, with no significant changes in overweight status or average BMI. Event study shows that the obesity decline emerge…

Multi-omic modelling of body mass index response to a dietary weight loss intervention.

Obesity is a multifactorial condition, and there is wide heterogeneity in responses to weight loss interventions. Although it remains challenging, modeling responses to weight loss interventions can help tailor treatments, increase weight loss success, or improve our understanding of underlying pathophysiology. We leveraged multi-omic (genetics; gut microbiota: taxonomy, inferred gene pathways and metabolite dynamics; blood metabolomics) and clinical data (e.g., lipids, blood glucose) from a 12-month behavioral weight loss trial of adults (n = 150) with overweight/obesity, to forecast longitud

Body weight and mortality in Takotsubo syndrome: insights from the International Takotsubo (InterTAK) Registry.

AIMS: The obesity paradox has been described in different cardiovascular conditions. Data on the association between obesity and outcomes in patients with Takotsubo syndrome (TTS) are lacking. The aim of this study was to determine the relationship between body weight and mortality in TTS patients. METHODS: Patients enrolled in the International Takotsubo Registry from January 2011 to July 2021 and with available data on body mass index (BMI) were included in the analysis. Patients were stratified according to BMI (underweight, <18.5 kg/m2; normal weight, 18.5-24.9 kg/m2; overweight, 25.0-29.…

POMC neuron METTL14/m6A/YTHDC1/YTHDF2 pathways safeguard energy balance, body weight, and metabolism.

METTL14 mediates N6-methyladenosine (m6A) RNA modification, while YTHDC1 and YTHDF2 specifically bind m6A-methylated RNA to regulate RNA fate. POMC neurons constitute the core of the central melanocortin circuit, and POMC deficiency causes obesity in both mice and humans. However, how m6A-based epitranscriptomics regulates melanocortin circuit function remains unclear. Here, we generated and characterized POMC neuron-specific knockout mice lacking Mettl14 (Mettl14ΔPOMC), Ythdc1 (Ythdc1ΔPOMC), or Ythdf2 (Ythdf2 ΔPOMC). Mettl14ΔPOMC and Ythdc1ΔPOMC mice develop hyperphagia, obesity, glucose into

Persistent body-weight change on achalasia and peroral endoscopic myotomy: a multicenter cohort study.

BACKGROUND: The distribution of body weight in patients with achalasia and after peroral endoscopic myotomy (POEM) has not been investigated. The role of body weight assessment after treatment remains unclear. METHODS: Using the multicenter achalasia cohort, the frequency of underweight (body mass index [BMI] < 18.5 kg/m2) and overweight (BMI ≥ 25.0 kg/m2) and their associated clinical characteristics were analyzed. After POEM, risk factors for insufficient- (underweight persistently) and excessive- (responded to overweight) weight gainers were investigated. The correlation between BMI-increa…

Unusual body weight loss due to primary hyperparathyroidism: A case study with literature review.

Brown tumors (osteitis fibrosa cystica) are rare pathognomonic signs that occur in patients with primary hyperparathyroidism (PHPT). Brown tumors can exist in multiple bones and can easily be misdiagnosed as a metastatic tumor or multiple myeloma. It is also localized in the forearm, humerus, and leg. The symptoms of hypercalcemia, pathologic fracture, and bodyweight loss may increase the diagnostic difficulty of brown tumors because multiple myeloma and bone metastasis also show the same symptoms. We studied a 68-year-old woman who had experienced unusual bodyweight loss in the past 6 months

Distinct Reproductive Phenotypes Segregate With Differences in Body Weight in Adolescent Polycystic Ovary Syndrome.

INTRODUCTION: Polycystic ovary syndrome (PCOS) is a heterogenous clinical syndrome defined by hyperandrogenism and irregular menses. In adult women with PCOS, discrete metabolic and reproductive subgroups have been identified. We hypothesize that distinct phenotypes can be distinguished between adolescent girls who are lean (LN-G) and girls with obesity (OB-G) at the time of PCOS diagnosis. METHODS: Data were extracted from the CALICO multisite PCOS database. Clinical data collected at the time of diagnosis were available in 354 patients (81% with obesity) from 7 academic centers. Patients wi

How cardiologists can manage excess body weight and related cardiovascular risk. An expert opinion.

Obesity is an important independent cardiovascular (CV) risk factor and a chronic inflammatory disease related to the development of insulin resistance, type 2 diabetes, dyslipidaemia, coronary artery disease, hypertension, heart failure, atrial fibrillation and obstructive sleep apnoea. Body Mass Index (BMI) values >27 kg/m2 are associated with an exponential increase in the risk for Major Adverse Cardiac Events (MACE). On the other hand, weight reduction can significantly reduce metabolic, CV and oncological risk. Orlistat, bupropion/naltrexone, liraglutide and semaglutide, combined with lif
