Escin alleviates stress-induced intestinal dysfunction to protect brain injury by regulating the gut-brain axis in ischemic stroke rats.

Hyperactivity of HPA axis results in intestinal dysfunction, which may play a role in brain injury caused by ischemic stroke (IS). Escin shows a neuroprotective effect but it may not penetrate blood brain barrier (BBB). Previous work in our laboratory showed that escin ameliorated intestinal injury in animals. The aim of this study is to investigate whether escin attenuates brain injury by improving intestinal dysfunction in middle cerebral artery occlusion (MCAO) rats, to mimic IS. MCAO rats and lipopolysaccharides (LPS)-induced Caco-2 cells were used to evaluate the effects of escin in vivo

Bi-directional elucidation of Lactiplantibacillus plantarum (RTA 8) intervention on the pathophysiology of gut-brain axis during Salmonella brain infection.

BACKGROUND: There have been reports of patients suffering from typhoid fever, particularly those involving infants and immunocompromised patients, which at times present with Salmonella induced brain infection. Although rare, it has frequently been associated with adverse neurological complications and increased mortality. In this context, the gut-brain axis, involving two-way communication between the gut and the brain, holds immense significance as various gut ailments have been associated with psychiatric complications. In turn, several neurodegenerative diseases have been associated with a

Selective brain cooling achieves peripheral organs protection in hemorrhagic shock resuscitation via preserving the integrity of the brain-gut axis.

Background: Although whole-body cooling has been reported to improve the ischemic/reperfusion injury in hemorrhagic shock (HS) resuscitation, it is limited by its adverse reactions following therapeutic hypothermia. HS affects the experimental and clinical bowel disorders via activation of the brain-gut axis. It is unknown whether selective brain cooling achieves beneficial effects in HS resuscitation via preserving the integrity of the brain-gut axis. Methods: Male Sprague-Dawley rats were bled to hypovolemic HS and resuscitated with blood transfusion followed by retrograde jugular vein flush…

Bioactive compounds and exercise in aging and neurodegeneration: mechanistic insights from the gut-brain-metabolic axis.

Aging and neurodegenerative disorders are associated with impaired hippocampal plasticity, yet existing literature largely examines exercise, nutrition, or metabolic regulation in isolation. This review synthesizes emerging evidence supporting an integrative neuro-nutritional-metabolic framework in which bioactive compounds and physical exercise converge to modulate hippocampal neurogenesis, synaptic plasticity, and cognitive resilience. Recent investigative efforts elucidate the neuro-nutritional-metabolic axis as a pivotal interface that integrates bioactive compounds derived from diet, syst

Microbiome-gut-brain axis modulation for the management of Parkinson's disease: Emerging mechanisms and translational insights.

Parkinson's disease (PD) is a progressive neurodegenerative disease that is associated with the loss of dopaminergic neurons and the formation of α-synuclein aggregates in the brain. Changes in the gut microbiota have been closely associated with PD pathophysiology and may contribute to disease-related processes through neuroinflammatory, metabolic, and immune-mediated mechanisms. This review explores the connection between the microbiome-gut-brain axis and Parkinson's disease and highlights new microbiome-based therapeutic interventions to help manage the disease. The extensive literature rev

From intake to impact: dietary modulation of the gut-brain axis across health and neurological disease.

UNLABELLED: Sinusoidal obstruction syndrome (SOS) is a distinctive and potentially fatal form of hepatic injury that mainly occurs after hematopoietic-stem cell transplantation but also due to many other conditions including drug or toxin exposure. Recently, immune checkpoint inhibitors (ICIs) have revolutionised the treatment of many solid organ malignancies. Furthermore, as their use has become more widespread, rare toxicities have emerged. The difficulty lies in diagnosing these unusual toxicities with an incidence of as low as less than 1% hence defined as SOS. The development of the disea

The microbiota-gut-brain axis: novel mechanisms and therapeutic frontiers in obesity and type 2 diabetes.

The microbiota-gut-brain axis (MGBA) is a bidirectional network linking gut microbes to the CNS. This review elucidates how microbial messengers-short-chain fatty acids, bile acids, neuroactive metabolites, and extracellular vesicles-drive obesity and type 2 diabetes via neural, endocrine, and immune pathways. MGBA dysregulation triggers hypothalamic dysfunction, adipose inflammation, incretin axis impairment, and β‑cell failure. We discuss MGBA-targeted therapies and their translational challenges.

Natural products targeting the gut-brain axis for the treatment of post-cardiac procedures anxiety or depression.

BACKGROUND: Post-cardiac surgery anxiety or depression (PCPAD) is a common neuropsychiatric complication following cardiovascular interventional procedures, which significantly increases the risk of adverse cardiovascular events and long-term mortality. Existing treatment strategies have limitations, and clinical needs remain unmet. The gut-brain axis (GBA) serves as a core network regulating neuroimmune and endocrine responses, and its imbalance involves key links such as intestinal flora dysbiosis and neuroimmune crosstalk disorders. It is closely related to the pathogenesis of this complica

Neural and metabolic mechanisms of emetogenic foodborne toxins via the brain-gut axis in ageing and cachexia.

Emetic foodborne toxins (bacterial enterotoxins, mycotoxins like DON/ZEA, marine toxins, cereulide, T-2 toxin, domoic acid, and biogenic amines such as histamine) cause acute gastrointestinal reactions and chronic systemic pathologies. This review outlines their activation of the brainstem's area postrema (AP) via intestinal barrier disruption, dysbiosis, vagus nerve stimulation, or direct central penetration. Acute exposure induces anorexia/defense responses through AP/NTS receptor pathways (GLP-1R, CCK, PYY, ghrelin, GDF15-GFRAL, 5-HT3). Chronic exposure impairs the AP-hypothalamus-locus coe

The Gut-Brain-Immune Axis: Role of Microbiota Dysbiosis and Autonomic Nervous System in Infectious Diseases.

The human gastrointestinal tract harbors a complex microbial ecosystem that plays a fundamental role in maintaining immune balance and defending against infectious diseases. Disruption of this ecosystem, known as dysbiosis, has been increasingly associated with impaired immune responses and heightened susceptibility to infections. Emerging evidence highlights the importance of the gut-brain-immune axis, particularly the role of the autonomic nervous system (ANS), in regulating these interactions. This systematic review aimed to examine the relationship between gut microbiota dysbiosis and infe

Interplay Between the Gut-Microbiota-Brain Axis and the Immune System in Viral Neuroinvasion and CNS Pathogenesis.

Evolving demographic factors have contributed to the continuous emergence of viral pathogens in humans, a great portion of which target the central nervous system (CNS). These viral pathogens influence multiple layers of host physiology, including the host's microbial communities. Gut microbiota consist of a plethora of microscopic organisms that serve as an integral part of the gut-brain axis. Emerging evidence suggests that the microbiota-gut-brain axis regulates many physiological processes, which are critical to the disease outcome, such as the immune response in several viral infections.

cGAS/STING is associated with brain-gut-liver axis disturbance and systemic inflammation in cerebral ischemia.

Cerebral ischemia triggers a cascade of systemic inflammatory responses closely associated with brain-gut-liver axis disturbance, yet the underlying molecular mechanism remains incompletely understood. This study aimed to elucidate the mechanistic role of the cyclic GMP-AMP synthase/stimulator of interferon genes (cGAS/STING) signaling pathway in disrupting gut-liver axis homeostasis following cerebral ischemia. Using a middle cerebral artery occlusion (MCAO) rat model, we employed a multidisciplinary approach combining behavioral, histological, molecular, and biochemical assays. We demonstrat

Synergistic Modulation of the Gut-Brain-Immune Axis by a Dual Lactobacillus Combination in a Murine IBS Model.

The gut microbiota is a key regulator of immune and neuroendocrine pathways along the gut-brain axis. Disruption of this bidirectional communication contributes to irritable bowel syndrome (IBS), a multifactorial disorder associated with gastrointestinal dysfunction and psychiatric comorbidities. Although microbiota-targeted therapies are promising, most current studies rely on single-strain interventions with limited efficacy, and the bioactive components as well as their host-mediated mechanisms remain insufficiently characterized. Here, we demonstrate that oral co-administration of Lactobac

An open window: the crucial role of the gut-brain axis in neurodevelopmental outcomes post-neurocritical illness.

Among patients admitted to the pediatric intensive care unit, approximately 10% are discharged with a new functional morbidity. For those who were admitted with a neurocritical illness, the number can be as high as 60%. The most common diagnoses for a neurocritical illness admission include traumatic brain injury, status epilepticus, post-cardiac arrest, hypoxic ischemic encephalopathy, meningo/encephalitis, and stroke. The gut-brain axis is crucial to childhood development, particularly neurodevelopment. Alterations on either side of the bidirectional communication of the gut-brain axis have

Recent drug delivery systems targeting the gut-brain-microbiome axis for the management of chronic diseases.

In recent years, the study of microorganisms and the brain has become increasingly connected. The gut-brain-microbiome axis (GBMA), a bi-directional communication system, is the key part of how the body's bacteria and the brain interact. This system can influence the brain and behaviour. Changes in this relationship have been linked to various mental and physical health conditions. The immune system, tryptophan metabolism, the vagus nerve, and the enteric nervous system all facilitate connections between the gut and brain. Microbes produce Peptidoglycans, branched-chain amino acids, and short-
