Senecavirus a 3C protease cleaves RETREG1 to antagonize ER-phagy and promote viral replication via endoplasmic reticulum calcium signaling.

Co-evolution between viruses and autophagy has led to the emergence of viral strategies that manipulate host endoplasmic reticulum (ER) homeostasis, ultimately promoting viral replication. ER turnover is achieved through selective autophagy, also referred to as ER-phagy, which is regulated by the RETREG1/FAM134B (reticulophagy regulator 1) family of reticulon proteins. Nevertheless, how viruses target RETREG1, a receptor for ER-phagy, remains largely unclear. In this study, we demonstrate that infection with Senecavirus A (SVA), an emerging picornavirus, triggers the cleavage of RETREG1, which

Swelling-induced calcium signaling in wild-type and Mlc1-null astrocytes: paradoxical effects of pharmacological TRPV4 and Piezo1 inhibition.

INTRODUCTION: Astrocytes play a crucial role in brain ion and water homeostasis and continuously adapt their volume in response to osmotic challenges. This process is disturbed in the leukodystrophy megalencephalic leukoencephalopathy with subcortical cysts (MLC). Intracellular calcium signaling in response to cell swelling has been implicated in astrocytic volume regulation. METHODS: We examined swelling-induced calcium signals in cultured astrocytes isolated from brains of wild-type and Mlc1-null mice, utilizing a microplate reader and Fura-2AM-based imaging. RESULTS: We showed that wild-t…

Effects of CdTe quantum dot exposure on the calcium signaling pathway in rat dorsal root ganglion cells ND7/23.

Previous study has shown that CdTe QDs exposure reduced ND7/23 cells activity and induced cell apoptosis in a time-dependent manner. The mitochondrial pathway was involved in CdTe QDs-induced ND7/23 cell apoptosis. The toxic effects at the subcellular level of ND7/23 cells after CdTe QDs exposure was still unclear. Whether the two classical mechanisms, endoplasmic reticulum stress and calcium ion imbalance, were involved in the subcellular structural and functional dysfunction of ND7/23 cells induced by CdTe QDs, and whether the Ca2 + -calpain2 pathway played a significant role in the CdTe QDs

IP3 Receptors Mediate Calcium and Anabolic Signaling Associated with Muscle Atrophy upon 3-Day Hind Limb Unloading in Rats.

Skeletal muscle unloading results in muscle atrophy associated with the upregulation of proteolytic genes and suppression of protein synthesis, often accompanied by altered calcium signaling. Here, we used the inositol trisphosphate receptor (IP3R) inhibitor aminoethoxydiphenyl borate (2-APB) to explore the hypothesis that these changes are mediated by IP3Rs. Male Wistar rats were divided into 4 groups: (i) control, (ii) control with daily injections of 2-APB, (iii) 3 days of hind limb suspension, (iv) 3 days of hind limb suspension with daily administration of 2-APB. At the end-point, soleus …

PLCγ1-PKCε-IP3R1 signaling plays an important role in hypoxia-induced calcium response in pulmonary artery smooth muscle cells.

Hypoxia-induced pulmonary vasoconstriction (HPV) is attributed to an increase in intracellular Ca2+ concentration ([Ca2+]i) in pulmonary artery smooth muscle cells (PASMCs). We have reported that phospholipase C-γ1 (PLCγ1) plays a significant role in the hypoxia-induced increase in [Ca2+]i in PASMCs and attendant HPV. In this study, we intended to determine molecular mechanisms for hypoxic Ca2+ and contractile responses in PASMCs. Our data reveal that hypoxic vasoconstriction occurs in pulmonary arteries, but not in mesenteric arteries. Hypoxia caused a large increase in [Ca2+]i in PASMCs, whi

Calcium-calmodulin signaling elicits mitochondrial dysfunction and the release of cytochrome c during cadmium-induced apoptosis in primary osteoblasts.

Cadmium (Cd) is a toxic heavy metal used in industry and is associated with adverse effects on human health following long- or short-term environmental exposure. Although Cd is known to induce apoptosis in many human organ systems, the mechanism that underlies its toxicity in primary osteoblasts (OBs) is not yet established. In the present study, we confirmed that Cd induced apoptosis in OBs isolated from the craniums of fetal Sprague-Dawley rats. We then showed that exposure to Cd transiently increased intracellular calcium ([Ca(2+)]i) levels for up to 1.5h, after which the levels returned to

Further insights into the antinociceptive potential of a peptide disrupting the N-type calcium channel-CRMP-2 signaling complex.

The N-type voltage-gated calcium channel (Cav 2.2) has gained immense prominence in the treatment of chronic pain. While decreased channel function is ultimately anti-nociceptive, directly targeting the channel can lead to multiple adverse side effects. Targeting modulators of channel activity may facilitate improved analgesic properties associated with channel block and a broader therapeutic window. A novel interaction between Cav 2.2 and collapsin response mediator protein 2 (CRMP-2) positively regulates channel function by increasing surface trafficking. We recently identified a CRMP-2 pept

Mitogen-activated protein kinase signaling in bovine articular chondrocytes in response to fluid flow does not require calcium mobilization.

In the present study, the role of mitogen-activated protein kinases (MAPKs) in chondrocyte mechanotransduction was investigated. We hypothesized that MAPKs participate in fluid flow-induced chondrocyte mechanotransduction. To test our hypothesis, we studied cultured chondrocytes subjected to a well-defined mechanical stimulus generated with a laminar flow chamber. The extracellular signal-regulated kinases 1 and 2 (ERK1/2) were activated 1.6-3-fold after 5-15 min of fluid flow exposure corresponding to a chamber wall shear stress of 1.6 Pa. Activation of ERK1/2 was observed in the presence of

A sonogenetic antigen-presenting nanoplatform enables spatiotemporal control of calcium signalling in T cells for enhanced cancer immunotherapy.

Effective T-cell activation plays the critical role in cancer immunotherapy; however, strategies to precisely and sustainably stimulate antitumor T-cell responses while minimizing systemic toxicity remain limited. Here, we develop a sonogenetic artificial antigen-presenting cell (aAPC) platform for remote, spatiotemporally controlled Ca2+ influx in tumor-infiltrating T cells. The platform consists of poly (lactic-co-glycolic acid) nanoparticles loaded with the plasmid encoding the Mechanosensitive channel of large conductance (MSCL), coated with tumor-pulsed dendritic cell membranes to provide
