The effect of a unilateral costochondral graft on the growth of the marmoset mandible.

PURPOSE: The purpose of the investigation was to examine growth of the mandible after costochondral grafting with special reference to the amount of cartilage included in the grafts. MATERIALS AND METHODS: The material consisted of 5 growing and 3 adult marmoset monkeys, Callithrix jacchus, in which the condylar process was removed unilaterally and replaced with a costochondral graft (CCG) containing either a short or long cartilage end. Growing animals were followed for 15 months until they attained maturity; and adult animals were followed for 13 months. Measurements made on frontal radiogr…

Prion protein immunohistochemical staining in the brains of monkeys with transmissible spongiform encephalopathy.

Prion protein (PrP) immunohistochemical staining of the brains of common marmosets (Callithrix jacchus) with experimental transmissible spongiform encephalopathy is described. The monkeys (n = 17) had been injected, intracerebrally, 17-49 months previously with homogenates of brain tissue taken post mortem from a cow with BSE (n = 2 monkeys), a sheep with natural scrapie (n = 2 monkeys), human cases of growth hormone related Creutzfeldt-Jakob disease (CJD) (n = 2 monkeys), sporadic CJD (n = 5 monkeys), or Gerstmann-Sträussler-Scheinker disease (GSS) (n = 4 monkeys), or from monkeys with spongi…

Gene regulatory innovations from transposable elements in primate cerebellum development.

Transposable elements are hypothesized to have driven gene regulatory innovation, yet their contributions to primate brain development at the cell type level remain underexplored. Here, we use single-cell multiomics data from human, macaque, marmoset, and mouse cerebella to show that transposable element contributions to different cell types are shaped by varying degrees of constraints across cell types, as well as the preferential co-option of certain transposable elements in specific cell states. Using a sequence-based deep-learning model that predicts cell-type-specific chromatin accessibil

Callitrichine herpesvirus 3 in the common marmoset is a model of Epstein-Barr virus infection and associated lymphoma.

Herpesviruses, such as Epstein-Barr virus (EBV), are thought to potentially play a significant role in multiple disease processes, including neoplasia, multiple sclerosis (MS), and more recently, Alzheimer's disease (AD). Animal models remain vital tools for understanding these diseases and developing therapeutics. Callitrichine herpesvirus 3 (CalHV-3) was identified in the early 2000s in the common marmoset (Callithrix jacchus). Although phylogenetically related to human EBV, the biological similarities between CalHV-3 and EBV have not been thoroughly characterized. Over 450 marmosets from fi…

Experimental basis for generating nonhuman primate models of frontotemporal dementia and Alzheimer's disease.

To our knowledge, no reports have described nonhuman primate (NHP) models of frontotemporal dementia with Parkinsonism linked to chromosome 17 (FTDP-17) that do not depend on an overexpression paradigm. Based on our recent success in generating single human MAPT knock-in mouse models of FTDP-17, we describe the experimental basis for generating knock-in marmoset models of FTDP-17. In addition, successful generation of mutant PSEN1 knock-in marmoset models lacking exon 9 (PSEN1-Δ9) of Alzheimer's disease (AD) indicates that we will be able to reconstitute two major pathological features of AD,

Distributed representations of prediction error signals across the cortical hierarchy are synergistic.

A relevant question concerning inter-areal communication in the cortex is whether these interactions are synergistic. Synergy refers to the complementary effect of multiple brain signals conveying more information than the sum of each isolated signal. Redundancy, on the other hand, refers to the common information shared between brain signals. Here, we dissociated cortical interactions encoding complementary information (synergy) from those sharing common information (redundancy) during prediction error (PE) processing. We analyzed auditory and frontal electrocorticography (ECoG) signals in fi

Cellular-resolution gene expression profiling in the neonatal marmoset brain reveals dynamic species- and region-specific differences.

Precise spatiotemporal control of gene expression in the developing brain is critical for neural circuit formation, and comprehensive expression mapping in the developing primate brain is crucial to understand brain function in health and disease. Here, we developed an unbiased, automated, large-scale, cellular-resolution in situ hybridization (ISH)-based gene expression profiling system (GePS) and companion analysis to reveal gene expression patterns in the neonatal New World marmoset cortex, thalamus, and striatum that are distinct from those in mice. Gene-ontology analysis of marmoset-speci

Histamine release from intestinal mast cells induced by staphylococcal enterotoxin A (SEA) evokes vomiting reflex in common marmoset.

Staphylococcal enterotoxins (SEs) produced by Staphylococcus aureus are known as causative agents of emetic food poisoning. We previously demonstrated that SEA binds with submucosal mast cells and evokes mast cell degranulation in a small emetic house musk shrew model. Notably, primates have been recognized as the standard model for emetic assays and analysis of SE emetic activity. However, the mechanism involved in SEA-induced vomiting in primates has not yet been elucidated. In the present study, we established common marmosets as an emetic animal model. Common marmosets were administered cl

Selective Neuronal Activation by Cochlear Implant Stimulation in Auditory Cortex of Awake Primate.

UNLABELLED: Despite the success of cochlear implants (CIs) in human populations, most users perform poorly in noisy environments and music and tonal language perception. How CI devices engage the brain at the single neuron level has remained largely unknown, in particular in the primate brain. By comparing neuronal responses with acoustic and CI stimulation in marmoset monkeys unilaterally implanted with a CI electrode array, we discovered that CI stimulation was surprisingly ineffective at activating many neurons in auditory cortex, particularly in the hemisphere ipsilateral to the CI. Furthe

The common marmoset as a model for the study of nonalcoholic fatty liver disease and nonalcoholic steatohepatitis.

Nonalcoholic fatty liver disease (NAFLD) is considered the hepatic manifestation of metabolic syndrome. The more clinically concerning form of the disease, nonalcoholic steatohepatitis (NASH), is characterized by steatosis, lobular inflammation, and ballooning degeneration. Here we describe a naturally occurring syndrome in the common marmoset that recapitulates the pathologic findings associated with NAFLD/NASH in humans. Hepatomegaly determined to result from NAFLD was observed in 33 of 183 marmosets. A comprehensive histopathologic assessment performed in 31 marmosets demonstrated that NAFL

MPTP treatment of common marmosets impairs proteasomal enzyme activity and decreases expression of structural and regulatory elements of the 26S proteasome.

Dysfunction of the ubiquitin-proteasome system occurs in the substantia nigra (SN) in Parkinson's disease (PD). However, it is unknown whether this is a primary cause or a secondary consequence of other components of the pathogenic process. We have investigated in nonhuman primates whether initiating cell death through mitochondrial complex I inhibition using 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine hydrochloride (MPTP) altered proteasomal activity or the proteasomal components in the SN. Chymotrypsin-like, trypsin-like and peptidylglutamyl-peptide hydrolase (PGPH) activating of 20S protea

Immunolocalisation of oestrogen receptor-alpha within the testis and excurrent ducts of the rat and marmoset monkey from perinatal life to adulthood.

The sites of action and the physiological role of oestrogens in the male reproductive tract are poorly understood. We have undertaken a systematic study of the immunoexpression of oestrogen receptor-alpha (ER alpha) in the male rat from late fetal life through to adulthood and compared the findings with results obtained in the marmoset monkey (Callithrix jacchus) from neonatal to adult life. The testes, rete testis, efferent ducts and epididymis were examined from normal male rats (aged 4, 8, 10, 15, 20, 25, 38, 48 and 90 days) and from male rat fetuses on days 17.5 and 18.5 of gestation; comp…

Acute upper genital-tract disease in female monkeys provoked experimentally by Mycoplasma genitalium.

The oviducts of two grivet monkeys and three marmosets, all sexually mature animals, were inoculated with Mycoplasma genitalium at laparotomy. The mycoplasma was not recovered from the grivet monkeys, nor from the oviducts of the marmosets although it was isolated intermittently from the vagina of two of the latter animals up to 4-6 weeks after inoculation. In contrast, all of the animals developed antibody to M. genitalium measured by a micro-immunofluorescence technique. It developed rapidly in the grivet monkeys but slowly in the marmosets, being detected first about 1 month after inoculati

Immune profile of an atypical EAE model in marmoset monkeys immunized with recombinant human myelin oligodendrocyte glycoprotein in incomplete Freund's adjuvant.

BACKGROUND: Experimental autoimmune encephalomyelitis (EAE) in the common marmoset monkey (Callithrix jacchus) is a relevant preclinical model for translational research into immunopathogenic mechanisms operating in multiple sclerosis (MS). Prior studies showed a core pathogenic role of T and B cells specific for myelin oligodendrocyte glycoprotein (MOG). However, in those studies, the quality of the response against MOG epitopes was strongly biased by bacterial antigens in the complete Freund's adjuvant (CFA), in which the immunizing recombinant human (rh) MOG protein had been formulated. In …

Distributed neural codes of the 3D position in the marmoset frontal cortex and hippocampus.

The neural encoding of the animal's position is essential for spatial navigation and cognition, yet the three-dimensional (3D) positioning system in the primate brain remains poorly understood. By combining wireless multi-channel recordings from common marmosets with a free-forging task in the 3D space, we identified 42% and 24% neurons in the frontal cortex (FC) and hippocampus (HPC) as place cells, respectively. Place cells in FC and HPC encoded the 3D position with similar spatial information, sparsity and stability, and exhibited stronger functional connections than non-place cells. Notabl

Spatial Adaptation of Primate Retinal Ganglion Cells Between Artificial and Natural Stimuli.

The retina encodes a broad range of stimuli, adapting its computations to features like brightness, contrast, and motion. However, it is unclear whether it also adapts when switching between natural scenes and white noise (WN). To address this, we analyzed the neural activity of male marmoset retinal ganglion cells (RGCs) in response to WN and naturalistic movies. We trained linear-nonlinear models on both stimuli, evaluated their performance, and compared their receptive fields across stimulus domains. We found that models with spatial filters trained on one stimulus ensemble were less accura

Mapping the metagenomic landscape: combined shotgun sequencing and quantitative PCR to profile gut metagenome-assembled genomes in marmosets following treatment with a broad-spectrum antibiotic cocktail.

Broad-spectrum antibiotics are invaluable tools for treating pathogenic infections, but their sustained use can contribute to changes in gut microbiome membership and the emergence of antimicrobial resistance. While these unintended side effects are independently well documented, the relationship between them has seldom been investigated. To address this, we quantified the effects of 28-d antibiotic cocktail exposure on metagenome-assembled genomes and antibiotic resistance genes in common marmosets using a custom whole-genome shotgun sequencing pipeline and quantitative polymerase chain react

An expanded subventricular zone supports postnatal cortical interneuron migration in gyrencephalic brains.

Cortical GABAergic interneurons generated in the ventral developing brain travel long distances to their final destinations. While there are examples of interneuron migration in the neonatal human brain, the extent of postnatal migration across species and how it contributes to cortical interneuron composition remains unknown. Here we demonstrate that neonatal gyrencephalic brains, including humans, nonhuman primates and piglets, harbor an elaborate subventricular zone, termed the Arc, due to its curved morphology and expanded neuroblast populations. The Arc is absent in lissencephalic marmose

Filter-based models of suppression in retinal ganglion cells: Comparison and generalization across species and stimuli.

The dichotomy of excitation and suppression is one of the canonical mechanisms explaining the complexity of neural activity. Computational models of the interplay of excitation and suppression in single neurons aim at investigating how this interaction affects a neuron's spiking responses and shapes the encoding of sensory stimuli. Here, we compare the performance of three filter-based stimulus-encoding models for predicting retinal ganglion cell responses recorded from axolotl, mouse, and marmoset retina to different types of temporally varying visual stimuli. Suppression in these models is i
