A limbic circuit selectively links active escape to food suppression.

Stress has pleiotropic physiologic effects, but the neural circuits linking stress to these responses are not well understood. Here, we describe a novel population of lateral septum neurons expressing neurotensin (LSNts) in mice that are selectively tuned to specific types of stress. LSNts neurons increase their activity during active escape, responding to stress when flight is a viable option, but not when associated with freezing or immobility. Chemogenetic activation of LSNts neurons decreases food intake and body weight, without altering locomotion and anxiety. LSNts neurons co-express sev

Basolateral amygdala input to the medial prefrontal cortex controls obsessive-compulsive disorder-like checking behavior.

Obsessive-compulsive disorder (OCD) affects ∼1 to 3% of the world's population. However, the neural mechanisms underlying the excessive checking symptoms in OCD are not fully understood. Using viral neuronal tracing in mice, we found that glutamatergic neurons from the basolateral amygdala (BLAGlu) project onto both medial prefrontal cortex glutamate (mPFCGlu) and GABA (mPFCGABA) neurons that locally innervate mPFCGlu neurons. Next, we developed an OCD checking mouse model with quinpirole-induced repetitive checking behaviors. This model demonstrated decreased glutamatergic mPFC microcircuit a

Dopamine neurons projecting to medial shell of the nucleus accumbens drive heroin reinforcement.

The dopamine (DA) hypothesis posits the increase of mesolimbic dopamine levels as a defining commonality of addictive drugs, initially causing reinforcement, eventually leading to compulsive consumption. While much experimental evidence from psychostimulants supports this hypothesis, it has been challenged for opioid reinforcement. Here, we monitor genetically encoded DA and calcium indicators as well as cFos in mice to reveal that heroin activates DA neurons located in the medial part of the VTA, preferentially projecting to the medial shell of the nucleus accumbens (NAc). Chemogenetic and op

Electroacupuncture pretreatment ameliorates sleep deprivation-induced cardiac dysfunction via the regulation of GABAergic neuron activity in the cerebellar fastigial nucleus.

OBJECTIVE: Sleep deprivation (SD) is increasingly prevalent and associated with impaired cardiac function. Diminished γ-aminobutyric acid (GABA)-ergic neuronal activity in the cerebellar fastigial nucleus (FN) constitutes a key mechanism driving this disorder. Although electroacupuncture (EA) preconditioning has shown potential for mitigating SD-induced cardiac impairment, the underlying mechanisms remain unclear. Additionally, while it is known that FNGABA neurons contribute to the structural and functional integrity of cardiac autonomic regulation, the effects of EA on these neurons, specifi

Reducing ventral hippocampal CA1/subiculum hyperexcitability restores social memory and alleviates anxiety-related behavior in a mouse model of temporal lobe epilepsy.

BACKGROUND: Interictal cognitive and affective comorbidities in temporal lobe epilepsy (TLE) often remain refractory to seizure-directed therapies. We tested the causal role of ventral hippocampal CA1/subiculum (vCA1/Sub) hyperexcitability in social memory failure and anxiety-related behavior, and whether normalizing principal-cell excitability restores function. METHODS: In pilocarpine-treated mice we combined blinded behavioral assays (social approach/discrimination, open field, olfaction), whole-cell recordings from mCherry-labeled vCA1/Sub principal neurons, alveus stimulation to assay sy

General Anesthesia-activated Neurons in the Central Amygdala Mediate Antinociception: Distinct Roles in Acute versus Chronic Phases of Nerve Injury.

BACKGROUND: General anesthesia, such as isoflurane, induces analgesia (loss of pain) and loss of consciousness through mechanisms that are not fully understood. A distinct population of γ-aminobutyric acid-mediated neurons has been recently identified in the central amygdala (CeA) that can be activated by general anesthesia (CeA GA ) and exert antinociceptive functions. In this study, the authors aimed to explore the underlying cellular mechanisms of CeA GA neurons across different phases of nerve injury-induced nociceptive sensitization in mice. METHODS: This study used 107 mice, including 5

Circuit Mechanisms Underlying the Contribution of Leptin Receptor-Expressing Neurons in the Nucleus Tractus Solitarius to Ventilatory Homeostasis.

Sleep apnea syndrome (SAS) is a prevalent disorder characterized by recurrent respiratory pauses during sleep; however, the neural mechanisms governing respiratory stability remain poorly understood. In this study, we identify the ratio of sigh expiratory volume to eupneic expiratory volume as a potential predictor of post-sigh apnea in susceptible C57BL/6J mice. We demonstrate that leptin signaling within the nucleus tractus solitarius (NTS) is critical for maintaining respiratory drive and suppressing apnea. Chemogenetic activation of Leptin receptor b-expressing NTS (NTSLepRb) neurons signi

Immature olfactory sensory neurons provide complementary input in the intact olfactory system.

Adult neurogenesis of olfactory sensory neurons (OSNs) provides the unique opportunity to understand how new neurons functionally integrate into existing circuitry and contribute to behaviors. Previous studies have shown that immature OSNs provide odor input to neurons in the mouse OB, and that, in mice lacking mature OSNs, sensory input from immature OSNs can mediate odor detection and discrimination. What remains unknown is how immature OSNs contribute to odor-guided behavior in the intact olfactory system. Here we show that acute chemogenetic silencing of immature OSNs impairs performance i

Astrocyte activation in the ventrolateral medulla modulates breathing and arousal states.

Astrocytes play vital roles in regulating brain states across organisms. Specifically, they regulate breathing behaviors and associated brain states, including facilitating transitions between breathing phases by sensing changes in O₂ and CO₂ levels, regulating the sleep-wake cycle, and impacting arousal and wakefulness. Here, we test the hypothesis that astrocytes in the ventral respiratory column (VRC) are important for arousal and sigh generation in alert mice. Our results reveal that a subset of Aldh1l1 cells in the VRC are activated prior to sigh generation and are recruited by hypoxia. C

Periaqueductal gray astrocytes contribute to depression-pain comorbidity via the CXCL10/CXCR3 axis.

Depression-pain comorbidity is a prevalent neuropsychiatric disorder with unclear pathogenic mechanisms. The periaqueductal gray (PAG) is one of the brain regions involved in emotional and pain processing. While astrocytes are known to be involved in various neurological disorders, their specific role in this comorbidity within the PAG is unclear. In this study, a mouse model of depression-pain comorbidity was established via chronic restraint stress (CRS). We found that CRS exposure induced marked depressive-like behaviors and mechanical allodynia in mice. This was accompanied by significant

A paraventricular hypothalamic control of social stress susceptibility by targeting brown adipose tissue β3 adrenoceptor.

Adipose tissue disturbance underlies the pathogenesis of major depressive disorder (MDD); however, the underlying neural and molecular mechanisms remain elusive. Utilizing a social defeat stress mouse model of MDD, we identified a paraventricular hypothalamic (PVH) control of susceptibility to social stress via β3 adrenoceptor (β3AR) signaling in the brown adipose tissue (BAT). Susceptible mice exhibited a significantly elevated expression of BAT β3AR. Pharmacological activation of β3AR increased immobility time in the forced swim test in naïve mice, and decreased social interaction time in mi

A Neuroendocrine Circuit That Suppresses Excretion and Pelvic Pain During Activity.

Mate-seeking during peak sexual receptivity is tightly coupled with exploratory movement, a process made more efficient by transient suppression of excretion. Here, we define a functional hypothalamic-hindbrain circuit in mice that promotes locomotion while eliminating urination and defecation for hours. Chemogenetic stimulation of excitatory estrogen-melanocortin-responsive MC4R + neurons in the ventrolateral ventromedial hypothalamus (VMHvl) effectively silences bladder and colonic visceral reflexes, even at noxious distension pressures, underscoring the potency of this anti-excretion circui

Gut-brain cholinergic signaling mediates the antiseizure effects of Bacteroides fragilis.

Gut dysbiosis has been implicated in epilepsy, yet probiotic efficacy and mechanisms remain unclear. Here, we identify that Bacteroides fragilis (B. fragilis) is markedly reduced in children with epilepsy and show that oral B. fragilis administration suppresses seizures in both pentylenetetrazole- and kainic-acid-induced mouse models. Mechanistically, B. fragilis activates colonic choline acetyltransferase-positive (ChAT+) cells and enhances gut-vagus-brain cholinergic signaling, as demonstrated by vagal recordings, pharmacological blockade, and chemogenetic manipulation, identifying a colonic

Glycosaminoglycan sugar side chains on perineuronal nets in the fastigial nucleus regulate motor skills.

Perineuronal nets (PNNs), composed primarily of chondroitin sulfate proteoglycans with core proteins and glycosaminoglycan (GAG) side chains, are specialized extracellular matrices that modulate synaptic transmission. However, the specific function of PNNs in the cerebellum, particularly in the fastigial nucleus (FN), and their influence on motor skill regulation remains unclear. Here, we initially performed chemogenetic inactivation of the FN and confirmed that the activity of glutamatergic neurons in the FN is required for both gross and fine motor skills. We then demonstrated that motor ski

Galectin-related protein, a key contributor, drives diabetes-associated neuropathic pain.

Neuropathic pain associated with central sensitization is common in diabetic patients, but the underlying mechanisms remain unclear. Here, a proteomics screen identified a previously uncharacterized protein, galectin-related protein (LGALSL), which was significantly upregulated in cerebrospinal fluid and extracellular fluid of the anterior cingulate cortex (ACC) in diabetes-related neuropathic pain (DNP) model rats. Exogenous LGALSL administration reduced mechanical nociceptive thresholds by activating glutamatergic neurons in the ACC (ACCGlu). Chemogenetic manipulations and functional assays

The Bed Nucleus of the Stria Terminalis-Paraventricular Nucleus of the Hypothalamus Neural Circuit Regulates Neuropathic Pain Through the Brain-Spleen Axis.

Neuropathic pain is a chronic condition caused by damage or dysfunction in the nervous system. While the spleen may influence neuropathic pain, its role has been poorly understood. This study demonstrates that the spleen plays a crucial role in regulating neuropathic pain through the bed nucleus of the stria terminalis (BNST) - paraventricular nucleus of the hypothalamus (PVN) neural circuit in a chronic constriction injury (CCI) mouse model. Splenectomy, splenic denervation, or splenic sympathectomy significantly increased the mechanical withdrawal threshold (MWT) and reduced macrophage infil

Prolactin modulation of thermoregulatory circuits provides resilience to thermal challenge of pregnancy.

Pregnancy represents a significant challenge to thermal homeostasis, with adaptive changes required to cope with the increased generation of metabolic heat. We have investigated the hypothesis that prolactin action in the preoptic area (POA) of the hypothalamus contributes to adaptive changes in thermoregulation during pregnancy. Prolactin receptors (Prlr) are expressed in warm-responding neurons (WRNs) in the POA, and chemogenetic activation of this subpopulation induces profound hypothermia. Adeno-associates virus (AAV)-Cre-mediated deletion of Prlr in the POA results in significant hyperthe

Cortical Control of Itch Sensation by Vasoactive Intestinal Polypeptide-Expressing Interneurons in the Anterior Cingulate Cortex.

The anterior cingulate cortex (ACC) has recently been proposed as a key player in the representation of itch stimuli. However, to date, little is known about the contribution of specific ACC interneuron populations to itch processing. Using c-Fos immunolabeling and in vivo Ca2+ imaging, we reported that both histamine and chloroquine stimuli-induced acute itch caused a marked enhancement of vasoactive intestinal peptide (VIP)-expressing interneuron activity in the ACC. Behavioral data indicated that optogenetic and chemogenetic activation of these neurons reduced scratching responses related t

Astrocytic RIPK3 exerts protective anti-inflammatory activity in mice with viral encephalitis by transcriptional induction of serpins.

Flaviviruses pose a substantial threat to public health because of their ability to infect the central nervous system (CNS). Receptor-interacting protein kinase 3 (RIPK3) is a central coordinator that promotes neuroinflammation during viral infection of the CNS, a role that occurs independently of its canonical function in inducing necroptosis. Here, we used mouse genetic tools to induce astrocyte-specific deletion, overexpression, and chemogenetic activation of RIPK3 to demonstrate an anti-inflammatory function for astrocytic RIPK3. RIPK3 activation in astrocytes promoted host survival during

Activation of Ventral Tegmental Area Dopaminergic Neurons Projecting to the Parabrachial Nucleus Promotes Emergence from Propofol Anesthesia in Male Rats.

Since the clinical introduction of general anesthesia, its underlying mechanisms have not been fully elucidated. The ventral tegmental area (VTA) and parabrachial nucleus (PBN) play pivotal roles in the mechanisms underlying general anesthesia. However, whether dopaminergic (DA) projections from the VTA to the PBN play a role in mediating the effects of general anesthesia is unclear. We microinjected 6-hydroxydopamine into the PBN to damage tyrosine hydroxylase positive (TH+) neurons and found a prolonged recovery time from propofol anesthesia. We used calcium fiber photometry recording to exp
