Immune Responses in Cecal Tonsils of Marek's Disease Virus-Infected Chickens.

Marek's disease (MD) is a lymphoproliferative disease of domestic chickens that is caused by a highly cell-associated oncogenic α-herpesvirus, Marek's disease virus (MDV). MDV replicates in chicken lymphocytes and establishes a latent infection within CD4+ T cells. Clinical signs of MD include depression, crippling, weight loss, bursal/thymic atrophy, neurologic disorders, and rapid onset of T cell lymphomas that infiltrate lymphoid tissues, visceral organs, and peripheral nerves. The cecal tonsils (CTs) are considered the largest lymphoid aggregates of avian gut-associated lymphoid tissue. Al…

In vivo lipopolysaccharide injection alters CD4+CD25+ cell properties in chickens.

In chickens, thymic CD4(+)CD25(+) cells are characterized as regulatory T cells. The objectives of this experiment were to study the effects of an in vivo lipopolysaccharide (LPS) injection on the percentage of CD4(+)CD25(+) cells in peripheral organs and the suppressive properties of splenic CD4(+)CD25(+) cells in chickens. Chickens were injected with LPS and CD4(+)CD25(+) cells were analyzed at 1, 2, 3, 5, and 10 d post LPS injection. The LPS injection increased CD4(+)CD25(+) cell percentage approximately 5-fold in the blood at 1 d post LPS injection (P < 0.001), 3-fold in the thymus at 3 d …

Molecular typing and antimicrobial susceptibility of Clostridium perfringens from broiler chickens.

Clostridium perfringens (Cp) causes necrotic enteritis disease in commercial poultry. Antimicrobials are used to control and treat this disease and sometimes clinical outbreaks do not respond well to certain treatments. This study was designed to isolate Cp from clinical cases, type these isolates by multiplex PCR, and determine their antimicrobial susceptibility by micro-dilution method. A total of 67 Cp isolates were obtained from 155 broiler chicken flocks. All isolates were classified as type A and non-enterotoxin producers. Lincomycin, erythromycins, and tilmicosin showed very high minima

Construction of EGFP-tagged rBCG of E.tenella and distribution in chickens.

Chicken coccidiosis is a major parasitic disease with substantial economic burden to the poultry industry. Enhanced green fluorescent protein (EGFP) tagged recombinant Bacille Calmette-Guerin (rBCG), as a fusion protein with coccidian rhomboid antigen was constructed to track rBCG in vivo in chickens in this study. Immunization of chickens with one dose of rBCG pMV361-Rho/EGFP induced humoral immune response. The colonization of rBCG in liver, spleen, lung, kidney and caecum was observed by laser confocal microscopy. Real-time quantitative RT-PCR showed a rise expression level of rhomboid prot…

Mice as potential carriers of infectious bursal disease virus in chickens.

To investigate the role of mice as potential carriers of infectious bursal disease virus (IBDV), three mice were inoculated with a very virulent strain of IBDV and allowed to have contact with three uninoculated mice. Faeces, intestine and pooled liver and spleen collected from inoculated mice 12 and 24 h post-inoculation were positive for IBDV by reverse transcriptase-polymerase chain reaction (PCR)-nested PCR (RT-PCR-nPCR). IBDV was detected by RT-PCR-nPCR in 3/3 samples of intestine and 2/3 samples of pooled liver and spleen from uninoculated in-contact mice at 24 h after exposure. Chickens…

Hepatic lesions caused by migrating larvae of Ascaris suum in chickens.

Group A consisted of chickens infected with a single dose of Ascaris suum and group B of chickens infected with two successive doses. At days 1, 3, 7, 14 and 21 after the first or second infection dose, six chickens from each group were sacrificed. In both groups, larvae were recovered from the livers on days 1, 3, and 7 and lungs on days 3 and 7. No larvae were detected in chickens on day 14. Clear white lesions were noticed only on the livers from chickens of group B at day 7 but had disappeared at day 14. A comparison with group B showed mild histological changes that developed relative to …

Campylobacter antimicrobial drug resistance among humans, broiler chickens, and pigs, France.

We describe isolates from human Campylobacter infection in the French population and the isolates' antimicrobial drug resistance patterns since 1986 and compare the trends with those of isolates from broiler chickens and pigs from 1999 through 2004. Among 5,685 human Campylobacter isolates, 76.2% were C. jejuni, 17.2% C. coli, and 5.0% C. fetus. Resistance to nalidixic acid increased from 8.2% in 1990 to 26.3% in 2004 (p < 10(-3)), and resistance to ampicillin was high over time. Nalidixic acid resistance was greater for C. coli (21.3%) than for C. jejuni (14.9%, p < 10(-3)). C. jejuni resista…

West Nile virus circulation detected in northern Italy in sentinel chickens.

Ninety percent (56/62) of sentinel chickens introduced to two regions within the Italian Alps seroconverted to West Nile virus (WNV) during the summer of 2005, showing a range of antibody titres from 1/20 to 1/320 in a virus neutralization test. Neutralization specificity for WNV antibodies was confirmed on an additional 34 sera that were tested in parallel against WNV (16/34 seropositivity), Usutu virus (3/34 seropositivity) and Koutango virus. The geometric mean neutralizing titre (GMT) calculated for WN-specific antibodies was 33.68 and did not differ significantly amongst sample sites, alt…

Histomonas meleagridis in chickens: attempted transmission in the absence of vectors.

The progress and transmission of blackhead disease in chickens was studied in battery cages and floor pens in the absence of vectors. Two-week-old chicks were inoculated intracloacally with Histomonas meleagridis and allowed to commingle with others in floor pens. There was no confirmed transmission of blackhead to other birds in the pen, whether stocked at 10% or 25% with infected birds. A second experiment evaluated the effects of feed restriction of chickens on spread of blackhead within floor pens. Inoculated seeder birds had severe cecal lesions of blackhead at necropsy, regardless of fee…

Outbreaks of adenoviral gizzard erosion in slaughtered broiler chickens in Japan.

Gizzard erosion in broiler chickens was investigated at 18 slaughterhouses in Japan. The condition was observed in 13 of them, and adenoviral gizzard erosion (AGE) was diagnosed histologically, immunohistochemically and virologically in the eroded gizzards from nine of these 13. The antigen-positive intranuclear inclusion body of group 1 fowl adenovirus was observed in the epithelial cells of the affected gizzards, and fowl adenoviruses were isolated from the lesions. In two of the slaughterhouses the total weights of the gizzards disposed of in three years were 3590 kg (0.40 per cent of the g…

Production of free radical species during Eimeria maxima infections in chickens.

Five experiments were conducted to investigate the production of nitric oxide (NO) and superoxide anion (O2-) during infections of chickens with the coccidial parasite, Eimeria maxima, in order to assess the importance of these free radical species in the pathogenesis of the infections. Nitric oxide production was estimated by analyzing NO2(-)+NO3-, stable metabolites of NO, in the plasma and intestinal mucosa. The potential for O2- production was estimated from activities of beta-nicotinamide adenine dinucleotide phosphate (NADPH) oxidase in mucosal homogenates. Levels of NO2(-)+NO3- reached …

Comparative reproduction of Cryptosporidium baileyi in embryonated eggs and in chickens.

At 2 days of age, each of 20 chickens was perorally or intracloacally infected with 3 x 10(5) oocysts of Cryptosporidium baileyi and maintained for 13 days post infection. In parallel, 20 embryonated chicken eggs were inoculated with 3 x 10(5) oocysts at day 10 of embryonation and were incubated for a further 7 days. The average reproduction rates in the two groups of chickens were x560 after peroral infection and x533 after intracloacal infection as compared with x256 in the eggs. Although the rate of reproduction of parasites seen in the eggs was only about 50% of that observed in chickens, …

T cells in regenerating feathers of Smyth line chickens with vitiligo.

Smyth line (SL) chickens exhibit a spontaneous, autoimmune, depigmentation (SL vitiligo, SLV) in regenerating feather tissue due to the death of pigment cells (melanocytes). It was the purpose of this study to characterize mononuclear leukocytes which infiltrate the feather as SLV develops. Frozen sections of regenerating features from SL and control chickens were labeled with a panel of mAbs specific for chicken mononuclear leukocytes. SL chickens were found to have significantly greater numbers of T cells in the feather pulp prior to, and throughout, the development of SLV. Initially, both S…

Variations in oocyst output associated with Cryptosporidium baileyi infections in chickens.

Groups of birds, aged 2, 16 and 30 days, were experimentally infected with 400, 20,000 or 1,000,000 oocysts of Cryptosporidium baileyi. The faecal oocyst output for each group was determined by collection of faeces on Days 0, 5, 7 and 13-21 post-infection (p.i.). The duration of patency of infection, as determined by oocyst excretion, declined more rapidly in the older age groups but persisted longer in birds challenged with the lower oocyst numbers.

Thyroidal iodine metabolism in obese-strain chickens before immune-mediated damage.

Several studies have shown that iodine plays a role in spontaneous autoimmune thyroiditis in man and other animals. In addition, abnormalities of iodine metabolism have been found in patients with Hashimoto's thyroiditis and in chickens of the obese strain (OS), an animal model of spontaneous autoimmune thyroiditis. We have examined several parameters of iodine metabolism before immune damage in this model and in the related Cornell strain (CS), a strain which develops a late-onset mild thyroiditis, to discover a possible causal relationship between altered iodine metabolism and the initiation…

Marek's disease virus-induced transient paralysis in chickens: electron microscopic lesions.

A study was made to determine the causative lesion of Marek's disease virus (MDV)-induced transient paralysis (TP) in chickens by comparing the ultrastructure of brain tissue from MDV-infected genetically susceptible and resistant birds. There were numerous intramyelinic vacuoles in the brains of TP-affected birds. Many of these vacuoles contained particulate material compatible with precipitated protein from edema. Astrocyte processes often were distended with similar particulate material. Most intramyelinic vacuoles were either adjacent to the axolemma or within inner myelin lamellae. Myelin…
