A key aspect of human cognitive flexibility concerns the ability to convert complex symbolic instructions into novel behaviors. Previous research proposes that this transformation is supported by two neurocognitive states: an initial declarative maintenance of task knowledge, and an implementation state necessary for optimal task execution. Furthermore, current models predict a crucial role of frontal and parietal brain regions in this process. However, whether declarative and procedural signals independently contribute to implementation remains unknown. We report the results of an fMRI experi…

Chronic stress during childhood negatively impacts cognition and physical and mental health. Exposure to stressors over time can cause hypot … to investigate the associations of HCC, DHEA, and their ratio with measures of executive function (cognitive flexibility and working memory) in a sample of kindergarten children (N=100). We found that the expected negative association between HCC and working memory approached significance, and DHEA was significantly and positively related to cognitive flexibility. We …

Exposure to stress can have significant negative consequences on long-term health. Among modifiable targets for promoting adaptive stress responses, executive functioning has emerged as a promising candidate. The current study sought to examine the extent to which executive functioning facets (inhibition, flexibility, updating) and working memory capacity played a beneficial role in reactivity and recovery to an acute stressor. Participants completed executive functioning tasks in the laboratory before undergoing either an acute stressor (Maastricht Acute Stress Test, MAST) or its control equi

While deficits in 'cold' executive functioning (EF) have been linked to Generalized Anxiety Disorder (GAD), no study has systematically exam … osis and GAD symptoms/severity revealed similar patterns, such that, GAD was associated with poorer cognitive flexibility and working memory, but not inhibitory control. Effect sizes were similar across youths and young/middle-aged adults; however, studies involving older adults yielded no association between GAD and any EF domain. Furthermore, effect sizes were notabl …

Traumatic brain injuries (TBI) have led to lasting deficits for an estimated 5.3 million American patients. Effective therapies for these patients remain scarce and each of the clinical trials stemming from success in experimental models has failed. We believe that the failures may be, in part, due to the lack of preclinical assessment of cognitive domains that widely affect clinical TBI. Specifically, the behavioral tasks in the TBI literature often do not focus on common executive impairments related to the frontal lobe such as cognitive flexibility. In previous work, we have demonstrated th…

Increasing evidence shows that the developmental perturbation of immune activity can lead to long-lasting neurodevelopmental and behavioral abnormalities. In our previous study, we found that deficiencies of microglia and TNFα in the medial prefrontal cortex (mPFC) were involved in the impairment of cognitive flexibility induced by adolescent social stress in adult mice. It remains unclear how and when immune changes occur following adolescent stress exposure and whether it is possible to prevent the delayed occurrence of cognitive impairment through early immune intervention. Firstly, the pre…

Deficits in cognitive flexibility, i.e. the ability to modify behavior in response to changes in the environment, are present in several psychiatric disorders and are often refractory to treatment. However, improving treatment response has been hindered by a lack of understanding of the neurobiology of cognitive flexibility. Using a rat model of chronic stress (chronic intermittent cold stress, CIC) that produces selective deficits in reversal learning, a form of cognitive flexibility dependent on orbitofrontal cortex (OFC) function, we have previously shown that JAK2 signaling is required for…

This study examined how the change in trait resilience, an increasing tendency to effectively cope with challenges and flexibly adapt to stress, affected the change in transdiagnostic depressive symptoms over 6 months of clinical follow-up among outpatient adolescents, and the role of psychological inflexibility/experiential avoidance (PI/EA), characterised by rigid and avoidant emotion regulation, in this association. Trait resilience was measured with the Brief Resilience Scale, PI/EA with the Acceptance and Action Questionnaire-II and depressive symptoms with the Beck Depression Inventory-I

Anti-N-methyl-d-aspartate-receptor (NMDAR) encephalitis often leads to long-term cognitive impairments, including deficits in executive function, even after acute symptoms resolution, but the underlying mechanisms remain unclear. To explore this, a murine model was established via 14-day intracerebroventricular (ICV) infusion of anti-GluN1 IgG. Mice exhibited deficits in cognitive flexibility and short-term recognition memory, as evidenced by impaired performance in reversal learning and in the novel object recognition test, while spatial learning and anxiety-related behaviors were spared. Mol…

BACKGROUND: Executive functions (EF) encompass abilities such as planning, decision-making, and inhibitory control, critical for learning, establishing and maintaining behavioral change. The association between alcohol use disorder (AUD) and impairments in EF are well established. However, prior research is dominated by studies on convenience samples including individuals with severe AUD with high levels of psychiatric comorbidity, which limits generalizability. The present study therefore aimed to investigate the degree of impairment and predictive ability of EF, on alcohol consumption, among

This study is aimed to investigate the status of general anxiety and depression among suspected patients of COVID-19 and explore whether psychological flexibility can serve as a mediator between perceived stress and general anxiety or depression. Cognitive flexibility, the ability to switch behavior in response to changing rules in an uncertain environment, is crucial for adaptive decision-making. Prior research has hypothesized a key role of prediction error and theta oscillations in the medial frontal cortex in this process. However, the causal link between such processes remains to be established. To address this, we combined neural stimulation, EEG, behavioral measurement, and computational modeling. Specifically, we applied high-definition transcranial direct current stimulation to modulate theta oscillations as measured via EEG f…

BACKGROUND: In this peculiar case study, we extensively explored the effects of 6 weeks of supervised yoga on the endogenous pain modulatory system, autonomic profile, and corticomotor excitability.
CASE REPORT: A 29-year-old woman diagnosed with fibromyalgia (FMS) was suggested 6 weeks of supervised yoga because the pharmaceutical treatments were ineffective. Assessment was done at baseline, 2 weeks, and post-6 weeks for pain, cognitive skills, sleep quality, anxiety, and depression status. An autonomic profile was done, followed by flexibility and corticomotor excitability assessments.

Cognitive flexibility, attributed to frontal cortex, is vital for navigating the complexities of everyday life. The mediodorsal thalamus (MD), interconnected to frontal cortex, may influence cognitive flexibility. Here, male rats performed an attentional set-shifting task measuring intradimensional (ID) and extradimensional (ED) shifts in sensory discriminations. MD lesion rats needed more trials to learn the rewarded sensory dimension. However, once the choice response strategy was established, learning further two-choice discriminations in the same sensory dimension, and reversals of the rew…

METHODS: Three carefully diagnosed groups of children, aged between 7 and 12 years (35 ADHD, 22 RD and 30 typically developing children), were tested on a wide range of tasks related to five major domains of executive functioning (EF): inhibition, visual working memory, planning, cognitive flexibility, and verbal fluency. Additional tasks were selected for each domain to control fo…
