Atomic Force Microscopy to Investigate Collagen Fibrils.

This chapter presents a comprehensive overview of the synthesis and atomic force microscopy (AFM) imaging of collagen fibers. Collagen, a key structural protein in the extracellular matrix, plays a crucial role in tissue strength and integrity. Synthetic and in vitro assembled collagen fibers are widely used in biomedical research and regenerative medicine. The chapter begins with a description of collagen's hierarchical structure and its self-assembly mechanisms under physiological conditions. Different synthesis methods are discussed, including extraction from biological sources and recombin…

A collagen orientation switch reshapes fin architecture.

Fibrillar-collagen architecture is a fundamental determinant of vertebrate tissue morphology; yet, its organizational principles remain poorly understood because collagen structures are challenging to visualize in vivo. Actinotrichia, the fibrillar-collagen structures in fish fins, provide an ideal system: they are readily visualized and exhibit a simple, tractable spatial pattern optimal for studying collagen organization. Using high-sensitivity 3D imaging, we revealed the essential role of the fish-specific actinodin genes in establishing this pattern. Our findings demonstrate that actinodin…

Collagen Scaffold Viscoelasticity Regulates Muscle Cell Phenotype.

Current biomaterial strategies are typically unable to return skeletal muscle to pre-injury function following damage, resulting in permanent loss of muscle function. Recently, there has been a growing appreciation for the role of matrix viscoelasticity in regenerative processes, and here we address the hypothesis that changes in matrix viscoelasticity regulate muscle cell function. Using norbornene-modified type I collagen hydrogels with a tetrazine-based crosslinker, it is found that myoblast spreading, proliferation, and differentiation are improved on and within slow-relaxing hydrogels. Ho…

Collagen Crosslinking for Keratoconus: Cellular Signaling Mechanisms.

Collagen crosslinking (CXL) is a widely used treatment to halt the progression of keratoconus (KC). Unfortunately, a significant number of patients with progressive KC will not qualify for CXL, including those with corneas thinner than 400 µm. The present study aimed to investigate the molecular effects of CXL using in vitro models, mirroring the normal, as well as thinner corneal stroma seen in KCs. Primary human corneal stromal cells were isolated from healthy (HCFs) and keratoconus (HKCs) donors. Cells were cultured and stimulated with stable Vitamin C resulting in 3D self-assembled extrace…

Skin Grafting Using Collagen Fiber Orientation Matching.

UNLABELLED: Skin contracture after skin grafting is undesirable. It is generally accepted that full-thickness skin grafts contract less than split-thickness skin grafts. However, unexpected secondary skin-graft contracture sometimes occurs after full-thickness skin grafting. We tried to elucidate the causes of skin contracture from the viewpoint of the orientation of collagen fibers to find a way to reduce skin-graft contracture. METHOD: First, we examined the collagen fiber orientation of the skin over the whole body in Sprague-Dawley rats. Next, two pieces of skin (width: 30 mm × 30 mm; thi…

Solvent-Free Direct PEGylation of Collagen Fibers.

The addition of poly(ethylene glycol) (PEG) to biomolecules and biomaterials is a well-established approach to modify their properties for therapeutic applications. For biomaterials, the approach is typically to blend or electrospray the synthetic polymer with the biomaterial. Effective surface modification approaches such as surface-initiated polymer brushes are challenging since the harsh solvents required for brush synthesis may destroy the biomaterial. Herein, we describe the PEGylation of collagen fibers by surface-initiated PEG brushes using a living anionic grafting-from mechanism. This…

Prokaryotic Collagen-Like Proteins as Novel Biomaterials.

Collagens are the major structural component in animal extracellular matrices and are critical signaling molecules in various cell-matrix interactions. Its unique triple helical structure is enabled by tripeptide Gly-X-Y repeats. Understanding of sequence requirements for animal-derived collagen led to the discovery of prokaryotic collagen-like protein in the early 2000s. These prokaryotic collagen-like proteins are structurally similar to mammalian collagens in many ways. However, unlike the challenges associated with recombinant expression of mammalian collagens, these prokaryotic collagen-l…

Current methods of collagen cross-linking: Review.

This review provides a report on cross-linking methods used for collagen modifications. Collagen materials have attracted significant academic interest due to its biological properties in native state. However, in many cases the mechanical properties and degradation rate should be tailored to especial biomedical and cosmetic applications. In the proposed review paper, the structure, preparation, and properties of several collagen based materials have been discussed in general, and detailed examples of collagen cross-linking methods have been drawn from scientific literature and practical work.…

Collagen XVII Processing and Blistering Skin Diseases.

Collagen XVII (COL17) is a hemidesmosomal transmembrane protein in the skin, which, in several autoimmune blistering skin diseases, may be targeted by autoantibodies. In addition, loss-of-function mutations in the COL17A1 gene induce a subtype of junctional epidermolysis bullosa. The extracellular domain of COL17 can be physiologically cleaved from the cell surface by ADAM family proteins in a process known as ectodomain shedding. COL17 ectodomain shedding is thought to be associated with the migration and proliferation of keratinocytes. Furthermore, the C-terminal cleavage of COL17 may be ass…

Current perspectives on corneal collagen crosslinking (CXL).

Corneal collagen crosslinking has revolutionized the treatment of keratoconus and post-refractive corneal ectasia in the past decade. Corneal crosslinking with riboflavin and ultraviolet A is proposed to halt the progression of keratectasia. In the original "Conventional Dresden Protocol" (C-CXL), the epithelium is removed prior to the crosslinking process to facilitate better absorption of riboflavin into the corneal stroma. Studies analyzing its short- and long-term outcomes revealed that although there are inconsistencies as to the effectiveness of this technique, the advantages prevail ove…

Recent advances in corneal collagen cross-linking.

Corneal collagen cross-linking has become the preferred modality of treatment for corneal ectasia since its inception in late 1990s. Numerous studies have demonstrated the safety and efficacy of the conventional protocol. Our understanding of the cross-linking process is ever evolving, with its wide implications in the form of accelerated and pulsed protocols. Newer advancements in technology include various riboflavin formulations and the ability to deliver higher fluence protocols with customised irradiation patterns. A greater degree of customisation is likely the path forward, which will a…

Corneal collagen cross-linking for treating keratoconus.

BACKGROUND: Keratoconus is a condition of the eye that affects approximately 1 in 2000 people. The disease leads to a gradual increase in corneal curvature and decrease in visual acuity with consequent impact on quality of life. Collagen cross-linking (CXL) with ultraviolet A (UVA) light and riboflavin (vitamin B2) is a relatively new treatment that has been reported to slow or halt the progression of the disease in its early stages. OBJECTIVES: The objective of this review was to assess whether there is evidence that CXL is an effective and safe treatment for halting the progression of kerat…

Tumor matrix protein collagen XIα1 in cancer.

The extracellular matrix is increasingly recognized as an essential player in cancer development and progression. Collagens are one of the most important components of the extracellular matrix, and have themselves been implicated in many aspects of neoplastic transformation. Collagen XI is a minor collagen whose main physiologic function is to regulate the diameter of major collagen fibrils. The α1 chain of collagen XI (colXIα1) has known pathogenic roles in several musculoskeletal disorders. Recent research has highlighted the importance of colXIα1 in many types of cancer, including its roles…
