Computerized Tailored Interventions to Enhance Prevention and Screening for Hepatitis C Virus Among People Who Inject Drugs: Protocol for a Randomized Pilot Study.

BACKGROUND: Hepatitis C virus (HCV) infection is a growing problem among people who inject drugs. Strategies to reduce disease transmission (eg, syringe exchange programs) and facilitate HCV screening and linkage are available but are under-utilized in many communities affected by injection drug use. Novel approaches to increasing the use of these strategies are needed. OBJECTIVE: The goals of this project are to (1) develop and pilot test a computerized tailored intervention for increasing HCV screening and decreasing risky drug use behavior among people who inject drugs and (2) determine th

Automated image analysis of instagram posts: Implications for risk perception and communication in public health using a case study of #HIV.

People's perceptions about health risks, including their risk of acquiring HIV, are impacted in part by who they see portrayed as at risk in the media. Viewers in these cases are asking themselves "do those portrayed as at risk look like me?" An accurate perception of risk is critical for high-risk populations, who already suffer from a range of health disparities. Yet, to date no study has evaluated the demographic representation of health-related content from social media. The objective of this case study was to apply automated image recognition software to examine the demographic profile of

Comparative Study of Single-Cell and Bulk RNA Sequencing Data from Metastatic Bone Marrow Neuroblastoma Samples.

Neuroblastoma is characterized by frequent involvement of bone marrow (BM) as a site of cell dissemination and spread. In this study, single-cell RNA sequencing (scRNA-seq) was used to analyze the cellular heterogeneity of a subset of metastatic BM samples collected at initial diagnosis. Comparison of the single-cell data with bulk RNA sequencing further refined the analysis. An enrichment of regulatory T cells relative to a healthy control and activation of the CD24, CD47, and CD200 "don't eat me" signals were documented. Computational analyses highlighted communication between neuroblastoma

A Synergy of Convolutional Neural Networks for Sensor-Based EEG Brain-Computer Interfaces to Enhance Motor Imagery Classification.

Enhancing motor disability assessment and its imagery classification is a significant concern in contemporary medical practice, necessitating reliable solutions to improve patient outcomes. One promising avenue is the use of brain-computer interfaces (BCIs), which establish a direct communication pathway between users and machines. This technology holds the potential to revolutionize human-machine interaction, especially for individuals diagnosed with motor disabilities. Despite this promise, extracting reliable control signals from noisy brain data remains a critical challenge. In this paper,

Immune Dysregulation and Prognosis in Sepsis: Insights From a Posttranslational Perspective.

Sepsis is a life-threatening condition triggered by infection, resulting in widespread inflammation, immune dysfunction, and multiorgan failure. Despite advances in medical science, early detection, treatment, and prognosis remain significant challenges. In this study, we investigated the expression and functional role of ZDHHC19, a palmitoyltransferase enzyme, in sepsis. Our results demonstrate that ZDHHC19 is significantly upregulated in neutrophils during sepsis and correlates with inflammatory pathways critical to disease progression. High ZDHHC19 expression was associated with increased a

Dynamic event-triggered fuzzy non-fragile control of DC microgrids.

This paper studies the event-triggered fuzzy non-fragile control of uncertain DC microgrids subject to false data injection (FDI) attacks, controller saturation, network delays and premise mismatching. Firstly, a dynamic event-triggered mechanism (ETM) is proposed, which can save more communication bandwidth than the static ETMs, and remove the complex Zeno-free computation required by the continuous-time ETMs. Secondly, a fuzzy time-delay closed-loop system model is established, which provides a unified framework to study the effects of the dynamic ETM, FDI attacks, uncertainties, saturation,

LSTMs and Neural Attention Models for Blood Glucose Prediction: Comparative Experiments on Real and Synthetic Data.

We have shown in previous work that LSTM networks are effective at predicting blood glucose levels in patients with type I diabetes, outperforming human experts and an SVR model trained with features computed by manually engineered physiological models. In this paper we present the results of a much larger set of experiments on real and synthetic datasets in what-if, agnostic, and inertial scenarios. Experiments on a more recent real-patient dataset, which we are releasing to the research community, demonstrate that LSTMs are robust to noise and can easily incorporate additional features, such

Remodeling of the tumor microenvironment in muscle-invasive bladder cancer: insights from single-cell and spatial transcriptomics after neoadjuvant immunochemotherapy.

BACKGROUND: Neoadjuvant immunochemotherapy (nICT), defined in this study as PD-L1 blockade combined with cisplatin-based chemotherapy, provides substantial clinical benefits in muscle-invasive bladder cancer (MIBC). However, the tumor immune microenvironment (TIME) is highly heterogeneous, resulting in variable patient responses and persistent therapy resistance. This study investigates key immune cell subsets, intercellular communication networks, and spatial distributions within the TIME of MIBC, with a focus on cellular and spatial features associated with nICT response. METHODS: Patients

Predictive Functional Profiling of Allergy-Associated Microbiota with PICRUSt2.

Functional profiling of host-associated microbiota is essential for elucidating its role in immune dysregulation, such as in allergic diseases. Predictive bioinformatics tools like PICRUSt2 (Phylogenetic Investigation of Communities by Reconstruction of Unobserved States, version 2) enable the inference of microbial metabolic potential from 16 S rRNA gene amplicon data. This chapter describes the computational workflow for using PICRUSt2 to analyze allergy-associated microbiota. The protocol begins with a standardized amplicon sequence variant (ASV) table and its corresponding taxonomy and seq

Multi-Class Classification Methods for EEG Signals of Lower-Limb Rehabilitation Movements.

Brain-Computer Interfaces (BCIs) enable direct communication between the brain and external devices by decoding motor intentions from EEG signals. However, the existing multi-class classification methods for motor imagery EEG (MI-EEG) signals are hindered by low signal quality and limited accuracy, restricting their practical application. This study focuses on rehabilitation training scenarios, aiming to capture the motor intentions of patients with partial or complete motor impairments (such as stroke survivors) and provide feedforward control commands for exoskeletons. This study developed a

Metabolic reprogramming and taxonomic drivers in bacterial vaginosis: A large-scale metagenomic meta-analysis.

OBJECTIVE: Bacterial vaginosis (BV) represents a profound ecological shift from a Lactobacillus-dominated microbiota to a diverse polymicrobial biofilm associated with adverse outcomes. While taxonomic signatures are well-documented, the functional mechanisms driving this transition remain obscured. This study elucidates the genomic potential for metabolic reprogramming and the putative "functional handover" underpinning the stability of the dysbiotic state. METHODS: A computational meta-analysis of 3557 vaginal microbiomes from diverse global cohorts was performed using the standardized MGni

Collapse of interictal suppressive networks permits seizure spread.

How do brain networks limit seizure activity? In the interictal suppression hypothesis, we recently postulated that high inward connectivity to seizure onset zones (SOZs) from non-involved zones (NIZs) is a sign of broader network suppression. If broad networks appear to be responsible for interictal SOZ suppression, what changes during seizure initiation, spread and termination? For patients with drug-resistant epilepsy, intracranial monitoring offers a view into the electrographic networks that organize around and in response to the SOZ. In this manuscript, we investigate network dynamics in

Decoding cellular communication networks and signaling pathways in bone, skeletal muscle, and bone-muscle crosstalk through spatial transcriptomics in a young male mouse.

Bone and skeletal muscle are essential components of musculoskeletal system, enabling movement, load-bearing, and systemic homeostasis. These tissues communicate through dynamic bone-muscle crosstalk mediated by cytokines, growth factors, and extracellular-matrix (ECM) proteins. The spatial organization of these mediators is critical for maintaining tissue integrity, and its disruption contributes to diseases, such as osteoporosis, sarcopenia, and metabolic syndrome. Despite this importance, spatial transcriptomics (ST) studies of bone-muscle interactions remain limited. Here, we applied 10x G

Exploring the Role of the Rich Club in Network Control of Neurocognitive States.

The brain's rich club is a network of particularly densely interconnected regions, metabolically costly to maintain but central to the balance between functional segregation and integration. We assessed whether the rich club can accordingly be described as a control center of the brain, and present a systematic analysis of its involvement in maintenance of and traversal between various cognitively relevant functional states. Brain states were defined based on fMRI task-evoked and resting-state patterns of activity as provided by the Human Connectome Project (HCP). Using tools from network cont
