Species-specific divergence in Phytophthora infestans resistance between Nicotiana benthamiana and potato mediated by 1-aminocyclopropane-1-carboxylic acid pretreatment.

BACKGROUND: Late blight, caused by Phytophthora infestans, is a devastating oomycete disease threatening global potato (Solanum tuberosum) production. 1-Aminocyclopropane-1-carboxylic acid (ACC), has also been proposed to act as a bioactive signal in plant stress responses, but its contribution to plant immunity remains elusive. RESULTS: Here, we show that ACC pretreatment exerts opposite effects on P. infestans resistance in Nicotiana benthamiana and the potato cultivar 'Favorita', revealing strong species-specific transcriptional reprogramming. In N. benthamiana, differentially expressed ge

Arabica coffee SBP transcription factor family members respond to brown leaf spot stress by regulating their expression.

Coffee brown spot is a prevalent foliar fungal disease that seriously threatens the survival and yield of Coffea arabica L. To explore the molecular basis of disease resistance, we conducted a genome-wide identification and analysis of the SBP gene family in C. arabica L. A total of 22 CaSBP genes were identified and classified into three phylogenetic subfamilies, distributed across 12 chromosomes and including nine segmentally duplicated gene pairs. Cis-element analysis revealed a high proportion (55.37%) of hormone-related regulatory elements in CaSBP promoters. Integrated transcriptomic and

Integrated proteomic analysis reveals differential remodelling of defense responses in potato to herbivory by Phthorimaea operculella.

Plants rely on both transcriptional reprogramming and post-translational machinery to regulate their responses to biotic stress. While transcriptome-wide dynamics underlying plant defense against insect herbivores have been extensively documented, systematic investigations into protein-level regulation and integrative analyses of these multi-layered regulatory networks remain limited. In this study, we employed quantitative proteomics and phospho-proteomics to characterize the protein-level responses of potato plants to attacks by Phthorimaea operculella. A total of 60 differentially expressed

Microbial Volatile 3-Methyl-1-Butanol Enhances Stomatal Closure and Salt Stress Tolerance via Ethylene and Jasmonate Pathways in Arabidopsis.

Microbial biostimulants can be applied to reliably improve crop productivity and quality. One such biostimulant is 3-methyl-1-butanol (3MB), a microbial volatile compound released by various plant growth-promoting microbes, which has gained recent attention due to its ability to promote plant growth and enhance stress tolerance. Previous studies have shown that 3MB induces stomatal closure and enhances salt stress tolerance in plants; however, its mode of action in these processes remains poorly understood. In this study, 6-day-old Arabidopsis seedlings were fumigated with 3MB (1 mg L-1) for 3

Methyl Jasmonate Was Involved in Hydrogen Sulfide-Alleviated Cadmium Stress in Cucumber Plants Through ROS Homeostasis and Chlorophyll Metabolism.

Cadmium (Cd), as one of the most toxic nonessential elements, severely prohibits plant growth and development. Hydrogen sulfide (H2S) and methyl jasmonate (MeJA) play essential roles in plant response to abiotic stress. However, the potential mechanism of H2S and MeJA in alleviating Cd stress in plants remains unclear. In the current study, the importance and crosstalk of H2S and MeJA in the Cd tolerance of cucumber seedlings have been investigated. Our results revealed that Cd stress obviously prohibited the growth of cucumber seedlings. Optimal concentrations of H2S donor sodium hydrosulfide

Characterization of four terpene synthase genes involved in nerolidol and linalool biosynthesis in Santalum album leaves induced by methyl jasmonate.

Santalum album is highly valued for its fragrant essential oil from heartwood. Volatile terpenoids involved in aroma formation in plants can be emitted in response to a variety of environmental stresses. However, the regulatory mechanisms underpinning the response of S. album to external stresses are not yet known. In this study, the regulatory mechanism of S. album leaves was investigated after the application of methyl jasmonate (MeJA), a stressor. Eight classes of volatile organic compounds were identified in S. album leaves, including terpenes, aldehydes, alcohols, ketones, esters, benzeno

A novel approach to enhance resistance to vascular disease by expressing cell-death-inducing fungal elicitors in the xylem tissue.

Constitutive expression of hypersensitive cell-death-inducing effectors secreted by pathogens has proved to be difficult in plants. Herein, xylem-specific promoters (PVND7 or PVND2) were utilized to express three cell-death-inducing proteins (FocSIX1A, FocSIX6 or TvSm1) of fungal origin in banana plants. The regenerated plants were not only healthy but also showed improved resistance to fungal (Foc vcg0124 and Foc vcg0125) infection. These banana plants developed systemic resistance-like symptoms, including elevated jasmonic acid content, callose depositions, increased xylem wall thickness and

TREX1 activators: A novel therapeutic strategy for rheumatoid arthritis management via cfDNA clearance.

Rheumatoid arthritis (RA) is a chronic autoimmune disease characterized by systemic inflammation and progressive joint destruction. Although current treatments, including conventional disease-modifying antirheumatic drugs (DMARDs) and biologics, offer therapeutic benefits, they are often associated with drug resistance and significant adverse effects with long-term use. Our previous research identified the accumulation of circulating cell-free DNA (cfDNA) as a critical pathogenic factor in RA, contributing to immune dysregulation and sustained inflammatory responses. A B-Box (BBX) Transcription Factor from Cucumber, CsCOL9 Positively Regulates Resistance of Host Plant to Bemisia tabaci.

B-box (BBX) transcription factors play crucial roles in plant growth, development, and defense responses to biotic and abiotic stresses. In this study, we cloned a BBX transcription factor gene, CsCOL9I, from cucumber and analyzed its role in the plant's defense against the feeding of Bemisia tabaci. CsCOL9 is expressed throughout all developmental stages in cucumber, with the highest expression in the leaves. CsCOL9 is induced by B. tabaci feeding, salicylic acid (SA), methyl jasmonate (MeJA), and hydrogen peroxide (H2O2). Cucumber plants with CsCOL9 silence (TRV2-CsCOL9) and overexpression (

Geniposide and asperuloside alter the COX-2 and GluN2B receptor expression after pilocarpine-induced seizures in mice.

Asperuloside (ASP) and geniposide (GP) are iridoids that have shown various biological properties, such as reduction of inflammation, oxidative stress, and neuroprotection. The aim of this study was to investigate the mechanism of action of ASP and GP through the experimental model of pilocarpine-induced seizures. Mice were treated daily with saline, valproic acid (VPA), GP (5, 25, or 50 mg/kg), or ASP (20 or 40 mg/kg) for 8 days. Pilocarpine (PILO) treatment was administered after the last day of treatment, and the epileptic behavior was recorded for 1 h and analyzed by an adapted scale. Afte

Estrogen-dependent KCa1.1 modulation is essential for retaining neuroexcitation of female-specific subpopulation of myelinated Ah-type baroreceptor neurons in rats.

Female-specific subpopulation of myelinated Ah-type baroreceptor neurons (BRNs) in nodose ganglia is the neuroanatomical base of sexual-dimorphic autonomic control of blood pressure regulation, and KCa1.1 is a key player in modulating the neuroexcitation in nodose ganglia. In this study we investigated the exact mechanisms underlying KCa1.1-mediated neuroexcitation of myelinated Ah-type BRNs in the presence or absence of estrogen. BRNs were isolated from adult ovary intact (OVI) or ovariectomized (OVX) female rats, and identified electrophysiologically and fluorescently. Action potential (AP)

Susceptibility of epidemic viruses to neuraminidase inhibitors and treatment-emergent resistance in the Japanese 2019-20 influenza season.

OBJECTIVES: To investigate the susceptibility of epidemic influenza viruses to neuraminidase inhibitors (NAIs) and the emergence of resistant viruses after treatment, a prospective, observational study was done in the 2019-20 Japanese influenza season. METHODS: Influenza viruses were isolated before and twice after treatment, the first at day 5 and the second at day 10. The 50% inhibitory concentrations (IC50s) to oseltamivir, zanamivir, peramivir, and laninamivir were measured and compared with those of 2010-11 to 2018-19 seasons. NA amino acid sequences were analyzed by next generation sequ

Wood profiling by non-targeted high-resolution mass spectrometry: Part 1, Metabolite profiling in Cedrela wood for the determination of the geographical origin.

The determination of the geographical origin of wood can be highly relevant for several reasons: On the one hand, it can help to prevent illegal logging and timber trade, on the other hand, it is of special interest for archaeological artefacts made of wood, as well as for a variety of biological questions. For this reason, different extraction methods were first tested for the analysis of polar and non-polar metabolites using liquid chromatography coupled electrospray ionization quadrupole time-of-flight mass spectrometry (UHPLC-ESI-QTOF-MS). Disclosure of salicylic acid and jasmonic acid-responsive genes provides a molecular tool for deciphering stress responses in soybean.

Hormones orchestrate the physiology of organisms. Measuring the activity of defense hormone-responsive genes can help understanding immune signaling and facilitate breeding for plant health. However, different from model species like Arabidopsis, genes that respond to defense hormones salicylic acid (SA) and jasmonic acid (JA) have not been disclosed in the soybean crop. We performed global transcriptome analyses to fill this knowledge gap. Upon exogenous application, endogenous levels of SA and JA increased in leaves. SA predominantly activated genes linked to systemic acquired resistance and

Expression dynamics indicate the role of Jasmonic acid biosynthesis pathway in regulating macronutrient (N, P and K+) deficiency tolerance in rice (Oryza sativa L.).

Expression pattern indicates that JA biosynthesis pathway via regulating JA levels might control root system architecture to improve nutrient use efficiency (NUE) and N, P, K+ deficiency tolerance in rice. Deficiencies of macronutrients (N, P and K+) and consequent excessive use of fertilizers have dramatically reduced soil fertility. It calls for development of nutrient use efficient plants. Plants combat nutrient deficiencies by altering their root system architecture (RSA) to enhance the acquisition of nutrients from the soil. Amongst various phytohormones, Jasmonic acid (JA) is known to re

Resistance analysis of cherry rootstock 'CDR-1' (Prunus mahaleb) to crown gall disease.

BACKGROUND: Crown gall disease, caused by the pathogenic bacterium Agrobacterium tumefaciens, is responsible for extensive economic losses in orchards. Cherry rootstock 'CDR-1' (Prunus mahaleb) shows high resistance but the mechanism remains unclear. Here, we examined the morphology of pathogen-infected root neck surface, determined the activity of 10 defense-related enzymes and the content of salicylic acid (SA) and jasmonic acid (JA), and also applied transcriptome analysis, transient expression and transgenic verification to explore the crown gall resistance genes in 'CDR-1' plants. Induced tolerance to abiotic and biotic stresses of broccoli and Arabidopsis after treatment with elicitor molecules.

The plant hormones salicylic acid (SA) and jasmonic acid (JA) regulate defense mechanisms capable of overcoming different plant stress conditions and constitute distinct but interconnected signaling pathways. Interestingly, several other molecules are reported to trigger stress-specific defense responses to biotic and abiotic stresses. In this study, we investigated the effect of 14 elicitors against diverse but pivotal types of abiotic (drought) and biotic (the chewing insect Ascia monuste, the hemibiotrophic bacterium Pseudomonas syringae DC 3000 and the necrotrophic fungus Alternaria altern

Arabidopsis C-terminal binding protein ANGUSTIFOLIA modulates transcriptional co-regulation of MYB46 and WRKY33.

The apparent antagonism between salicylic acid (SA) and jasmonic acid (JA)/ethylene (ET) signalling resulting in trade-offs between defence against (hemi)biotrophic and necrotrophic pathogens has been widely described across multiple plant species. However, the underlying mechanism remains to be fully established. The molecular and cellular functions of ANGUSTIFOLIA (AN) were characterised, and its role in regulating the pathogenic response was studied in Arabidopsis. We demonstrated that AN, a plant homologue of mammalian C-TERMINAL BINDING PROTEIN (CtBP), antagonistically regulates plant res
