Human Epidermal Growth Factor Receptor 2 Quantification Using Computational Pathology to Identify Novel Biomarkers for Trastuzumab Deruxtecan-Treated Human Epidermal Growth Factor Receptor 2-Positive Gastric Cancer.

PURPOSE: Trastuzumab deruxtecan (T-DXd) is an antibody-drug conjugate approved for human epidermal growth factor receptor 2 (HER2)-positive gastric cancer (GC) and gastroesophageal junction adenocarcinoma (GEJA). Eligibility for T-DXd therapy currently relies on conventional HER2 immunohistochemistry (IHC) scoring per ASCO/CAP guidelines. Computational pathology tools such as quantitative continuous scoring (QCS) offer precise quantification of HER2 expression, enabling improved scores reflective of T-DXd's mechanism of action. METHODS: We used HER2-QCS scoring to identify biomarker signature

Postoperative Pain in Children: A Real Emergency.

The topic of acute pain, especially post-operative pain, risks being unfairly relegated to the background. Although significant progress has been made in pediatric anesthesia, with a drastic reduction in perioperative mortality and morbidity, much remains to be done in terms of organization and training. Artificial intelligence can potentially help here. Combining data from large, well-structured datasets can provide predictive models that guide patients at higher risk of developing pain. Having reliable alerts would allow resources to be used not only more effectively, but also more rationall

Automatic Segmentation of Vestibular Schwannoma From MRI Using Two Cascaded Deep Learning Networks.

OBJECTIVE: Automatic segmentation and detection of vestibular schwannoma (VS) in MRI by deep learning is an upcoming topic. However, deep learning faces generalization challenges due to tumor variability even though measurements and segmentation of VS are essential for growth monitoring and treatment planning. Therefore, we introduce a novel model combining two Convolutional Neural Network (CNN) models for the detection of VS by deep learning aiming to improve performance of automatic segmentation. METHODS: Deep learning techniques have been employed for automatic VS tumor segmentation, inclu

Role of individual and population heterogeneity in shaping dynamics of multi-pathogen shedding in an island endemic bat.

Understanding processes driving pathogen transmission in bats is critical to prevent spillovers and emergence events. Although substantial research has addressed this topic, few studies have directly examined shedding dynamics (as opposed to serological studies) and co-infection patterns using fine-scale spatio-temporal datasets. Here, based on the monitoring of 5,714 Reunion free-tailed bats (Mormopterus francoismoutoui) in 17 roosts over 24 months, we studied the co-shedding dynamics of paramyxoviruses (PMV) and Leptospira bacteria (LEPTO) in urine, and herpesviruses (HSV) in saliva. We evid

Intrapartum Care for People with Diabetes-Working towards Evidence-Based Management.

The consensus in the literature supports the need for careful monitoring and management of maternal blood glucose during labor to optimize neonatal outcomes. Guidelines generally recommend strict control of maternal blood glucose during labor, involving frequent checks, and the use of dextrose and insulin as needed. However, recent evidence has not consistently shown a strong association between strict control of blood glucose and a reduction in the rate of neonatal hypoglycemia. This raises questions about the extent to which intrapartum blood glucose control impacts neonatal hypoglycemia. Th

Early detection of mental health on social media using a hybrid Bi-LSTM-XGBoost model: a comparative study.

The case of mental health disorders has been a main topic in the clinical and psychological field. The advancement of computing studies, especially in Natural Language Processing (NLP)-a subset of Machine Learning, created a system of detection that can detect the mental health state of a person in early stage to prevent the eventuality of the worst case. This is crucial since there has been a lot of case of mental health disorder-such as depression and suicide, remains undetected and untreated-especially when the internet usage is more prevalent than ever even among the most vulnerable users,

Cross-Subject Tinnitus Diagnosis Based on Multi-Band EEG Contrastive Representation Learning.

Electroencephalogram (EEG) is an important technology to explore the central nervous mechanism of tinnitus. However, it is hard to obtain consistent results in many previous studies for the high heterogeneity of tinnitus. In order to identify tinnitus and provide theoretical guidance for the diagnosis and treatment, we propose a robust, data-efficient multi-task learning framework called Multi-band EEG Contrastive Representation Learning (MECRL). In this study, we collect resting-state EEG data from 187 tinnitus patients and 80 healthy subjects to generate a high-quality large-scale EEG datase

Integrated quantitative sensory testing and psychological phenotyping dataset for capsaicin-induced neuropathic pain susceptibility.

Experimental pain models are used to investigate the analgesic effects in healthy volunteers during the early stages of drug development. However, the predictive value of these models for clinical pain requires continuous reinforcement. One question is whether neuropathic pain patterns can be induced in healthy individuals. Another question is whether healthy volunteers can be pre-selected based on the inducibility of neuropathic-like sensory patterns under controlled experimental conditions. A third question is whether psychological traits contribute to this susceptibility. The present datase

Epidermal retinoid starvation driven by AKR1B10 orchestrates barrier disruption and hyperinflammation in atopic dermatitis.

BACKGROUND: Atopic dermatitis (AD) is driven by a feed-forward loop of epidermal barrier disruption and immune dysregulation. Although retinoic acid (RA) signaling is indispensable for epidermal homeostasis and terminal differentiation, the molecular mechanisms underlying its functional exhaustion in eczematous lesions remain incompletely elucidated. To investigate whether the aldo-keto reductase AKR1B10 mediates intracellular RA depletion and epidermal dysfunction in AD, and to evaluate the therapeutic potential of topical aldo-keto reductase inhibition. METHODS: We integrated single-cell RN

Nuclear receptor corepressor 1 is a potential diagnostic and prognostic biomarker in clear cell renal cell carcinoma.

This study investigates the potential of nuclear receptor corepressor 1 (NCOR1) as a diagnostic and prognostic biomarker for clear cell renal cell carcinoma (ccRCC). Through the analysis of data from The Cancer Genome Atlas and Gene Expression Omnibus databases, along with immunohistochemical testing of clinical samples, this study revealed that NCOR1 expression was significantly downregulated in ccRCC tissues when compared to normal tissues. This downregulation was also more pronounced in 11 other types of tumors. The results of functional enrichment analysis indicated that NCOR1-related diff

The effect of selection bias on the performance of a deep learning-based intraoperative hypotension prediction model using real-world samples from a publicly available database.

BACKGROUND: There are models to predict intraoperative hypotension from arterial pressure waveforms. Selection bias in datasets used for model development and validation could impact model performance. We aimed to evaluate how selection bias affects the predictive performance of a deep learning (DL)-based model and a model using only mean arterial pressure (MAP) as input (MAP-only model). METHODS: We used the VitalDB open dataset. A hypotensive event was defined as a MAP <65 mm Hg for 1 min. For the 'biased dataset', 'non-hypotensive events' needed to be (a) at the centre of a 'non-hypotensiv

Integrative Analyses of Biomarkers and Pathways in Oxidative Stress-Related Genes for Gestational Diabetes Mellitus.

PROBLEM: Oxidative stress (OS) plays a key role in the pathogenesis of gestational diabetes mellitus (GDM), but it was not well understood. We aimed to investigate the biomarkers and underlying mechanisms of OS-related genes in GDM. METHOD OF STUDY: The GSE103552 and GSE70493 datasets of GDM were acquired from the Gene Expression Omnibus (GEO) database. Then, oxidative stress-related differentially expressed genes (OSDEGs) were screened between GDM and normal samples from these two datasets. Further analyses were conducted by gene ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG),

A high-resolution transcriptomic and spatial atlas of cell types in the whole mouse brain.

The mammalian brain consists of millions to billions of cells that are organized into many cell types with specific spatial distribution patterns and structural and functional properties1-3. Here we report a comprehensive and high-resolution transcriptomic and spatial cell-type atlas for the whole adult mouse brain. The cell-type atlas was created by combining a single-cell RNA-sequencing (scRNA-seq) dataset of around 7 million cells profiled (approximately 4.0 million cells passing quality control), and a spatial transcriptomic dataset of approximately 4.3 million cells using multiplexed erro

Unified AI framework to uncover deep interrelationships between gene expression and Alzheimer's disease neuropathologies.

Deep neural networks (DNNs) capture complex relationships among variables, however, because they require copious samples, their potential has yet to be fully tapped for understanding relationships between gene expression and human phenotypes. Here we introduce an analysis framework, namely MD-AD (Multi-task Deep learning for Alzheimer's Disease neuropathology), which leverages an unexpected synergy between DNNs and multi-cohort settings. In these settings, true joint analysis can be stymied using conventional statistical methods, which require "harmonized" phenotypes and tend to capture cohort

XGSEA: CROSS-species gene set enrichment analysis via domain adaptation.

MOTIVATION: Gene set enrichment analysis (GSEA) has been widely used to identify gene sets with statistically significant difference between cases and controls against a large gene set. GSEA needs both phenotype labels and expression of genes. However, gene expression are assessed more often for model organisms than minor species. Also, importantly gene expression are not measured well under specific conditions for human, due to high risk of direct experiments, such as non-approved treatment or gene knockout, and then often substituted by mouse. Thus, predicting enrichment significance (on a p

Confronting false discoveries in single-cell differential expression.

Differential expression analysis in single-cell transcriptomics enables the dissection of cell-type-specific responses to perturbations such as disease, trauma, or experimental manipulations. While many statistical methods are available to identify differentially expressed genes, the principles that distinguish these methods and their performance remain unclear. Here, we show that the relative performance of these methods is contingent on their ability to account for variation between biological replicates. Methods that ignore this inevitable variation are biased and prone to false discoveries

Implementation of a deep learning model for automated classification of Aedes aegypti (Linnaeus) and Aedes albopictus (Skuse) in real time.

Classification of Aedes aegypti (Linnaeus) and Aedes albopictus (Skuse) by humans remains challenging. We proposed a highly accessible method to develop a deep learning (DL) model and implement the model for mosquito image classification by using hardware that could regulate the development process. In particular, we constructed a dataset with 4120 images of Aedes mosquitoes that were older than 12 days old and had common morphological features that disappeared, and we illustrated how to set up supervised deep convolutional neural networks (DCNNs) with hyperparameter adjustment. The model appl

Predicting Resolvin D1 Pharmacokinetics in Humans with Physiologically-Based Pharmacokinetic Modeling.

Sjögren's syndrome (SS) is an autoimmune disease with no effective treatment options. Resolvin D1 (RvD1) belongs to a class of lipid-based specialized pro-resolving mediators that showed efficacy in preclinical models of SS. We developed a physiologically-based pharmacokinetic (PBPK) model of RvD1 in mice and optimized the model using plasma and salivary gland pharmacokinetic (PK) studies performed in NOD/ShiLtJ mice with SS-like features. The predictive performance of the PBPK model was also evaluated with two external datasets from the literature reporting RvD1 PKs. The PBPK model adequately
