Human dendritic cell targeting peptide can be targeted to porcine dendritic cells to improve antigen capture efficiency to stimulate stronger immune response.

Dendritic cells (DCs) play a key role in the natural recognition of pathogens and subsequent activation of adaptive immune responses due to their potent antigen-presenting ability. Dendritic cell-targeting peptide (DCpep) is strongly targeted to DCs, which often express antigens, to enhance the efficacy of vaccines. Our previous study showed that recombinant Lactobacillus expressing human DCpep could significantly induce stronger immune responses than recombinant Lactobacillus without DCpep, but the mechanism remains unclear. In this study, the mechanism by which DCpep enhances the immune resp

A novel dendritic cell-induced model of erosive inflammatory arthritis: distinct roles for dendritic cells in T cell activation and induction of local inflammation.

Transferring collagen-pulsed, bone marrow-derived dendritic cells (DCs) into congenic DBA/1 recipient mice produced arthritis in joints adjacent to the site of DC transfer and could be inhibited by treatment with TNF antagonists. Disease was Ag specific, as transfer of control, unpulsed DCs, or DCs pulsed with OVA did not produce arthritis. In contrast to other experimental arthritis models, DC-induced arthritis localized to the site of injection and did not spontaneously generalize to uninvolved joints, despite the demonstration of circulating collagen-reactive T cells. Similarly, transfer of

Production of functionally deficient dendritic cells from HTLV-I-infected monocytes: implications for the dendritic cell defect in adult T cell leukemia.

Adult T cell leukemia (ATL) is induced by an infection with human T lymphotropic virus type I (HTLV-I) and is accompanied by immunodeficiency. Monocyte-derived immature dendritic cells (DCs) donated by 11 ATL patients were suppressed in the ability to take up fluorescein isothiocyanate (FITC)-dextran and were down-regulated in the expression of CD1a and CD86 antigens (Ags). Monocytes from the patients showed impaired expression of CD14 and HLA-DR Ags. These results suggest intrinsic abnormalities of monocytes and a defect of DC maturation in ATL patients. Therefore, we examined the influence o

Expression of dendritic cell lysosome-associated membrane protein and dendritic cell-specific intercellular adhesion molecule-3 grabbing nonintegrin in condyloma acuminatum lesions.

OBJECTIVE: Retrospective, observational study to explore the role of dendritic cells (DCs) in condyloma acuminatum lesions (genital warts) and their relationship with duration of the disease. METHODS: Condyloma acuminatum lesion samples were collected from male patients with the condition and compared with normal foreskin samples from male volunteers. Cellular locations of dendritic cell lysosome-associated membrane protein (DC-LAMP) and dendritic cell-specific intercellular adhesion molecule-3 grabbing nonintegrin (DC-SIGN) were detected using immunohistochemistry. Levels of both proteins we

Peripheral T-cell lymphoma with extensive dendritic cell network mimicking follicular dendritic cell tumor: a case report with pathologic, immunophenotypic, and molecular findings.

Peripheral T-cell lymphoma (PTCL) with a nodular pattern of growth is uncommon and may be misdiagnosed initially as a B-cell lymphoma or reactive process. We report a case of a rapidly growing PTCL with a distinctly nodular pattern in an axillary lymph node from an 89-year-old man. Immunohistochemical stains for CD21, CD23, and CD35 highlighted an extensive dendritic cell network that imparted the nodular appearance and, in addition, was associated intimately with the neoplastic cells. The neoplastic cells otherwise had an immunophenotype similar to previously reported cases of PTCL with a nod

The dendritic cell-specific CC-chemokine DC-CK1 is expressed by germinal center dendritic cells and attracts CD38-negative mantle zone B lymphocytes.

DC-CK1 (CCL18) is a dendritic cell (DC)-specific chemokine expressed in both T and B cell areas of secondary lymphoid organs that preferentially attracts CD45RA(+) T cells. In this study, we further explored the nature of DC-CK1 expressing cells in germinal centers (GCs) of secondary lymphoid organs using a newly developed anti-DC-CK1 mAb. Immunohistochemical analysis demonstrated a remarkable difference in the number of DC-CK1 expressing cells in adjacent GCs within one tonsil, implicating that the expression of DC-CK1 in GCs depends on the activation and/or progression stage of the GC reacti

Sézary syndrome patients demonstrate a defect in dendritic cell populations: effects of CD40 ligand and treatment with GM-CSF on dendritic cell numbers and the production of cytokines.

Sézary syndrome (SzS) is an advanced form of cutaneous T-cell lymphoma associated with involvement of the peripheral blood by malignant T cells. The disease is defined by impaired cell-mediated immunity and the production of interferon-gamma (IFN-gamma) and interleukin-2 (IL-2), possibly as a result of deficient IL-12 production. To understand the mechanism of this impairment, we examined the composition and function of dendritic cells and monocytes in the blood of SzS patients with different levels of peripheral blood tumor burden. Consistent with our previous observations, numbers of monocyt

Carfilzomib preserves dendritic cell phenotype and function compared with bortezomib.

INTRODUCTION: Despite major therapeutic advances, multiple myeloma (MM) remains largely incurable. Moreover, side effects and toxicities of treatments compromise patients' quality of life. Dendritic cell (DC)-based cancer vaccines can induce long-term tumor-specific immune responses and are generally well tolerated. To improve the potency of this immunotherapeutic approach, researchers are now exploring combination strategies that integrate approved agents with DC-based vaccines. Proteasome inhibitors (PIs), including bortezomib and carfilzomib, are the standard of care for MM. PIs have been s

Dendritic cells as orchestrators in the allergen-specific immunotherapy of allergic diseases.

Dendritic cells (DCs) play a pivotal role in regulating immune responses, encompassing both tolerance and activation. Dysregulation of DC function is fundamental in the development of allergic conditions, underscoring their significance as a major focus for allergen-specific immunotherapy (AIT). This review systematically delineates the intricate landscape of DC subsets, comprising conventional DCs, plasmacytoid DCs, monocyte-derived DCs, and Langerhans cells, elucidating their specific contributions to either fostering Th2-driven inflammation or driving regulatory T cell generation. We dissec

The complement-dendritic cell-endothelial cell crosstalk in vascular inflammation.

Inflammation is a highly co-ordinated process involving multiple immune components, including the complement system, dendritic cells (DCs), and endothelial cells (ECs). All of them play pivotal roles in immunity and host defence and have a significant impact on cardiovascular health and disease. While individual functions of the complement system, DCs and ECs are well-characterized, their multifaceted interplay in modulating inflammation and disease is insufficiently elucidated. The complement system exerts predominantly pro-inflammatory effects, influencing the immune, cardiovascular, and ner

Dendritic Cell α-Ketoglutarate Regulates Tfh Polarization in Allergy.

Initiation of allergic responses requires activation of specialized subsets of T follicular helper (Tfh) cells called Tfh13 cells. Differentiation of these cells from naïve CD4+ T cells is induced by antigen presenting cells, such as type 2 conventional dendritic cells (cDC2s), following allergen exposure. Here we show that allergen stimulation induces a distinct metabolic program in cDC2s characterized by increased glutaminolysis which results in intracellular accumulation of α-ketoglutarate. Inhibition of glutaminolysis suppresses Tfh13 cell differentiation and promotes inhibitory T follicul

Mechanically driven Li dendrite penetration in garnet solid electrolyte.

All-solid-state batteries promise improved safety and higher energy density by replacing flammable liquid electrolytes and graphite anodes with solid electrolytes and lithium metal1-4. However, the penetration of soft lithium dendrites into hard ceramic electrolytes remains a substantial obstacle to realizing all-solid-state lithium metal batteries5-7. The mechanism by which mechanically soft lithium dendrites fracture hard ceramic electrolytes remains under debate7-10 owing to the challenges of characterizing nanoscale lithium distribution and its microstructure at the dendrite tip11. Here we…

Engineered immunosuppressive dendritic cells protect against cardiac remodelling.

Heart failure remains a leading cause of morbidity and mortality, yet no approved therapies effectively prevent or reverse pathological cardiac fibrosis and the associated decline in cardiac function1-4. Chronic inflammation is a central driver of pathological fibrosis after ischaemic or haemodynamic stress, but strategies that locally rebalance injurious and reparative immune responses without systemic immunosuppression are lacking5,6. Dendritic cells (DCs) are key regulators of immune activation and tolerance, providing an opportunity for therapeutic immune reprogramming in cardiac diseases7

Protocadherin γC4 regulates neuronal survival and dendritic self-avoidance.

Animal models are indispensable for linking human genetic findings to disease mechanisms. Mutations in protocadherin gamma C4 (γC4), one of the 22 isoforms encoded by the protocadherin-γ (Pcdh-γ) gene cluster, cause a human neurodevelopmental syndrome with progressive microcephaly, seizures, and intellectual disability. Here, we established a γC4 mutant mouse model that exhibits motor dysfunction, seizures, reduced brain size, and increased embryonic neuronal apoptosis. Using DOMINO (Double Mutation-Induced Open Reading Frame Switch), a two-step CRISPR/Cas9-based genome-editing strategy, we al

Transcription factor Etv3 controls the tolerogenic function of dendritic cells.

Dendritic cells (DCs) facilitate the maintenance of immunological tolerance in the steady state. We report that transcription factor Etv3 is preferentially expressed in mature DCs, including tissue-derived migratory DCs (migDCs), and facilitates their homeostatic maturation and CCR7-dependent migration. Mice with global or DC-specific deletion of Etv3 manifested the expansion of CD25low regulatory T (Treg) cells, spontaneous activation of conventional T cells, and multiorgan T cell infiltration. Etv3 deficiency exacerbated TLR7-driven systemic lupus erythematosus (SLE)-like disease, supporting
