Cerebral vasoreactivity by transcranial Doppler in carbon disulfide poisoning cases in Korea.

The abnormalities of cerebral vasoreactivity were evaluated in CS2 poisoning cases. In 34 retired workers with CS2 poisoning (case group) and 20 healthy individuals (control group), blood flow velocities were measured in the middle cerebral arteries (MCA) by transcranial Doppler. Compared with the control group, the case group showed lower mean values of blood flow velocities and pulsatile indices. The differences in CO2 reactivities of both groups were remarkable (1.10-2.19% decrease/mmHg CO2 for cases and 3.35-5.08 for controls). Multiple regression analyses were conducted to adjust the effe

A mortality study of workers with reported chronic occupational carbon disulfide poisoning.

A mortality cohort study was carried out on 2,291 workers, including 169 women, with chronic occupational CS2 poisoning diagnosed during the years 1970-90. Information on vital status was available for 98.2% as of December 31, 1992. Mortality assessment was based on the standardized mortality ratio using the person-years method. The general population of Poland was the reference population. The number of subjects who died during the period of observation accounted for 658 men and 21 women. The analysis of mortality in male subjects showed statistically significant excess of deaths from the cir

Carbon disulfide mediated protein cross-linking by N,N-diethyldithiocarbamate.

N,N-Diethyldithiocarbamate and its disulfide are used as pesticides, in industrial processes, and as therapeutic agents, providing numerous opportunities for human exposure. Animal studies and in vitro investigations have demonstrated adverse effects following exposure to dithiocarbamates. The ability of dithiocarbamates to decompose to parent amine and CS2 suggests that these adverse effects may be mediated through release of CS2. The toxicity of CS2 is well established, and covalent cross-linking of proteins has been presented as a potential molecular mechanism of CS2 induced neuropathy. In

Occurrence of autoantibody to protein disulfide isomerase in rats with xenobiotic-induced hepatitis.

The occurrence of an autoantibody to protein disulfide isomerase (PDI) in rats after administration of various hepatotoxic drugs was investigated by immunoblotting and radioimmunoassay. An anti-PDI autoantibody was detected with high frequency in rats treated with D-galactosamine, acetaminophen with diethylmaleate, and carbon tetrachloride with diethylmaleate. The antibody-positive rate was relatively low in the groups of rats given carbon tetrachloride, acetaminophen or DL-ethionine alone. The anti-PDI antibody was not detected in rats treated with diethylmaleate alone. Although the mechanism

Carbon disulfide exposure and neurotoxic sequelae among viscose rayon workers.

In Norway's only viscose rayon plant, carbon disulfide (CS2) concentrations in ambient air usually were between 30 and 50 mg/m3 during the first 23 years of production. From 1970/1971 until the factory was closed in 1982, corresponding values were 10-25 mg/m3. Through all of these years, high peak exposures of CS2 and H2S occurred. In 1986, 16 of the 24 men still at work in 1982 and with at least 10 years' experience in the spinning room agreed to participate in this study. Clinical neurological examination demonstrated abnormalities in 15; neuropsychological tests showed impairments of probab

Study of the effects of simultaneous exposure to noise and carbon disulfide on workers' hearing.

The aim of this study was to explore the effects of simultaneous exposure to noise and carbon disulfide on workers' hearing and balance. The study was conducted by interviews and by audiometric and balance tests on workers in a rayon factory in the city of São Paulo, Brazil (n = 258). The workers studied had a history of exposure both to excessive noise levels (86-89 dBA) and to excessive levels of carbon disulfide (89.92 mg/m3). The percentage of hearing loss found was much higher than expected for this occupational activity, which reinforced the possible connection between the exposure to no

Hepatic cholesterol metabolism as a function of carbon disulfide concentration and treatment with phenobarbital.

Male F344 rats were exposed to carbon disulfide (CS2) at 0, 30, 75, 150, 300, or 600 ppm for 6 hr by inhalation in the presence or absence of 0.1% phenobarbital (PB) in the drinking water starting 5 days before exposure to CS2. Exposure to 600 ppm CS2 only resulted in a decrease in hepatic cholesterol synthesis and an increase in the liver-to-body-weight ratio (relative liver weight); however, it caused no histopathological damage and had little or no consistent effect on the concentration of hepatic cholesterol or on hepatic water content. Treatment with PB alone resulted in increases in the

[Biochemical and histological research on lung tissue in experimental carbon disulfide exposure].

During an experiment on two species of test animals (guinea pigs and white male rats) is studied the effect of carbon sulfide on the biochemical and structural characteristic of the lung. Duration of the experiments 30 days with guinea pigs and 90 days-white male rats. A complex of biochemical and histological methods is applied. Disturbances in the metabolic processes of the lung are established: a) raised activity of the hydrolytic enzymes; b) increased activity of the glycolytic enzymes and c) stronger protein synthesis. The observed metabolic disturbances are based, most probably, on the c

Effect of carbon disulfide on the anticholinesterase action of several organophosphorus insecticides in mice.

Effect of carbon disulfide (CS2) on toxic action of 11 organophosphorus (OP) insecticides were examined by determining the plasma cholinesterase activity in mice. CS2 pretreatment potentiated the anticholinesterase action of parathion and EPN, but suppressed that of dimethoate and diazinon. CS2 had no significant effect or a slightly suppressive effect on the other compounds. Some of these effects were contrasted with the reported alteration of the toxicity following phenobarbital pretreatment. CS2 administration suppressed both detoxification and activation of parathion and EPN by liver micro

[Early behavioral and neurofunctional deviations following prenatal carbon disulfide exposure].

The effect of prenatal carbon disulphide exposure with a concentration 10 and 0.33 mg/m3 is studied (MAC for a working zone and atmospheric air resp.) upon the behavioural and neurofunctional development of the generation. A neurotoxicological screening was employed for the assessment of the early postnatal development. It was established that carbon disulphide with a MAC level of the working zone, disturbed the postnatal development without inducing congenital malformations, inducing buit sensory, neurofunctional and behavioural deviations. The results stress upon the high sensitivity of the

Carbon disulfide neuropathy in rats. A morphological and ultrastructural study of degeneration and regeneration.

The aim of this study was to elucidate the site and detailed nature of peripheral nerve damage induced in the rat by chronic CS2 inhalation exposure in the light of the relationship between pathological and neurophysiological data. Adult male rats were exposed to 700 ppm of CS2 2 h/d, 5 d/week for 12 weeks and then followed-up for 18 weeks. The first alteration observed was a decrease in the nerve conduction velocity, discovered after only 3 weeks of exposure. Pathological lesions were first observed in the 10th week and consisted of a typical "giant axon" axonopathy. Obvious pathological lesi

[Effect of carbon disulfide on the metabolism of connective tissue of rats' aorta and lungs].

Female Wistar rats were exposed to CS2 for 13 months. Afterwards collagen and elastin content in aorta and lungs was determined. In addition, collagen and non-collagen protein biosynthesis was determined using 14-C glycine incorporation to proteins. Collagen and elastin content in aorta wall and elastin concentration in lungs were found to be increased. Isotopic method indicated that the specific activity of collagen in aorta wall was increased and activity of pulmonary collagen and non-collagen proteins of aorta and lungs was decreased. The above is indicative of: lowered biosynthesis of nonc

[Lysosomal and cytoplasmic enzymatic activity under the experimental action of carbon disulfide].

Activity of lysosomal enzymes and content of carbohydrate containing biopolymers (glycosaminoglycans, glycoproteins) were altered in various tissues (liver, kidney, brain, aorta) of experimental animals intoxicated with carbon disulfide. Possible role of the impairments observed, related to neurotropic and hepatotoxic effects, is discussed. Subsequent development of atherogenic and embryotoxic effects of carbon disulfide--one of contaminators of surroundings--are considered.
