For pathogens that must be treated with combinations of antibiotics and acquire resistance through genetic mutation, knowledge of the order in which drug-resistance mutations occur may be important for determining treatment policies. Diagnostic specimens collected from patients are often available; this makes it possible to determine the presence of individual drug resistance-conferring mutations and combinations of these mutations. In most cases, these specimens are only available from a patient at a single point in time; it is very rare to have access to multiple specimens from a single pati

The rapid spread of antimalarial drug resistance in Plasmodium falciparum over the past few decades has necessitated intensive monitoring of such resistance for an effective malaria control strategy. P. falciparum dihydropteroate synthase (Pfdhps) and P. falciparum dihydrofolate reductase (Pfdhfr) genes act as molecular markers for resistance against the antimalarial drugs sulphadoxine and pyrimethamine, respectively. Resistance to pyrimethamine which is used as a partner drug in artemisinin combination therapy (ACT) is associated with several mutations in the Pfdhfr gene, namely A16V, N51I, C

Salmonella enterica subsp. enterica serovar Typhi, a human enteric pathogen causing typhoid fever, developed resistance to multiple antibiotics over the years. The current study was dedicated to understand the multi-drug resistance (MDR) mechanism of S. enterica serovar Typhi CT18 and to identify potential drug targets that could be exploited for new drug discovery. We have employed gene interaction network analysis for 44 genes which had 275 interactions. Clustering analysis resulted in three highly interconnecting clusters (C1-C3). Functional enrichment analysis revealed the presence of drug

UNLABELLED: Multiple myeloma is marked by recurrent cytogenetic abnormalities and mutations that accumulate as the disease progresses. In this study, we sought to elucidate the transitions driving tumorigenesis and therapy resistance in multiple myeloma using a unique cohort of nearly 900 patients spanning premalignant to late-stage refractory multiple myeloma, comprehensively characterized at molecular and clinical levels. Waves of epigenetic dysregulation drove these critical transitions. In this paradigm, genomic and cytogenetic events unlocked epigenetic plasticity, reshaping multiple myel

Artemisinin (ART) has been found to exert anti-tumor activity by regulating the cell cycle, inducing apoptosis, inhibiting angiogenesis and tumor invasion and metastasis. Its derivatives (ARTs) can regulate the expression of drug-resistant proteins and reverse the multidrug resistance (MDR) of tumor cells by inhibiting intracellular drug efflux, inducing apoptosis and autophagy of tumor cells, thus enhancing the sensitivity of tumor cells to chemotherapy and radiotherapy. Recent studies have shown that nanodrugs play an important role in the diagnosis and treatment of cancer, which can effecti

Cancer is highly heterogeneous in nature and characterized by abnormal, uncontrolled cells' growth. It is responsible for the second leading cause of death in the world. Nanotechnology is explored profoundly for sitespecific delivery of cancer chemotherapeutics as well as overcome multidrug-resistance (MDR) challenges in cancer. The progress in the design of various smart biocompatible materials (such as polymers, lipids and inorganic materials) has now revolutionized the area of cancer research for the rational design of nanomedicine by surface engineering with targeting ligands. The present study was set up to determine vancomycin tolerance in Colombian, multiresistant, invasive, paediatric isolates of Streptococcus pneumoniae. A total of seven clinical isolates of penicillin-resistant S. pneumoniae, serotype 14, corresponding to the Spain(9v)-3 clone, were studied. Isolates were grown in Todd Hewitt broth at 37 degrees C. Growth (A(595)) and cell viability (cfu/ml) were monitored hourly for 4 h after addition of vancomycin at 10x MIC. S. pneumoniae R6 was used as a control for growth. For R6, the mean decrease in A(595) and cell viability (log(10) cfu/ml) after 4 h o

Drug resistance is a common concern for the development of novel antiviral, antimicrobial and anticancer therapies. To overcome this problem, several strategies have been developed, many of which involving the theme of this review, the use of structure-based drug design (SBDD) approaches. These include the successful design of new compounds that target resistant mutant proteins, as well as the development of drugs that target multiple proteins involved in specific biochemical pathways. Finally, drug resistance can also be considered in the early stages of drug discovery, through the use of str

BACKGROUND: Infections caused by bacteria such as multidrug resistant (MDR) Acinetobacter spp. and methicillin-resistant Staphylococcus aureus (MRSA) constitute a worldwide pandemic. Without gathering information about these strains, we cannot reduce the morbidity and mortality due to infections caused by these notorious bugs.
METHODS: This study was conducted to identify the status of MDR Acinetobacter spp. and MRSA in a tertiary care centre of Nepal. Sputum, endotracheal aspirate and bronchial washing specimens were collected and processed from patients suspected of lower respiratory tract

Drug resistance in tuberculosis is a growing global problem. The emergence of multi-drug resistant tuberculosis cases, particularly in the 1990s, has become an important health problem and threatens tuberculosis control worldwide. Resistance to isoniazid and rifampicin, two of the most potent anti-tuberculosis drugs currently available, in multi-drug resistant tuberculosis cases is clinically quite important. The treatment of multi-drug resistant tuberculosis requires prolonged use of costly second-line drugs with significant toxic potentials under supervision and long-term hospitalization of

Tuberculosis is a deadly airborne disease caused by Mycobacterium tuberculosis. Drug-resistant tuberculosis presents significant challenges for treatment and control of the disease. Resistant strains of Mycobacterium tuberculosis arise from specific mutations in the bacterium. Identification and characterization of resistance-associated mutations are crucial for effective treatment strategies because the first- and second-line drugs for the disease target distinct genes in the bacterium and serve different purposes. Our study developed a machine learning prediction model to analyze mutations a

Antibiotic combination therapy has a critical role in limiting emergent antibiotic resistance in bacterial pathogens. Collateral sensitivity (CS), resistance to one antibiotic that is inextricable from sensitivity to another antibiotic, presents an opportunity for combinations explicitly selecting against resistance. Complementing efforts to select against resistance, differential genetic vulnerability mapping unravels dependencies unique for resistant strains. In this review, we discuss the potential implications of CS and genetic vulnerability data for the design of bespoke antibiotic combin

Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), is a global health threat. Despite advances in diagnostics and treatment regimens, it infects millions annually. It remains a significant global health challenge, particularly due to the appearance of resistance towards anti-tuberculosis drugs (ATD), mainly multidrug-resistant (MDR) and extensively drug-resistant (XDR) strains. Mtb poses resistance towards the currently used first- and second-line regimen. Recent advancements in drug development have introduced therapeutic options focussed on improving treatment outcomes for both d

Indeed, multi-drug resistance (MDR) is a significant obstacle to effective chemotherapy. The overexpression of ATP-binding cassette (ABC) membrane transporters is a principal cause of enhanced cytotoxic drug efflux and treatment failure in various types of cancers. At cellular level, the pumps of ABC family regulate the transportation of numerous substances including drugs in and out of the cells. In past, the overexpression of ABC pumps suggested a well-known mechanism of drug resistance in cancers as well as infectious diseases. In oncology, the search for new compounds for the inhibition of

Within a relatively short period of time after the first antimicrobial drugs were introduced, bacteria began exhibiting varying degrees of resistance. The excessive use (and abuse) of antibiotics in agriculture, and in both human and veterinary medicine, has played a critical causative role in the development of antibiotic resistance, which is now recognized as a global public health threat. Increasing concern over this issue should impact the practice of cutaneous medicine and surgery, as dermatologists can easily adopt new healthcare delivery patterns that might reduce the development of ant
