Vaccine Formulation and Optimization for Human Herpes Virus-5 through an Immunoinformatics Framework.

In the current situation, the importance of vaccines for viral diseases has become the need of the hour. The scientific community in the field of virology has taken it upon themselves to develop vaccines for viral infections before an epidemic or pandemic situation arises. Human herpes virus-5 is an emerging situation that has alarming cases with major health concerns, including congenital impairments and infections leading to cancer states. Vaccination is the route most likely to succeed in the battleground with viral infections and consequences. Hence in the present manuscript, we have formu

T cell mechanisms in the immunopathogenesis of viral hepatitis B and C.

Considerable evidence suggests that immune mechanisms are involved in the pathogenesis of both hepatitis B and C. Both CD4+ and CD8+ T cell responses to viral antigens are important mechanisms that may be responsible for the hepatocyte damage in hepatitis B and C. CD4+ T cell proliferative responses to hepatitis B core antigen (HBcAg) in terms of stimulation index are correlated with hepatitis activity. These responses can be demonstrated in both adult and paediatric patients, and are more vigorous in patients with acute self-limited hepatitis B than in patients with chronic hepatitis B. Patie

Affinity-matured B cell responses neutralizing type-I interferons underlie severe viral infections.

Autoantibodies neutralizing type-I interferons (AAN-I-IFNs) emerge as global, common, and strong determinants of a growing number of severe viral diseases. We report that AAN-I-IFNs+ patients with life-threatening COVID-19 pneumonia harbor circulating type-I IFN-specific B cells indistinguishable from patients bearing T cell tolerance defects of genetic origin. This autoimmune response mobilizes a highly diverse and stable circulating B cell response that is detected prior to severe viral infection and acquires high affinity and neutralization potential to type-I IFNs through extended somatic

Bioorthogonal click chemistry to visualize an immunogenic HLA-A2-restricted hepatitis B virus epitope in human monocyte-derived dendritic cells.

Peptide-based therapeutic vaccines exploit cross-presentation by dendritic cells for the induction of effective T cell responses. Their clinical success, however, has been limited due to incomplete understanding of antigen processing and presentation (APP). Bioorthogonal chemistry (BOC) uses chemical "click" reactions that can be performed selectively without interference of the cellular environment. A "click handle" can be incorporated into a biomolecule with only minor effects on its characteristics, allowing selective visualization and tracking of the biomolecule. We generated 10 analogues

Identification of transiently produced IgG linear epitopes in Senecavirus A for differentiating infected from vaccinated animals.

Differentiating infected from vaccinated animals (DIVA) is critical for disease eradication and emerging infection surveillance. Senecavirus A (SVA), which causes vesicular disease and neonatal mortality in pigs, is clinically indistinguishable from foot-and-mouth disease virus (FMDV), posing significant economic risks to swine production. Therefore, a reliable DIVA diagnostic method is urgently needed for accurate SVA detection. In this study, we employed the IgG sero-dynamic curves to aid epitope discovery (IsDAED) approach, utilizing peptide microarrays to identify transiently produced IgG

Identification of PD-1-PD-L1 blockade epitopes in vitro utilizing porcine immunoglobulin and heterologous Fc-fused protein.

Programmed death 1 (PD-1) and its ligand, programmed death ligand 1 (PD-L1), function as pivotal immune checkpoints. Numerous studies have demonstrated the association between the malignant progression of various swine diseases and aberrant expression of PD-1 and PD-L1, hence the development and screening of high-affinity porcine PD-1 and PD-L1 monoclonal antibodies (mAbs) holds substantial significance for advancing research and therapeutic interventions. In this study, we produced porcine PD-1 and PD-L1 mAbs which exhibited robust reactivity in western blot (WB), indirect immunofluorescence

AminoAcid3D: An Algorithm for Designing HLA Single Antigen Panels and Predicting the Essential Regions of Epitopes Based on Antibody Reactivity Profiles.

The highly polymorphic yet homologous nature of HLA antigens (Ags) constitutes a milieu of public and private B-cell epitopes of varying degrees far exceeding the coverage of the existing cross-reactive groups (CREGs). To support the on-going efforts in updating serological categorisation and designing the next generation single antigen bead (SAB) panel of higher resolution and broader population coverage, an algorithm named AminoAcid3D (AA3D) has been developed as a research tool for designing SAB panels and predicting which amino acid residues (termed 'eps') could be involved in Ag-antibody

Pepscan and bioinformatic strategies for identification of potential B-cell epitopes for a peptide-based vaccine for tick control.

The cattle tick Rhipicephalus microplus poses a major problem to the livestock industry worldwide, with acaricides resistance presenting an increasing challenge. On other hand, vaccination has been suggested as a better strategy for tick control, and peptide-based vaccines could be developed to target multiple tick antigens. Nevertheless, there are still limitations to the identification of epitopes in tick candidate antigens, as the bioinformatics tools currently available were developed almost exclusively based on mammalian genomes. Therefore, improving the performance of B-cell epitope pred

Isolation and epitope characterization of human monoclonal antibodies to hepatitis C virus core antigen.

In this study we describe the establishment of two hybridoma cell lines secreting human monoclonal antibodies to the 22-kD nucleocapsid protein (core, p22) of the hepatitis C virus (HCV). For this purpose we isolated B lymphocytes from an anti-HCV positive blood donor and infected them with Epstein-Barr (EBV). We obtained several lymphoblastoid cell clones secreting antibodies to the recombinant HCV core protein. The B-cell cultures were oligoclonally expanded and two of them were fused with the (mouse:human) heteromyeloma cell line K6H6/B5. The resulting stable hybridomas produce antibodies o

Oxidation-Specific Epitopes (OSEs) Dominate the B Cell Response in Murine Polymicrobial Sepsis.

In murine abdominal sepsis by colon ascendens stent peritonitis (CASP), a strong increase in serum IgM and IgG antibodies was observed, which reached maximum values 14 days following sepsis induction. The specificity of this antibody response was studied in serum and at the single cell level using a broad panel of bacterial, sepsis-unrelated as well as self-antigens. Whereas an antibacterial IgM/IgG response was rarely observed, studies at the single-cell level revealed that IgM antibodies, in particular, were largely polyreactive. Interestingly, at least 16% of the IgM mAbs and 20% of the IgG

[Malignant histiocytosis: a study on clinicopathological features and cell origin].

Thirty-one autopsy cases previously diagnosed as malignant histiocytosis (MH) were studied by means of immunohistochemical staining. Antibodies detecting the formalin resistant epitopes on T-cells, B-cells and those of histiocyte/monocyte origin were used. It was shown that the malignant histiocytes reacted only to the cell markers derived from histiocyte/monocyte, and only a part of lymphocytes showed positive reaction to the T and B cell markers. It is suggested that the histiocyte/monocyte lineage is the possible origin of the malignant proliferating cells in MH. The clinicopathological fea

Designing of cytotoxic T lymphocyte-based multi-epitope vaccine against SARS-CoV2: a reverse vaccinology approach.

SARS-CoV2 is a single-stranded RNA virus, gaining much attention after it out broke in China in December 2019. The virus rapidly spread to several countries around the world and caused severe respiratory illness to humans. Since the outbreak, researchers around the world have devoted maximum resources and effort to develop a potent vaccine that would offer protection to uninfected individuals against SARS-CoV2. Reverse vaccinology is a relatively new approach that thrives faster in vaccine research. In this study, we constructed Cytotoxic T Lymphocytes (CTL)-based multi-epitope vaccine using h

Beyond the state of the art of reverse vaccinology: predicting vaccine efficacy with the universal immune system simulator for influenza.

When it was first introduced in 2000, reverse vaccinology was defined as an in silico approach that begins with the pathogen's genomic sequence. It concludes with a list of potential proteins with a possible, but not necessarily, list of peptide candidates that need to be experimentally confirmed for vaccine production. During the subsequent years, reverse vaccinology has dramatically changed: now it consists of a large number of bioinformatics tools and processes, namely subtractive proteomics, computational vaccinology, immunoinformatics, and in silico related procedures. However, the state

In silico Evaluation of PLAC1-fliC As a Chimeric Vaccine against Breast Cancer.

BACKGROUND: Breast cancer is one of the most prevalent cancers among women. Common cancer treatment methods are not effective enough, and there is a need for a more efficient treatment procedure. Cancer vaccine is a novel immunotherapy method that stimulates humoral and/or cellular immunity against cancer. Placenta-specific protein 1 (PLAC1) is a cancer/testis antigen, prevalent in breast cancer and rarely found in normal tissues. FliC, as a bacterial adjuvant, when fused to PLAC1 can elicit humoral and cellular responses. Therefore, PLAC1-fliC is a chimeric protein, which can be considered a

pcDNA3.1-rpfB or pcDNA3.1-rpfD recombinant DNA prevents Mycobacterium tuberculosis infection in BALB/c mice.

Tuberculosis (TB) is caused by an infection with Mycobacterium tuberculosis. The Bacille Calmette-Guérin (BCG) vaccine is used to treat TB. However, its efficacy varies in different countries. A new TB vaccine is urgently required to inhibit the spread of TB infection. DNA vaccines are promising for providing an immune response against the disease. The resuscitation-promoting factors B and D (rpfB/rpfD) genes are promising vaccine candidates. In this study, the vaccine candidates pcDNA3.1-rpfB and pcDNA3.1-rpfD were evaluated for their ability to inhibit M. tuberculosis infection in BALB/c mic

β1-Adrenergic Receptor Contains Multiple IAk and IEk Binding Epitopes That Induce T Cell Responses with Varying Degrees of Autoimmune Myocarditis in A/J Mice.

Myocarditis/dilated cardiomyopathy (DCM) patients can develop autoantibodies to various cardiac antigens and one major antigen is β1-adrenergic receptor (β1AR). Previous reports indicate that animals immunized with a β1AR fragment encompassing, 197-222 amino acids for a prolonged period can develop DCM by producing autoantibodies, but existence of T cell epitopes, if any, were unknown. Using A/J mice that are highly susceptible to lymphocytic myocarditis, we have identified β1AR 171-190, β1AR 181-200, and β1AR 211-230 as the major T cell epitopes that bind major histocompatibility complex clas

Identification and characterization of nematode specific protective epitopes of Brugia malayi TRX towards development of synthetic vaccine construct for lymphatic filariasis.

Although multi-epitope vaccines have been evaluated for various diseases, they have not yet been investigated for lymphatic filariasis. Here, we report for the first time identification of two immunodominant B epitopes (TRXP1 and TRXP2) from the antioxidant Brugia malayi thioredoxin by studying their immune responses in mice model and human subjects. TRXP1 was also found to harbor a T epitope recognized by human PBMCs and mice splenocytes. Further, the epitopic peptides were synthesized as a single peptide conjugate (PC1) and their prophylactic efficacy was tested in a murine model of filarias

Targeting the B-cell receptor signaling pathway in B lymphoid malignancies.

PURPOSE OF REVIEW: Normal B cells that have failed to productively rearrange immunoglobulin V region genes encoding a functional B-cell receptor (BCR) are destined to die. Likewise, the majority of B-cell malignancies remain dependent on functional BCR signaling, whereas in some subtypes BCR expression is missing and, apparently, counterselected. Here, we summarize the recent experimental evidence for the importance of BCR signaling and clinical concepts to target the BCR pathway in B-cell leukemia and lymphoma. RECENT FINDINGS: Although the dependency on pre-BCR signaling in pre-B acute lymp
