A novel injectable hydrogel containing polyetheretherketone for bone regeneration in the craniofacial region.

Polyetheretherketone (PEEK) is an organic material introduced as an alternative for titanium implants. Injectable hydrogels are the most promising approach for bone regeneration in the oral cavity to fill the defects with irregular shapes and contours conservatively. In the current study, injectable Aldehyde-cellulose nanocrystalline/silk fibroin (ADCNCs/SF) hydrogels containing PEEK were synthesized, and their bone regeneration capacity was evaluated. Structure, intermolecular interaction, and the reaction between the components were assessed in hydrogel structure. The cytocompatibility of th

Improving Chronic Diabetic Wound Healing through an Injectable and Self-Healing Hydrogel with Platelet-Rich Plasma Release.

Diabetic skin ulcer is one of the severe complications of diabetes mellitus, which has a high incidence and may cause death or disability. Platelet-rich plasma (PRP) is widely used in the treatment of diabetic wounds due to the effect of growth factors (GFs) derived from it. However, the relatively short half-life of GFs limits their applications in clinics. In addition, the presence of a large amount of proteases in the diabetic wound microenvironment results in the degradation of GFs, which further impedes angiogenesis and diabetic wound healing. In our study, we fabricated a self-healing an

A tubular gelatin scaffold capable of the time-dependent controlled release of epidermal growth factor and mitomycin C.

A tubular gelatin scaffold for the time-dependent controlled release of epidermal growth factor (EGF) and mitomycin C (MMC) was fabricated. EGF was incorporated using silk fibroin carriers, and MMC was planted using polylactide (PLA) microspheres. The relationship between scaffold properties and crosslinking degrees was evaluated. As the crosslinking degree was increased from 23.7% to 65.3%, the mechanical properties of the scaffold obviously improved, and the compressive modulus increased to approximately 65kPa. The mass degradation of the scaffold was also controlled from 9 days to approxima

A chitosan-based multifunctional hydrogel patch improves cardiac health and function by repairing the post-myocardial infarction microenvironment.

Myocardial infarction (MI) induced ischemia and hypoxia trigger a complex pathological microenvironment characterized by oxidative stress, inflammation, and fibrosis. However, the existing surgical and pharmacological treatments, as well as some tissue engineering techniques, fail to adequately therapy these symptoms, ultimately leading to cardiac dysfunction and poor prognosis. In this study, a multifunctional hydrogel patch composed of chitosan (CS), intrinsically hierarchical silk fibroin (SF) fibers, and reactive oxygen species (ROS) scavenging tannic acid (TA) for cardiac implantation was

Double Enzyme Active Hydrogel Program Regulates the Microenvironment of Staphylococcus aureus-Infected Pressure Ulcers.

The treatment of infected pressure ulcers (IUPs) requires addressing diverse microenvironments. A pressing challenge is to effectively enhance the regenerative microenvironment at different stages of the healing process, tailoring interventions as needed. Here, a dual enzyme mimetic and bacterial responsive self-activating antimicrobial hydrogel designed to enhance IPUs healing is introduced. This hydrogel incorporates pH-responsive dual enzyme-active nanoplatforms (HNTs-Fe-Ag) encapsulated within a methacrylate-modified silk fibroin (SFMA) and dopamine methacrylamide (DMA) matrix. This compos

A tree-inspired liquid-managing scaffold with radial-axial continuity for wound exudate management.

Effective exudate management remains challenging in complex wounds because most dressings rely on local absorption or surface wetting, which readily fail under persistent, heterogeneous, and three-dimensional fluid input. Here, inspired by the radial-axial transport organization of trees, we developed an annealed bioinspired radial-axial convergent scaffold (aBRACS) from regenerated silk fibroin (RSF) via directional ice templating. Featuring a seamless convergent-ascending channel architecture, aBRACS couples radial liquid capture with axial drainage in a continuous three-dimensional network,

Bidirectional optoelectronic switching in protein by photoinduced phase transition.

Protein polymers conduct electricity due to the delocalization of π electrons along the conjugated main chains, but their photoconductivity often shows unidirectional tunability. We report a protein polymer with bidirectional optoelectronic conductivity because of reversible light intensity-dependent secondary structural transformation. The protein device shows a positive photocurrent under high light intensity (2.22 to 3.18 μW·μm-2 and the wavelength of 405-nanometer illumination) illumination and a negative photocurrent under low light intensity (0.32 to 1.91 μW·μm-2). Under low light intens

Bioelectronic hydrogel driven by perovskite solar cells promotes healing of drug-resistant bacteria-infected wounds.

Self-powered dressings based on triboelectric or piezoelectric systems generate alternating electric stimulation (ES) that induces M1-type macrophage polarization and exacerbates inflammation, limiting therapeutic efficacy. Here, we report a self-powered dressing integrating portable perovskite solar cells to deliver direct ES that activates anti-inflammatory M2-type macrophage polarization, thereby regulating immune responses and reshaping infected-wound microenvironments. Under optimized conditions (130 mV/mm, 30 min), solar cell-derived ES combined with antimicrobial peptide-loaded silk fib

Down-Regulating Scar Formation by Microneedles Directly via a Mechanical Communication Pathway.

Excessive extracellular matrix deposition drives fibroblasts into a state of high mechanical stress, exacerbating pathological fibrosis and hypertrophic scar formation, leading to tissue dysfunction. This study reports a minimally invasive and convenient approach to obtaining scarless tissue using a silk fibroin microneedle patch (SF MNs). We found that by tuning the MN size and density only, the biocompatible MNs significantly decreased the scar elevation index in the rabbit ear hypertrophic scar model and increased ultimate tensile strength close to regular skin. To advance our understanding

Promising Biomolecules.

The osteochondral defect (OD) comprises the articular cartilage and its subchondral bone. The treatment of these lesions remains as one of the most problematic clinical issues, since these defects include different tissues, requiring distinct healing approaches. Among the growing applications of regenerative medicine, clinical articular cartilage repair has been used for two decades, and it is an effective example of translational medicine; one of the most used cell-based repair strategies includes implantation of autologous cells in degradable scaffolds such as alginate, agarose, collagen, ch

Antigen-Containing Silk Nanoparticles: A New Potential as Oral Vaccine Carriers.

PURPOSE: Oral vaccines are safer, easier to administer, and more cost-effective than injectable vaccines are. They induce systemic and mucosal immunity, thereby providing broad protection. However, developing an orally administered vaccine that can traverse the stomach and safely reach the intestinal tract to induce antigen-specific immune responses is challenging. Recently, many effective nanoparticle (NP) drug delivery systems have been developed. Various polymeric materials, including synthetic biodegradable and natural polymers, have been used as drug delivery matrices. Silk fibroin, a nat

SilMA-stabilized micro-wavy PLCL vascular grafts for native-like nonlinear mechanics and vascular remodeling.

Cardiovascular diseases remain a major cause of mortality, yet small-diameter vascular grafts (SDVGs) are still limited by thrombosis and pathological luminal remodeling caused in part by mechanical mismatch. Here, we developed electrospun poly(L-lactide-co-ε-caprolactone) (PLCL) grafts with a mechanically programmed micro-wavy architecture and artery-like nonlinear elasticity. A conformal methacrylated silk fibroin (SilMA) layer was photopolymerized onto the PLCL fibers to stabilize the programmed structure and improve the blood-contacting interface. The resulting graft preserved a porous arc

Potentiating Chemo-Immunotherapy via a Programmable Nanocapsule-Hydrogel Platform for Sequential Tumor Microenvironment Remodeling.

Durable responses to cancer immunotherapy require both robust antitumor immunity and sustained immune pressure within the immunosuppressive tumor microenvironment (TME). Chemotherapeutics can induce transient immune priming and modulate TME features but are limited by unpredictable immune response durations and systemic toxicity when combined with immunotherapy. To overcome these challenges, we develop a programmable delivery platform integrating chemotherapeutics and immune checkpoint blockade within a silk fibroin hydrogel containing pH-responsive nanocapsules for controlled temporal release

Silk-based materials: Emerging for tendon/tendon-bone regeneration.

The tendon and tendon-bone interface injuries have long been an issue for clinical practice. Autologous or allogeneic transplantation has been found to be a good method for repairing injuries; however, many factors limit the application of autologous and allogeneic transplants, which urges researchers to find a new type of strategy for regeneration. Silk fibroin (SF) scaffolds serve as promising exogenous biomaterials for alternative tendon and bone regeneration strategies. As its excellent biocompatibility, satisfactory mechanical properties, controllable biodegradability, and structural tuna

Biomimetic cellulose polysaccharide hydrogel with in situ "epidermal armor" for hemostasis, antimicrobial protection, and wound healing.

The skin serves as a critical barrier against infection and external damage. Effective wound management demands prompt hemostasis and infection control. Inspired by the hierarchical "epidermis-dermis" architecture of human skin, this work introduces a biomimetic bilayer hydrogel dressing. The hydrogel (named Fe@S/C) combines a dermis-mimetic layer of a double-crosslinked network of silk fibroin (Silk-GMA) and oxidized carboxymethyl cellulose (OXCMC-MA) with an in situ-formed epidermis-mimetic "Armor" antimicrobial protective layer via Fe3+ coordination. The hydrogel exhibits favorable mechanic

Phycocyanin-based multifunctional hydrogel with self-healing, hemostatic, antioxidative, and antibacterial activity for wound healing.

Hydrogels have gained significant attentions in the field of wound dressing since their resemblance to the extracellular matrix and excellent biocompatibility. Oxidative damage and bacterial infections resulting from an accumulation of reactive oxygen species (ROS) present significant challenges for chronic non-healing wounds. To address the issues of excessive ROS and bacterial infections during the wound healing process, we utilized C-phycocyanin, gelatin and silk fibroin as matrices to fabricate multifunctional hydrogels (PRG hydrogel), with rhein introduced as an antibacterial agent and te
