From Fusarium solani Keratitis to Full Visual Acuity: A Case of Infection Requiring Multiple Corneal Transplants.

Background and Clinical Significance: The Fusarium solani species complex is a group of filamentous fungi capable of causing severe ocular infections. It is a frequent cause of mycotic keratitis, which is associated with a poor prognosis due to aggressive biological behavior and resistance to most antifungal agents. Contact lens wear is a well-known risk factor; Case Presentation: A 48-year-old female contact lens wearer presented with a persistent corneal ulcer and hypopyon in the right eye. Initial visual acuity was hand movements. Hyphae of the Fusarium solani species complex were detected …

Wheat-associated Antagonistic Bacteria Exerts Biocontrol Activity Against Fungal Growth and Deoxynivalenol Production in Fusarium Graminearum.

Fusarium head blight (FHB), primarily caused by Fusarium graminearum, leads to severe contamination of wheat grains with the mycotoxin deoxynivalenol (DON), posing a significant threat to grain safety and quality. While chemical control remains common, its efficacy is increasingly challenged by fungicide resistance and potential stimulation of DON production. Biological control represents a promising alternative, yet the identification of effective antagonistic agents and elucidation of their mechanisms are still needed. This study evaluated the biocontrol potential of the endophytic bacterium…

Antifungal Activity of Electrochemically Etched Nanotextured Stainless Steel against Candida albicans and Fusarium oxysporum Fungal Cells.

Fungal adhesion to stainless steel, an alloy commonly used in the food and beverage sectors, public and healthcare settings, and numerous medical devices, can give rise to serious infections, ultimately leading to morbidity, mortality, and significant healthcare expenses. In this study, we demonstrate that nanotextured stainless steel (nSS) fabricated using an electrochemical technique is an antibiotic-free biocidal surface against Candida albicans and Fusarium oxysporum fungal cells with 98% and 97% reduction, respectively. The nanoprotrusion features on nSS can have both physical contact wit…

Mating type genes and genetic markers to decipher intraspecific variability among Fusarium udum isolates from pigeonpea.

To ascertain the variability in Fusarium udum (Fu) isolates associated with pigeonpea wilt is a difficult task, if based solely on morphological and cultural characters. In this respect, the robustness of five different genetic marker viz., random amplified polymorphic DNA (RAPD), enterobacterial repetitive intergenic consensus (ERIC), BOX elements, mating type locus, and microsatellite markers were employed to decipher intra-specific variability in Fu isolates. All techniques yielded intra-specific polymorphism, but different levels of discrimination were obtained. RAPD-PCR was more discrimin…

Ability of nonpathogenic Fusarium oxysporum strain Fo47 to induce resistance against Pythium ultimum infection in cucumber.

The influence exerted by nonpathogenic Fusarium oxysporum strain Fo47 in triggering cucumber protection against infection by Pythium ultimum was investigated ultrastructurally. Macroscopic and microscopic observations of the pathogen colony in dual cultures revealed that reduction of Pythium growth was associated with marked disorders, including generalized disorganization of the host cytoplasm, retraction of the plasmalemma, and complete loss of the protoplasm. Cytochemical labeling of cellulose with an exoglucanase-gold complex showed that the cellulose component of the host cell walls was s…

Transformation patterns of masked fusarium mycotoxins during barley malting: Mechanistic evidence and multimodal detection in industrial products.

Malt, a key food industry ingredient, faces safety risks from Fusarium mycotoxins and masked derivatives. While malt's agricultural role is clear, biotransformation pathways of masked mycotoxins during barley malting remain poorly understood. This study assesses mycotoxin contamination in commercial malt and links fungal communities to mycotoxin metabolism. Deoxynivalenol dominated, detected in 55% beer and 70% malt samples, with low adult exposure risk. Using ultra performance liquid chromatography-tandem mass spectrometry and internal transcribed spacer sequencing, natural malting showed bip…

Is within-field spatial variation in wheat Fusarium crown rot associated with rhizosphere microbiome and soil nutrients.

Fusarium crown rot (FCR) of wheat, caused predominantly by Fusarium pseudograminearum, frequently exhibits marked spatial heterogeneity within individual fields despite uniform cultivars and agronomic management. However, the mechanisms underlying this within-field variation remain poorly understood. In this study, we investigated six commercial wheat fields in Henan Province, China, each containing distinct highly diseased (HD) and lightly diseased (LD) zones. Soil physicochemical properties and rhizosphere microbial communities (characterised via 16S rRNA and ITS amplicon sequencing) were co…

Computational design of oleuropein as a potential antifungal agent and enzyme engineering targeting multidrug-resistant fusarium species.

Fusarium species, including Fusarium oxysporum and Fusarium solani, pose significant threats to global agriculture due to their multidrug resistance (MDR), which reduces the efficacy of conventional fungicides. This study investigates the potential of Oleuropein, a natural phenolic compound, as a candidate antifungal agent targeting Fusarium-associated proteins. Molecular docking analysis indicated that oleuropein exhibits favorable binding affinity (- 6.6 kcal/mol) toward key fungal targets, supported by hydrogen bonding and hydrophobic interactions. The stability of the protein-ligand comple…

New findings on ascus structure reveal forcible spore discharge mechanism in the fungal wheat pathogen Fusarium graminearum.

Fungi in the phylum Ascomycota disperse their sexual spores primarily by forcible discharge from the spore sac, the ascus, in which they develop. These fungi cause some of the most devastating diseases of agricultural crops worldwide. The mechanism of spore discharge is not well understood, despite its primary role in disease spread. Using confocal microscopy, we have elucidated the ascus structure and development as related to function for spore dispersal in Fusarium graminearum, a pathogen of small grains and corn. To determine the mechanism of spore discharge in this species, we examined as…
