Multiple origins for phenylketonuria in Europe.

Phenylketonuria (PKU), a disorder of amino acid metabolism prevalent among Caucasians and other ethnic groups, is caused primarily by a deficiency of the hepatic enzyme phenylalanine hydroxylase (PAH). PKU is a highly heterogeneous disorder, with more than 60 molecular lesions identified in the PAH gene. The haplotype associations, relative frequencies, and distributions of five prevalent PAH mutations (R158Q, R261Q, IVS10nt546, R408W, and IVS12n1) were established in a comprehensive European sample population and subsequently were examined to determine the potential roles of several genetic m

Histocompatibility lymphocytic antigen (HLA) typing in patients with acute exacerbation of Hashimoto's thyroiditis.

Histocompatibility lymphocytic antigen (HLA) typing was performed in 6 patients with acute exacerbation of Hashimoto's thyroiditis whose diagnoses were established on the basis of typical histological findings, and was compared with those of 12 with subacute thyroiditis, 33 with general Hashimoto's thyroiditis and also with a control group. There was a high incidence of BW35 in patients with subacute thyroiditis, although it was only seen in 1 of 6 patients with acute exacerbation. The difference was statistically significant (p less than 0.01). Four of 6 patients with acute exacerbation had D

Human leukocyte antigen (HLA) in Korean patients with autoimmune thyroid diseases.

In previous studies, there has been evidence of different allelic associations with a particular disease among various ethnic groups. The present study was done to investigate the associations between HLA and autoimmune thyroid diseases in the Korean. We found no association between Graves’ disease and HLA-B8 or -B35. However, increased frequencies of HLA-A11 and -DRw8, and the decreased frequencies of HLA-A10 and B12 were found in patients with Graves’ disease. In the cases of Hashimoto’s disease, the frequencies of HLA-A2 and -DRw8 were found to be significantly increased and the frequency o

Family and population studies of HLA and coeliac disease in the West of Ireland.

Tissue typing (HLA-A, -B and -DR) was carried out on 92 patients with coeliac disease (CD) and on a further 71 first degree relatives of 13 of these patients. Results of antigen frequencies in unrelated patients and segregation analysis in families shows that HLA-DR3 but not HLA-DR7 is positively associated with CD. Haplotype and sib-pair data from these coeliac families suggest that with all of them, genetic susceptibility to CD lies in the HLA region of the sixth chromosome. In families where the coeliac proband has HLA-DR3, the relative risk to siblings who also have this antigen is increas

Individual differences with respect to the sneezing reflex: an inherited physiological trait in man?

Individual differences with respect to the sneezing reflex are described. Visual exposure to strong light may induce a sneezing reaction in about 20% of the individuals in the Swedish population. Preliminary data indicate that the ' sneezer trait' may be inherited in an autosomal dominant fashion. The eventual selective advantage of ST is discussed.

Two locus models for gluten sensitive enteropathy: population genetic considerations.

Familial occurrence of coeliac disease (gluten-sensitive enteropathy, GSE) is well known, but the mode of inheritance remains unclear. Pena et al proposed that the genetic basis for GSE was due to disease-predisposing alleles at two loci: DRw3 at the HLA-D locus and a GSE-associated B-cell alloantigen at another, unlinked locus. They concluded that clinical disease required one DRw3 (dominant inheritance) and two B-cell alloantigen alleles (recessive inheritance), but the observed gene frequencies were not consistent with the observed disease prevalence. Here we examine the gene frequencies al

HLA-A,B,C and DR antigens in psoriasis.

51 psoriasis vulgaris patients and 93 controls were tested for HLA-A,B,C and DR antigenic frequencies. Significant increases of B17, Cw6 and DR7 were documented in the patient group, as well as a decreased frequency of DR1. The significance of these findings is discussed. DR7 occurred more often together with Cw6 in psoriasis patients than in controls, which might suggest that there are at least two interacting HLA linked genes which increase the disease susceptibility and possibly one DR1 linked gene associated with resistance to the disease.

HLA-DRw4 in pemphigus vulgaris patients in Israel.

Pemphigus vulgaris (PV) is relatively common in Jews. Three HLA antigens were significantly more frequent in 39 Israeli Jewish PV patients than in controls A26 - 59% vs 20%; Bw38 - 61% vs 20%; and DRw4 - 90% vs 38%. The joint occurrence of A26-Bw38-DRw4 was observed in 46% of PV patients and in 10% of controls. Similar results were recently reported for Jews in the Los Angeles area. Yet, when our patient sample was grouped into Ashkenazi and non-Ashkenazi Jews, it was evident that each of the three antigens had a higher frequency both in Ashkenazi patients and controls as compared to non-Ashke

DMRT3 gene variations in horse breeds selected for gaited movement - established research and novel findings.

BACKGROUND AND OBJECTIVES: The DMRT3 gene, often referred to as the "gait keeper," plays a key role in controlling alternative gaits in horses, such as tölt and pace. This study aimed to determine the frequency of known and to screen for potential novel polymorphisms within the second exon of the DMRT3 gene. DATA COLLECTION: A total of 244 blood or hair samples were collected from representative individuals of the six horse breeds: gaited (Icelandic Horse, French Trotter), non-gaited (Arabian Horse, Malopolski Horse), and pony breeds (Welsh Pony, Shetland Pony). RESEARCH METHODS: The second

Involvement of Gaucher Disease Mutations in Parkinson Disease.

Gaucher disease is an autosomal recessive lysosomal storage disorder, caused by mutations in the GBA gene. The frequency of Gaucher disease patients and heterozygote carriers that developed Parkinson disease has been found to be above that of the control population. This fact suggests that mutations in the GBA gene can be involved in Parkison's etiology. Analysis of large cohorts of patients with Parkinson disease has shown that there are significantly more cases bearing GBA mutations than those found among healthy individuals. Functional studies have proven an interaction between α-synuclein

Extensive genetic diversity among populations of the malaria mosquito Anopheles moucheti revealed by population genomics.

Malaria vectors are exposed to intense selective pressures due to large-scale intervention programs that are underway in most African countries. One of the current priorities is therefore to clearly assess the adaptive potential of Anopheline populations, which is critical to understand and anticipate the response mosquitoes can elicit against such adaptive challenges. The development of genomic resources that will empower robust examinations of evolutionary changes in all vectors including currently understudied species is an inevitable step toward this goal. Here we constructed double-digest

Isoenzyme differentiation of Aedes aegypti populations in French Guiana.

Population genetics of peri-domestic Aedes aegypti (Diptera: Culicidae), vector of dengue and yellow fever, were investigated by gel electrophoresis of 10 enzyme loci in 14 samples of mosquito larvae collected in 1997-1998 from localities separated by distances of 3-275 km in French Guiana. Genetic differentiation between geographical populations was generally high (mean FST = +0.111, P < 10(-5)) even among seven sites <30 km apart (FST = +0.137, P < 0.05), but not positively correlated with distance. Thus, Ae. aegypti comprises a mosaic of genetically differentiated populations in French Guia

Dose-effect relationship of dicentric and ring chromosomes in lymphocytes of individuals living in the high background radiation areas in China.

Chromosomes of 39 healthy family members (3 generations from 13 families) living both in the high-level background radiation areas (HBRA) and the control areas (CA) were studied. Cumulative dose from birth to the time of blood sampling was estimated by calculating measured exposure rate in each individual. The cumulative doses ranged 30.9-358.9 and 6.0-59.2 mGy for HBRA and CA, respectively. Peripheral lymphocyte chromosome preparations were made according to our improved method. Dicentric and ring chromosomes (Dic + Rc) were scored in average 2,527 cells per individual in HBRA and 2,694 cells

Association of apolipoprotein E allele epsilon 4 with late-onset familial and sporadic Alzheimer's disease.

Apolipoprotein E, type epsilon 4 allele (APOE epsilon 4), is associated with late-onset familial Alzheimer's disease (AD). There is high avidity and specific binding of amyloid beta-peptide with the protein ApoE. To test the hypothesis that late-onset familial AD may represent the clustering of sporadic AD in families large enough to be studied, we extended the analyses of APOE alleles to several series of sporadic AD patients. APOE epsilon 4 is significantly associated with a series of probable sporadic AD patients (0.36 +/- 0.042, AD, versus 0.16 +/- 0.027, controls [allele frequency estimat

Detection of major gene for Gilles de la Tourette syndrome.

The families of 250 consecutive, unselected patients with Tourette syndrome (TS) were analyzed. If the parents had either motor or vocal tic … 2 per 1,000 in the general population. A pure recessive major gene effect was also rejected. With a gene frequency of approximately .5%, the penetrance was estimated to be about 94% in abnormal Ts/Ts homozygotes, 50% in Ts/ts heterozygotes, and less than 0.3% in normal ts/ts homozygotes. More than two of every three cases are heterozygotes, and nearly all other …

Genetic study of Welsh gypsies.

A South Wales gypsy kindred of Romany origin had a high incidence of phenylketonuria along with other recessively inherited disorders. There was a high degree of consanguinity (F = 0-017) with an excess of non-specific mental subnormality among known consanguineous matings. Phenylketonuria and a number of other recessively inherited disorders have been recorded from other Romany gypsy populations, but it is uncertain whether this results from a generally high gene frequency for the disorders or from consanguinity and other more local factors.
