Alterations in T-Cell Transcription Factors and Cytokine Gene Expression in Late-Onset Alzheimer's Disease.

BACKGROUND: Late-onset Alzheimer's disease (LOAD) is associated with many environmental and genetic factors. The effect of systemic inflammation on the pathogenesis of neurodegenerative diseases such as AD has been strongly suggested. T helper cells (Th) are one of the important components of the immune system and can easily infiltrate the brain in pathological conditions. The development of each Th-subset depends on the production of unique cytokines and their main regulator. OBJECTIVE: This study aimed to compare the mRNA levels of Th-related genes derived from peripheral blood mononuclear

A case of paramyotonia congenita without periodic paralysis: electrophysiological and molecular genetic studies.

Paramyotonia congenita (PC), first described in 1886 by Eulenberg, is characterized by cold and exercise-induced muscle stiffness and intermittent flaccid paresis not necessarily related to cold or myotonia. Several authors segregated a pure form of PC, which has no periodic paralysis, even after cold exposure. The existence of this phenotype has been debated in the literature. We describe electrophysiological and molecular genetic features of a patient with PC who had no history of periodic paralysis. Immersion in cold water or potassium load could not induce clinical paralysis. However, repe

Sodium balance and peripheral resistance in arterial hypertension.

In this brief review, some aspects of the relation between sodium balance and peripheral resistance in four types of arterial hypertension are discussed. Renal hypertension, hypertension caused by reduced kidney mass and sodium load, mineralocorticoid hypertension, and spontaneous or "genetic" hypertension in rats are considered. In all these forms of hypertension, the increase in peripheral resistance is preceded by changes in body sodium and fluids. However, the precise role of these changes in the development of the subsequent rise of peripheral resistance is not yet clear. Changes in ion t

Sex-dependent effect of APOE on Alzheimer's disease and other age-related neurodegenerative disorders.

The importance of apolipoprotein E (APOE) in late-onset Alzheimer's disease (LOAD) has been firmly established, but the mechanisms through which it exerts its pathogenic effects remain elusive. In addition, the sex-dependent effects of APOE on LOAD risk and endophenotypes have yet to be explained. In this Review, we revisit the different aspects of APOE involvement in neurodegeneration and neurological diseases, with particular attention to sex differences in the contribution of APOE to LOAD susceptibility. We discuss the role of APOE in a broader range of age-related neurodegenerative disease

Host genetic determinants of resistance and susceptibility to bovine leukemia virus infection: A mini-review.

Bovine leukemia virus (BLV), a member of the Deltaretrovirus genus, causes enzootic bovine leukosis, leading to clinical outcomes that range from asymptomatic infection to malignant lymphoma. Host genetic factors significantly influence BLV susceptibility, proviral load (PVL), immune response, and disease progression. This mini-review synthesizes evidence on genetic polymorphisms in immune-related genes such as BoLA-DRB3, Tumor necrosis factor (TNF), and immunoglobulin loci, and examines novel findings from genome-wide association studies (GWAS). Beyond classical immune genes, recent GWAS have

Evaluating the Impact of Functional Genetic Variation on HIV-1 Control.

BACKGROUND: Previous genetic association studies of human immunodeficiency virus-1 (HIV-1) progression have focused on common human genetic variation ascertained through genome-wide genotyping. METHODS: We sought to systematically assess the full spectrum of functional variation in protein coding gene regions on HIV-1 progression through exome sequencing of 1327 individuals. Genetic variants were tested individually and in aggregate across genes and gene sets for an influence on HIV-1 viral load. RESULTS: Multiple single variants within the major histocompatibility complex (MHC) region were

Genetic control of asexual development in aspergillus fumigatus.

Aspergillus fumigatus is one of the most common fungi found in the environment. It is an opportunistic human pathogen causing invasive pulmonary aspergillosis with a high mortality rate in immunocompromised patients. Conidia, the asexual spores, serve as the main dispersal and infection agent allowing entrance of the fungus into the host through the respiratory tract. Therefore, understanding the asexual developmental process that gives rise to the conidia is of great interest to the scientific community and is currently the focus of an immense load of research being conducted. We have been st

Alzheimer's disease and intelligence.

A significant body of evidence has accumulated suggesting that individual variation in intellectual ability, whether assessed directly by intelligence tests or indirectly through proxy measures, is related to risk of developing Alzheimer's disease (AD) in later life. Important questions remain unanswered, however, such as the specificity of risk for AD vs. other forms of dementia, and the specific links between premorbid intelligence and development of the neuropathology characteristic of AD. Lower premorbid intelligence has also emerged as a risk factor for greater mortality across myriad hea

Revisiting hemochromatosis: genetic vs. phenotypic manifestations.

Iron overload disorders represent an important class of human diseases. Of the primary iron overload conditions, by far the most common and best studied is HFE-related hemochromatosis, which results from homozygosity for a mutation leading to the C282Y substitution in the HFE protein. This disease is characterized by reduced expression of the iron-regulatory hormone hepcidin, leading to increased dietary iron absorption and iron deposition in multiple tissues including the liver, pancreas, joints, heart and pituitary. The phenotype of HFE-related hemochromatosis is quite variable, with some in

Breeding for thermotolerance in dairy cattle: Production versus fertility traits.

The decline in production of milk and its components has been extensively studied as an indicator of heat tolerance for genetic evaluations. However, the antagonistic relationship between high production and functionality raises questions about the suitability of using productive traits as indicators of heat tolerance. This study aimed to estimate changes in the relationship between production and fertility under thermoneutral (TN) and heat stress (HS) conditions, to define breeding strategies that enhance adaptation to high heat loads while maintaining both productivity and functionality. The

Multiple Immune and Genetic Mechanisms Contribute to Cmv5s-Driven Susceptibility and Tissue Damage during Acute Murine Cytomegalovirus Infection.

The MHC class I molecule H-2Dk conveys resistance to acute murine CMV infection in both C57L (H-2Dk transgenic) and MA/My mice. M.H2k/b mice are on an MA/My background aside from a C57L-derived region spanning the MHC (Cmv5s), which diminishes this resistance and causes significant spleen histopathology. To hone in on the effector elements within the Cmv5s interval, we generated several Cmv5-recombinant congenic mouse strains and screened them in vivo, allowing us to narrow the phenotype-associated interval >6-fold and segment the genetic mechanism to at least two independent loci within the M

Genetic predisposition to major depressive disorder differentially impacts alcohol consumption and high-risk drinking situations in men and women with alcohol use disorder.

Lifetime history of major depressive disorder (MDD) has a sex-specific association with pretreatment alcohol consumption in patients with alcohol dependence. Here, we investigated the association of genetic load for MDD estimated using a polygenic risk score (PRS) with pretreatment alcohol consumption assessed with Timeline Follow Back in a sample of 287 men and 156 women meeting DSM-IV-TR criteria for alcohol dependence. Preferred drinking situations were assessed using the Inventory of Drug Taking Situations (IDTS). Linear models were used to test for association of normalized alcohol consum…

Targeted gene sequencing of Lynch syndrome-related and sporadic endometrial carcinomas.

About one-third of endometrial carcinomas (ECs), mainly of endometrioid histology, harbor the mismatch repair (MMR) defects and microsatellite instability (MSI). Among these, ECs arising in women with Lynch syndrome (LS) account for a large proportion. To date, no somatic genetic analyses have been published comparing LS-ECs with sporadic ECs. In this work, we examined the mutational profiles of a well-characterized series of sporadic and LS-related ECs, performing exonic targeted sequencing of 16 genes mainly involved in MSI ECs. Next-generation sequencing analysis was performed in 35 ECs on

Effect of genetic modifications by selection for immunological tolerance on fungus infection in mice.

Two strains of mice genetically selected for extreme phenotypes of immunological tolerance to ovalbumin, susceptible (TS) and resistant (TR), were experimentally infected with Sporothrix schenckii. The objective was to observe whether the genetic modifications produced by the selection might be associated with interstrain differences in adaptive immune and innate responses to infection. Therefore, we evaluated the LD(50), CFU, phagocytic index, fungicidal activity, pro-inflammatory cytokines, specific antibody titres, and the delayed-type hypersensitivity reactivity. TR mice were tenfold more

Maintain, not obtain: rethinking HIV care in sub-Saharan Africa in the era of dolutegravir.

Over the past four decades, the HIV epidemic in sub-Saharan Africa has shifted from an acute, high-mortality phase to an era of widespread antiretroviral therapy (ART), driven by global scale up and the introduction of potent regimens such as dolutegravir-based combinations. While viral suppression is now increasingly achievable, the central challenge has evolved toward maintaining durable virological control across the life course. We argue that program success can no longer be defined by the mere attainment of suppression, but by the sustained maintenance of undetectable viral load and prese

The Genetic Variability of UCP4 Affects the Individual Susceptibility to Late-Onset Alzheimer's Disease and Modifies the Disease's Risk in APOE-ɛ4 Carriers.

Uncoupling proteins (UCPs) are a group of five mitochondrial inner membrane transporters with a tissue specific expression that uncouple biofuel oxidation from ATP synthesis and function as regulators of energy homeostasis and antioxidants. Previous data suggested that neuronal UCPs (UCP2, UCP4, and UCP5) can directly influence synaptic plasticity, neurotransmission, and neurodegenerative processes, and have a crucial role in the function and protection of the central nervous system. In fact, it has been observed that the expression of neuronal UCPs significantly decreases in Alzheimer's disea

Contribution of polymorphic variation of inositol hexakisphosphate kinase 3 (IP6K3) gene promoter to the susceptibility to late onset Alzheimer's disease.

Maintenance of electric potential and synaptic transmission are energetically demanding tasks that neuronal metabolism must continually satisfy. Inability to fulfil these energy requirements leads to the development of neurodegenerative disorders, including Alzheimer's disease. A prominent feature of Alzheimer's disease is in fact neuronal glucose hypometabolism. Thus understanding the fine control of energetic metabolism might help to understand neurodegenerative disorders. Recent research has indicated that a novel class of signalling molecules, the inositol pyrophosphates, act as energy sen

Early-Onset Alzheimer Disease and Candidate Risk Genes Involved in Endolysosomal Transport.

IMPORTANCE: Mutations in APP, PSEN1, and PSEN2 lead to early-onset Alzheimer disease (EOAD) but account for only approximately 11% of EOAD overall, leaving most of the genetic risk for the most severe form of Alzheimer disease unexplained. This extreme phenotype likely harbors highly penetrant risk variants, making it primed for discovery of novel risk genes and pathways for AD. OBJECTIVE: To search for rare variants contributing to the risk for EOAD. DESIGN, SETTING, AND PARTICIPANTS: In this case-control study, whole-exome sequencing (WES) was performed in 51 non-Hispanic white (NHW) patie
