Genome-wide association studies of missing metabolite measures from two population-based studies.

BACKGROUND: Metabolomic analyses are increasingly applied in both etiological and predictive research, but frequently report missing values, which are then either imputed or removed from the analyses, and not examined as true missingness due to altered metabolism. We hypothesized that interindividual genetic variation may account for part of this missingness. RESULTS: We perform a logistic GWAS of metabolite missingness from an untargeted mass spectrometry-based platform in the Netherlands Epidemiology of Obesity Study (N = 594) and the Rhineland Study (N = 4,165). We consider metabolites mis

Association of hypothyroidism and respiratory diseases: A cross-sectional study among Europeans using Mendelian randomization.

To explore the causal relationship between hypothyroidism and various respiratory diseases through a 2-sample Mendelian randomization (MR) a … ept term of the MR-Egger regression had a P-value > .05, which indicated the absence of significant genetic pleiotropy. The results of heterogeneity tests with P-values > .05 suggested no heterogeneity among single nucleotide polymorphisms. MR analysis indicated that hypothyroidism elevated the risk of asthma.

Association between pelvic inflammatory disease and ovarian cancer: A bidirectional Mendelian randomization study.

Some observational studies have suggested an association between pelvic inflammatory disease (PID) and ovarian cancer (OC), but the causality between PID and OC remains unclear. To investigate the causal association between PID and 7 OCs, we performed two-sample bidirectional Mendelian randomization (MR) analyses using genetic variants associated with PID. Pooled data on OC were obtained from the Ovarian Cancer Association Consortium. Effect estimates were obtained by inverse-variance weighting, the weighted median method, MR-Egger regression, and MR pleiotropy residual sum and outlier (MR-PRE

Dissecting cross-population polygenic heterogeneity across respiratory and cardiometabolic diseases.

Biological mechanisms underlying multimorbidity remain elusive. To dissect the polygenic heterogeneity of multimorbidity in twelve complex traits across populations, we leveraged biobank resources of genome-wide association studies (GWAS) for 232,987 East Asian individuals (the 1st and 2nd cohorts of BioBank Japan) and 751,051 European individuals (UK Biobank and FinnGen). Cross-trait analyses of respiratory and cardiometabolic diseases, rheumatoid arthritis, and smoking identified negative genetic correlations between respiratory and cardiometabolic diseases in East Asian individuals, opposit

The Impact of Physical Activities on Men's Mental Health: A Focus on Bipolar Disorder Prevention.

Bipolar disorder (BD) is a prevalent psychiatric condition characterized by extreme mood fluctuations between manic and depressive episodes, significantly affecting social and occupational functioning. The etiology of BD is multifactorial, involving genetic, environmental, and lifestyle factors. While previous research has focused on the genetic and environmental contributors to BD, the role of physical activity as a modifiable lifestyle factor remains underexplored. This study investigates the causal relationship between different types of physical activity, particularly heavy do-it-yourself

Causal relationship between osteoporosis, bone mineral density, and osteonecrosis: a bidirectional two-sample Mendelian randomization study.

BACKGROUND: Osteonecrosis (ON) is a debilitating orthopedic condition characterized by bone cell death due to impaired blood supply, leading to structural changes and disability. Osteoporosis (OP), a systemic skeletal disease, results in reduced bone density and quality, making bones fragile and prone to fractures. Although distinct, OP and ON share several risk factors such as corticosteroid use and smoking. This study aims to investigate the causal relationships between OP, bone mineral density (BMD), and ON using a bidirectional two-sample Mendelian randomization (MR) approach. METHODS: Th

Rheumatoid arthritis increases the risk of malignant neoplasm of bone and articular cartilage: a two-sample bidirectional mendelian randomization study.

OBJECTIVE: Prior research has revealed a heightened prevalence of neoplasms in individuals diagnosed with rheumatoid arthritis (RA). The primary objective of this study is to delve into the causal association between RA and two distinct types of neoplasms: benign neoplasm of bone and articular cartilage (BNBAC) and malignant neoplasm of bone and articular cartilage (MNBAC). METHODS: We employed summary data from genome-wide association analyses (GWAS) to investigate the causal relationship between RA and two neoplasms, BNBAC and MNBAC, using a two-sample bidirectional Mendelian randomization

The genetic landscape of age-related hearing loss.

Age-related hearing loss (ARHL) is a prevalent concern in the elderly population. Recent genome-wide and phenome-wide association studies (GWASs and PheWASs) have delved into the identification of causative variants and the understanding of pleiotropy, highlighting the polygenic intricacies of this complex condition. While recent large-scale GWASs have pinpointed significant SNPs and risk variants associated with ARHL, the detailed mechanisms, encompassing both genetic and epigenetic modifications, remain to be fully elucidated. This review presents the latest advances in association studies,

COVID-19 and the Risks of Migraine and Headache: A Mendelian Randomization Study.

Several observational studies suggested that migraine headache attacks were associated with coronavirus disease 2019 (COVID-19). We investigated genetic causal links between COVID-19 phenotypes and the development of headache and migraine, including migraine with aura (MA) and migraine without aura (MO). We conducted a two-sample Mendelian randomization (MR) analysis to estimate the genetic association in European populations. The inverse-variance weighted (IVW) method was used as the main approach in the MR analyses, together with weighted median and MR-Egger methods. We also performed a seri

Elucidating mechanisms of genetic cross-disease associations at the PROCR vascular disease locus.

Many individual genetic risk loci have been associated with multiple common human diseases. However, the molecular basis of this pleiotropy often remains unclear. We present an integrative approach to reveal the molecular mechanism underlying the PROCR locus, associated with lower coronary artery disease (CAD) risk but higher venous thromboembolism (VTE) risk. We identify PROCR-p.Ser219Gly as the likely causal variant at the locus and protein C as a causal factor. Using genetic analyses, human recall-by-genotype and in vitro experimentation, we demonstrate that PROCR-219Gly increases plasma le

Investigating the genetic architecture of eye colour in a Canadian cohort.

Eye color is highly variable in populations with European ancestry, ranging from low to high quantities of melanin in the iris. Polymorphisms in the HERC2/OCA2 locus have the largest effect on eye color in these populations, although other genomic regions also influence eye color. We performed genome-wide association studies of eye color in a Canadian cohort of European ancestry (N = 5,641) and investigated candidate causal variants. We uncovered several candidate causal signals in the HERC2/OCA2 region, whereas other loci likely harbor a single causal signal. We observed colocalization of eye

Executive Function as an Underlying Mechanism of Alcohol Use, Aggression, and ADHD.

BACKGROUND: Executive functioning (EF) has been proposed as a transdiagnostic risk factor for externalizing disorders and behavior more broadly, including attention-deficit/hyperactivity disorder (ADHD), aggression, and alcohol use. Previous research has demonstrated both phenotypic and genetic overlap among these behaviors, but has yet to examine EF as a common causal mechanism. The current study examined reciprocal causal associations between EF and several externalizing behaviors using a Mendelian randomization (MR) approach. METHODS: Two-sample MR was conducted to test causal associations
