[Genetically based diseases].

Genetics is one of the greatest scientific advances of the XX century, which begins with the rediscovery of Mendel's laws and culminates in the elaboration of the first "Черновик" of the complete sequence of the human genome. Genetics employs different research strategies, such as the study of twins and adoption, investigating the influence of genetic and environmental factors, and strategies for identifying specific genes (molecular genetics). Besides the significant degree of disability they generate, the social impact of hereditary diseases is enormous, due to their potentially recurrent chara

Does genetic testing remain relevant in advanced Parkinson's disease?

Genetic testing is traditionally emphasized in early-onset or familial Parkinson's disease (PD), leaving its role in advanced disease uncertain. This mini-review explores, from the perspectives of patients, their family members, and clinical researchers, whether genetic investigation remains meaningful once patients reach advanced PD. In large prospective clinical-genetic screening cohorts, roughly one in seven PD patients has genetic PD. Established genotype-phenotype correlations, spanning cognitive trajectory, levodopa responsiveness, and deep brain stimulation outcomes, continue to inform

New insights and perspectives on the genetics of obsessive-compulsive disorder.

Psychiatric genetic research has exploded in search of polygenic risk factors over the past decade, but because of the complexity and heterogeneity of mental illnesses, using the current understanding of the genome has not reached the conclusion of finding a cause for psychiatric disorders. Obsessive-compulsive disorder is a relatively common and often debilitating neuropsychiatric disorder that has not been the primary focus in psychiatric research. Clinicians and researchers who have dedicated to investigate the genetics of obsessive-compulsive disorder have detected a strong genetic involve…

Clinical and Genetic Aspects of Alopecia Areata: A Cutting Edge Review.

Alopecia areata (AA) is a chronic, non-scarring, immune-mediated skin disease that affects approximately 0.5-2% of the global population. The etiology of AA is complex and involves genetic and environmental factors, with significant advancements in genetic research occurring in recent years. In addition to well-known genes such as PTPN22, CTLA4, and IL2, which have been widely supported as being associated with AA, an increasing number of specific gene-related loci have been discovered through advances in genetic research. For instance, gene analysis of microRNAs can reveal the critical role o…

"It's Not Deterministic and It Will Never Be Deterministic": A Qualitative Study on Stakeholder Perspectives of Polygenic Risk Score Testing for Post-Traumatic Stress Disorder.

Post-traumatic stress disorder (PTSD) causes significant mental and physical distress, yet only a small subset of individuals exposed to trauma develop the disorder. Scientists and clinicians are still unable to predict who will get the disorder or how it will manifest. Genetic factors contribute to PTSD risk, but integrating genetic findings into clinical practice has been challenging. Military veterans are especially impacted by PTSD and have different treatment outcomes when compared to civilians. Thus, genetic findings related to PTSD may be particularly valuable to this population. This q

The Effect of Genetic Taste Status on Swallowing: A Literature Review.

PURPOSE: Swallowing and taste share innervation pathways and are crucial to nutritive intake. Individuals vary in their perception of taste due to factors such as genetics; however, it is unclear to what extent genetic taste status influences swallowing physiology and function. The purpose of this review article is to provide background on genetic taste status, review the evidence on the association between genetic taste status and swallowing, and discuss research and clinical implications. METHOD: A comprehensive literature review was conducted using search terms related to swallowing and ge

Genetic counseling and testing practices for late-onset neurodegenerative disease: a systematic review.

OBJECTIVE: To understand contemporary genetic counseling and testing practices for late-onset neurodegenerative diseases (LONDs), and identify whether practices address the internationally accepted goals of genetic counseling: interpretation, counseling, education, and support. METHODS: Four databases were systematically searched for articles published from 2009 to 2020. Peer-reviewed research articles in English that reported research and clinical genetic counseling and testing practices for LONDs were included. A narrative synthesis was conducted to describe different practices and map gene

The emerging role of the physician in genetic counselling and testing for heritable breast, ovarian and colon cancer.

As a genetic testing for susceptibility to breast, ovarian and colon cancer becomes more readily available, physicians are faced with an increasing demand for information about inherited cancer risk. Because advances in treatment have not kept pace with advances in predictive testing, the provision of genetic counselling and testing marks a departure from the traditional role of the physician. A systematic framework is needed within which the physician's emerging role in predictive testing for heritable cancer can be delineated. The development of such a framework will require collaboration am

Human fetal gene therapy: moral and ethical questions.

This two-part paper discusses moral and ethical questions raised by future trials of human fetal gene therapy. The first part examines broad moral issues to explore whether fetal gene therapy is a morally praiseworthy goal. Ought it be done at all? These issues include (i) how the concept of fetal gene therapy originally arose as a goal envisioned at the beginning of prenatal diagnosis, (ii) preimplantation genetic diagnosis as a better preconceptual alternative for parents at higher genetic risk, (iii) alternatives to genetic abortions, (iv) the social and economic priority of fetal gene ther

Shared genetic architecture of gastroesophageal reflux disease and age related phenotypes.

Increasing age is a risk factor of gastroesophageal reflux disease. This study aims to uncover the shared genetic architecture of gastroesophageal reflux disease (GERD) and age-related phenotypes. Based on publicly available GWAS statistics, this genome-wide pleiotropic association research was performed with multiple genetic approaches sequentially to explore the pleiotropic associations from single-nucleotide polymorphism (SNP) and gene levels, to reveal the underlying shared genetic etiology between GERD and age-related phenotypes. This study featured shared genetic mechanisms between GERD

The contribution of genetic and environmental factors to the duration of pregnancy.

This review describes how improvements in biometric-genetic studies of twin kinships, half-sibships, and cousinships have now demonstrated a sizeable fetal genetic and maternal genetic contribution to the spontaneous onset of labor. This is an important development because previous literature for the most part reports only an influence of the maternal genome. Current estimates of the percent of variation that is attributable to fetal genetic factors range from 11-35%; the range for the maternal genetic contribution is 13-20%. These same studies demonstrate an even larger influence of environme

Twin study method: Unlocking genetic and environmental interactions.

Twin studies offer a powerful approach for disentangling genetic and environmental influences on human traits. By comparing monozygotic twins with identical genetic makeup, researchers can more accurately identify the environmental contributions to phenotypic variation. Structural equation modeling provides a theoretical framework for estimating the relative contributions of additive genetic effects, shared environmental factors, and unique environmental influences. This model allows researchers to determine the most suitable parameters for explaining the observed data in twin populations. In

Increase in linkage information by stratification of pedigree data into gold-standard and standard diagnoses: application to the NIMH Alzheimer Disease Genetics Initiative Dataset.

Patients diagnosed with a standard clinical method (subject to misclassification error) are often combined with patients diagnosed with a gold-standard method (with zero or very small misclassification error) in family-based studies of complex disease. For example, non-autopsied patients (NAP) are often included along with autopsy-proven (AP) patients in family-based studies of complex diseases, such as Alzheimer's disease (AD). Theoretical and simulation studies suggest that certain misclassification errors can result in severe reduction of power in genetic linkage and association analyses an

Decoding HuH-7: a comprehensive genetic and molecular portrait of a widely used hepatocellular carcinoma model.

INTRODUCTION: Immortalized cell lines play a crucial role in biomedical research by enabling reproducible experiments and enhancing our understanding of complex diseases. HuH-7, originally derived from a human hepatocellular carcinoma, is particularly valuable for studying liver cancer dynamics, viral hepatitis, and drug metabolism. However, concerns about cell line misidentification and genetic drift in cell lines highlight the importance of rigorous authentication to maintain the reliability of research outcomes, despite their widespread use. METHODS: In this study, we present a detailed (c

Examination of Huntington's disease with atypical clinical features in a Bangladeshi family tree.

Atypical manifestation of Huntington's disease (HD) could inform ongoing research into HD genetic modifiers not present in the primarily European populations studied to date. This work demonstrates that expanding HD genetic testing into under-resourced healthcare settings can benefit both local communities and ongoing research into HD etiology and new therapies.
