Effects of chemotherapeutic agents on male germ cells and possible ameliorating impact of antioxidants.

Treatment of cancer in young adults is associated with several side effects, particularly in the reproductive system. Detrimental effects of chemotherapy on the germ cells depend on many factors including primary semen parameters, the way of drug administration, the kind and dose of chemotherapeutic regimens, and the phase of spermatogenesis during the time of drug administration. Lack of appropriate fertility preservation treatments particularly in the affected children necessitates the introduction of methods to amend the harmful effects of chemotherapeutic agents on male germ cells. Several…

SYMPK Is Required for Meiosis and Involved in Alternative Splicing in Male Germ Cells.

SYMPK is a scaffold protein that supports polyadenylation machinery assembly on nascent transcripts and is also involved in alternative splicing in some mammalian somatic cells. However, the role of SYMPK in germ cells remains unknown. Here, we report that SYMPK is highly expressed in male germ cells, and germ cell-specific knockout (cKO) of Sympk in mouse leads to male infertility. Sympk cKODdx4-cre mice showed reduced spermatogonia at P4 and almost no germ cells at P18. Sympk cKOStra8-Cre spermatocytes exhibit defects in homologous chromosome synapsis, DNA double-strand break (DSB) repair, a…

CFTR is required for the migration of primordial germ cells during zebrafish early embryogenesis.

Mutations in the cystic fibrosis transmembrane conductance regulator (CFTR) gene affect fertility in both sexes. However, the involvement of CFTR in regulating germ cell development remains largely unknown. Here, we used zebrafish model to investigate the role of CFTR in primordial germ cells (PGCs) development. We generated a cftr frameshift mutant zebrafish line using CRISPR/Cas9 technique and investigated the migration of PGCs during early embryo development. Our results showed that loss of Cftr impairs the migration of PGCs from dome stages onward. The migration of PGCs was also perturbed …

Normal fertility in male mice with deletion of β-catenin gene in germ cells.

As a dual function protein, β-catenin affects both cell adhesion and mediates canonical Wnt/β-catenin cell signaling. β-Catenin is prominently expressed in somatic Sertoli cells in the testis and postmeiotic germ cells, suggesting an additional role in spermatogenesis. It was reported previously that Cre/loxP-mediated conditional inactivation of the β-catenin gene (Ctnnb1) in male gonads using a protamine promoter-driven Cre transgene (Prm-cre) resulted in partial infertility, reduced sperm count, and abnormal spermatogenesis. In this report, we demonstrated that the conditional deletion of Ct…

Effects of mono-(2-ethylhexyl) phthalate (MEHP) on chicken germ cells cultured in vitro.

In recent decades, many toxicological tests based on in vivo or in vitro models, mainly from mammalian (rat-mouse) and fish species, were used to assess the risks raised by contact or ingestion of molecules of pharmaceutical, agricultural, or natural origin. But no, or few, in vitro tests using other non-mammalian models such as bird have been explored despite their advantages: the embryonic gonads of birds have a high plasticity of development sensitive to estrogen, and sperm production is nearly two times faster than in rodents. Hence, we have established an in vitro culture of germ cells an…

Lineage enforcement by inductive mesenchyme on adult epithelial stem cells across developmental germ layers.

During development, cell differentiation is accompanied by the progressive loss of pluripotent gene expression and developmental potential, although de-differentiation in specialized cells can be induced by reprogramming strategies, indicating that transdifferentiation potential is retained in adult cells. The stromal niche provides differentiating cues to epithelial stem cells (SCs), but current evidence is restricted to tissue types within the same developmental germ layer lineage. Anticipating the use of adult SCs for tissue regeneration, we examined if stroma can enforce lineage commitment

FISH studies of chromosome abnormalities in germ cells and its relevance in reproductive counseling.

Chromosome abnormalities are one of the major causes of human infertility. In infertile males, abnormal karyotypes are more frequent than in the general population. Furthermore, meiotic disorders affecting the germ cell-line have been observed in men with normal somatic karyotypes consulting for infertility. In both cases, the production of unbalanced spermatozoa has been demonstrated. Basically addressed to establish reproductive risks, fluorescence in situ hybridization (FISH) on decondensed sperm heads has become the most frequently used method to evaluate the chromosomal constitution of sp

Repair in haploid male germ cells occurs late in differentiation as chromatin is condensing.

Huntington's Disease (HD) is one of eight progressive neurodegenerative disorders in which the underlying mutation is a CAG expansion encoding a polyglutamine tract. The mechanism of trinucleotide expansion remains poorly understood. We have followed heritable changes in CAG length in male transgenic mice. In germ cells, expansion is limited to the post-meiotic, haploid cell and therefore cannot involve mitotic replication or recombination between a homologous chromosome and a sister chromatid. Expansion occurs by gap filling synthesis when DNA loops comprising the CAG trinucleotide repeats ar…

Analysis of meiosis in intratesticular germ cells from subjects affected by classic Klinefelter's syndrome.

Azoospermic subjects affected by Klinefelter's syndrome may occasionally show the presence of intratesticular residual foci of spermatogenesis, and the retrieval of mature spermatozoa from the testis may permit fertility and paternity by means of intracytoplasmic sperm injection. Previous studies have demonstrated that these subjects show the presence of an increased incidence of hyperaploid spermatozoa. Here we analyzed, by fluorescence in situ hybridization using specific probes for chromosomes 8, X, and Y, the spermatogenic process and the meiotic progression of 47,XXY germ cells retrieved …

Involvement of Fas in the apoptosis of mouse germ cells induced by experimental cryptorchidism.

The role of Fas in the apoptosis of testicular germ cells was investigated in BALB/c mice and Fas-deficient lpr/lpr mice. Spontaneous apoptosis of germ cells was observed in the testes of 40-day-old BALB/c mice, and experimentally induced cryptorchidism increased this apoptosis to such an extent that there was a decrease in the weight of the testis. Flow cytometry and immunohistochemistry using a Fas-specific monoclonal antibody demonstrated expression of Fas on germ cells including spermatogonia, spermatocytes, and spermatids. Furthermore, analysis by flow cytometry suggested that Fas express…

Prolonged expression of the c-kit receptor in germ cells of intersex fetal testes.

Stem cell factor (SCF) and its receptor Kit encoded by the c-kit proto-oncogene are crucial for the development and migration of primordial germ cells in rodents. The expression of Kit has been examined immunohistochemically in gonads obtained from five specimens of fetal tissues with intersex conditions which included 45,X/46,XY mosaicism; androgen insensitivity syndrome; and 46,XY/iso(p)Y mosaicism. Individuals with such disorders of sexual differentiation and Y-chromosome material carry a very high risk of developing testicular neoplasms. Fetal testicular germ cells of the intersex subjects…

Chromosomal aberrations in germ cells of male mice immunised with attenuated viral vaccines (human).

The cytogenetic effects of two attenuated viral vaccines (yellow fever vaccine and oral poliomyelitis vaccine) were assessed by means of the analysis of meiotic spermatocyte chromosomes in mice. In a dose of 0.5 ml, but not 0.1 ml, both vaccines induced a significant percentage of chromosomal aberrations after 7, 14 and 30 days. Euploidy was the major abnormality produced by yellow fever vaccine. The various abnormalities produced by oral polio vaccine were significant when pooled, but not when analysed individually. More abnormalities were produced by yellow fever vaccine than by oral polio v

Identification of testicular atypical germ cells by an immunohistochemical technique for placental alkaline phosphatase.

The identification of atypical testicular germ cells is often difficult by by routine histologic examination. By immunohistochemical detection of placental alkaline phosphatase (PLAP) and by periodic acid Schiff staining of glycogen, atypical germ cells were easily identified in testicular samples. Forty-one fetal and adult testes were used for a preliminary study, and 121 testes from infants and adults with either cryptorchidism or germ cell tumors were studied for the presence of atypical germ cells. Two types of clear germ cells were differentiated histochemically, and one with PLAP-positiv…

The piRNA protein Asz1 is essential for germ cell and gonad development in zebrafish and exhibits differential necessities in distinct types of germ granules.

Germ cells are essential for fertility, embryogenesis, and reproduction. Germline development requires distinct types of germ granules, which contains RNA-protein (RNP) complexes, including germ plasm in embryos, piRNA granules in gonadal germ cells, and the Balbiani body (Bb) in oocytes. However, the regulation of RNP assemblies in zebrafish germline development are still poorly understood. Asz1 is a piRNA protein in Drosophila and mice. Zebrafish Asz1 localizes to both piRNA and Bb granules, with yet unknown functions. Here, we hypothesized that Asz1 functions in germ granules and germline d…

The RNA-Binding Protein IGF2BP1 Marks Germ Cells but Is Dispensable for Mouse Fertility.

Insulin-like growth factor 2 mRNA binding protein 1 (IGF2BP1) is a key reader of N6-methyladenosine modifications that regulate target mRNA stability in eukaryotic cells; however, its role in germ cells has never been explored. Here, we analyzed the spatiotemporal expression of IGF2BP1 and revealed that it was present not only in oocytes of the mouse ovary but also in ZBTB16-positive undifferentiated spermatogonia in the mouse testis. Coimmunoprecipitation and fluorescence staining revealed that IGF2BP1 interacted with TRIM71, a regulator of spermatogonia differentiation, but that its expressi…

A Comparative Study of Transfection Techniques for Genetic Modification in Chicken Primordial Germ Cells.

Avian species possess a distinct reproductive system that differs fundamentally from mammals, resulting in relatively slower progress in transgenic poultry research compared to mammalian models. Primordial germ cells (PGCs), as the precursors to functional gametes, provide a promising avenue for the utilization of genetic resources and the application of transgenesis in chickens. However, the transfection of chicken PGCs remains challenging due to the low efficiency of conventional methods. This study systematically compared the transient transfection efficiency of chicken PGCs using lipofecti…

Characterization of Ucp1-iCre knockin mice reveals the recombination activity in male germ cells.

Ucp1 promoter-driven Cre transgenic mice are useful in the manipulation of gene expression specifically in thermogenic adipose tissues. However, the wildly used Ucp1-Cre line was generated by random insertion into the genome and showed ectopic activity in some tissues beyond adipose tissues. Here, we characterized a knockin mouse line Ucp1-iCre generated by targeting IRES-Cre cassette immediately downstream the stop codon of the Ucp1 gene. The Cre insertion had little to no effect on uncoupling protein 1 (UCP1) levels in brown adipose tissue. Ucp1-iCre mice of both genders exhibited normal the

Investigation of Signals and Transcription Factors for The Generation of Female Germ-Like Cells.

Objective: Primordial germ cell (PGCs) lines are a source of a highly specialized type of cells, characteristically oocytes,
during female germline development in vivo. The oocyte growth begins in the transition from the primary follicle. It is
associated with dynamic changes in gene expression, but the gene-regulating signals and transcription factors that control oocyte growth remain unknown. We aim to investigate the differentiation potential of mouse bone marrow mesenchymal stem cells (mMSCs) into female germ-like cells by testing several signals and transcriptio
