Dietary Tryptophan Enhances Aryl Hydrocarbon Receptor Activation and Reduces Colitis Through Microbial Metabolism.

BACKGROUND & AIMS: Disrupted microbial tryptophan metabolism and impaired aryl hydrocarbon receptor activation are implicated in inflammatory bowel disease pathogenesis. However, strategies to restore this pathway through diet or microbial modulation remain poorly defined. This study investigates how dietary tryptophan and human and mouse microbiota modulate metabolism, aryl hydrocarbon receptor activation, and intestinal inflammation in preclinical models. METHODS: Gnotobiotic mice colonized with microbiota of varying complexity or human fecal microbiota from patients with ulcerative colitis

Harnessing a germ‑free mouse gut bioreactor for directed evolution of probiotics to combat non-alcoholic fatty liver disease.

Directed evolution has transformed biomolecular engineering but remains largely untapped for probiotic optimization, despite its immense promise for human health maintenance and disease therapy. Here, we present an in vivo, host-mediated directed-evolution platform that harnesses the gut's endogenous selective pressures to drive functional enhancement of probiotics. Using Bifidobacterium animalis subsp. lactis as a model, we expose germ-free male mice to stepwise increases in bile-acid stress via a high-fat, high-cholesterol diet. Compared to in vitro evolution, which fails to produce any func

Hematopoietic MyD88 orchestrates the control of gut colonization by segmented filamentous bacteria.

Host-microbiota cooperation is critical for successful intestinal homeostasis. The commensal segmented filamentous bacteria (SFB) are crucial for orchestrating the post-natal maturation of the host gut immune system and establishing a healthy state of physiological inflammation, which largely depends on their intimate attachment to the ileal mucosa. However, the signaling pathways used by SFB to induce gut immune responses and how such responses ultimately control SFB colonization remain controversial. Using gnotobiotic approaches, we showed that SFB load is controlled by complex interactions

Role of gut microbiota in regulating gastrointestinal dysfunction and motor symptoms in a mouse model of Parkinson's disease.

Parkinson's disease (PD) is a common neurodegenerative disorder characterized primarily by motor and non-motor gastrointestinal (GI) deficits. GI symptoms' including compromised intestinal barrier function often accompanies altered gut microbiota composition and motor deficits in PD. Therefore, in this study, we set to investigate the role of gut microbiota and epithelial barrier dysfunction on motor symptom generation using a rotenone-induced mouse model of PD. We found that while six weeks of 10 mg/kg of chronic rotenone administration by oral gavage resulted in loss of tyrosine hydroxylase

The effect of Ganodermalucidum spore oil in early skin wound healing: interactions of skin microbiota and inflammation.

The mushroom Ganoderma lucidum (G. lucidum Leyss. ex Fr.) Karst has been a traditional Chinese medicine for millennia. In this study, we isolated the Ganoderma lucidum spore oil (GLSO) and evaluated the effect of GLSO on skin burn wound healing and the underlying mechanisms. Mice were used to perform skin wound healing assay. Wound analysis was performed by photography, hematoxylin/eosin staining, Masson's Trichrome staining and immunohistochemical analysis. Microbiota on the wounds were analyzed using the 16s rRNA sequence and quantitative statistics. The lipopolysaccharide (LPS) content was

Pathogenesis of Human Norovirus Genogroup II Genotype 4 in Post-Weaning Gnotobiotic Pigs.

Norovirus is the most common cause of acute gastroenteritis. Its pathogenesis is poorly understood owing to the difficulty of establishing viral infection in animal models. Here, post-weaning gnotobiotic pigs were infected with human norovirus genogroup II genotype 4 (HuNoV GII.4) to investigate the pathogenesis and replication of the virus. Three groups of four pigs were infected with 1 × 10⁵, 1 × 10⁶, or 1 × 10⁷ genomic equivalent (GE) copies of HuNoV GII.4. Four pigs were used as negative controls. Blood and rectal swab samples were collected after viral infection, and gross legions were ex

Effects of a gut pathobiont in a gnotobiotic mouse model of childhood undernutrition.

To model how interactions among enteropathogens and gut microbial community members contribute to undernutrition, we colonized gnotobiotic mice fed representative Bangladeshi diets with sequenced bacterial strains cultured from the fecal microbiota of two 24-month-old Bangladeshi children: one healthy and the other underweight. The undernourished donor's bacterial collection contained an enterotoxigenic Bacteroides fragilis strain (ETBF), whereas the healthy donor's bacterial collection contained two nontoxigenic strains of B. fragilis (NTBF). Analyses of mice harboring either the unmanipulate

Members of the human gut microbiota involved in recovery from Vibrio cholerae infection.

Given the global burden of diarrhoeal diseases, it is important to understand how members of the gut microbiota affect the risk for, course of, and recovery from disease in children and adults. The acute, voluminous diarrhoea caused by Vibrio cholerae represents a dramatic example of enteropathogen invasion and gut microbial community disruption. Here we conduct a detailed time-series metagenomic study of faecal microbiota collected during the acute diarrhoeal and recovery phases of cholera in a cohort of Bangladeshi adults living in an area with a high burden of disease. We find that recovery

Chronic active gastritis due to Helicobacter pylori in immunized gnotobiotic piglets.

In gnotobiotic piglets, parenteral vaccination with formalin-killed Helicobacter pylori and oral vaccination with live bacteria induced H. pylori-specific serum immunoglobulins G, M, and A. Vaccination reduced but did not prevent infection by subsequent challenge with viable H. pylori. Oral vaccination with killed bacteria was less effective in inducing serum antibody and had no effect on bacterial colonization. Immunization status influenced the histologic response of piglets to challenge by H. pylori. Lymphoplasmacytic gastritis was more severe in parenterally vaccinated piglets than in the

Evidence for tumor necrosis factor-induced pulmonary microvascular injury after intestinal ischemia-reperfusion injury.

Acute lung injury characterized by increased microvascular permeability is one feature of multiple-organ system failure and the adult respiratory distress syndrome. Intestinal ischemia-reperfusion injury has been linked to this type of acute lung injury. The purpose of these experiments was to examine the pathogenic mediators that link the two processes, with particular emphasis on the roles of endotoxin and tumor necrosis factor alpha (TNF alpha). Previously described characteristics of the acute lung injury in this rat model of intestinal ischemia-reperfusion include pulmonary neutrophil seq

Experimentally induced coronavirus infections in calves: viral replication in the respiratory and intestinal tracts.

Eleven 3- to 50-day-old colostrum-deprived gnotobiotic calves and seven 25- to 63-day-old colostrum-deprived conventional calves were allotted into 3 groups. Each group was inoculated with a fecal isolate of bovine coronavirus via different routes: orally/intranasally OR/IN, No. 1 through 8, group 1 calves; OR, No. 9 through 13, group 2 calves; IN, No. 14 through 18, group 3 calves. Nasal swab specimens and fecal specimens were collected daily and were examined for coronavirus antigen by use of direct immunofluorescent staining (nasal epithelial cells) or by use of immune electron microscopy (

The physiological importance of the colonic microflora.

The present concept of the intestinal microflora has been reviewed with stressing the fact that it represents an entity with a weight comparable to one of the larger organs of the body. It is composed of about 500 different strains, many of which as yet have not been isolated because 95% of them are anaerobic and fastidious in their growth requirements. The intimate relationship of the flora to the surface epithelium of the intestines and the Lieberkühn's crypts are brought forward. The production of germfree animals is reviewed as an accounting of the most obvious differences between the germ

Intestinal microbiota influence the early postnatal development of the enteric nervous system.

BACKGROUND: Normal gastrointestinal function depends on an intact and coordinated enteric nervous system (ENS). While the ENS is formed during fetal life, plasticity persists in the postnatal period during which the gastrointestinal tract is colonized by bacteria. We tested the hypothesis that colonization of the bowel by intestinal microbiota influences the postnatal development of the ENS. METHODS: The development of the ENS was studied in whole mount preparations of duodenum, jejunum, and ileum of specific pathogen-free (SPF), germ-free (GF), and altered Schaedler flora (ASF) NIH Swiss mic

MaaT013 fecal microbiotherapy modulates key gastro-intestinal and immunological processes involved in GvHD pathogenesis in vitro and in vivo.

BACKGROUND: Acute graft-vs.-host disease (aGvHD) is a life-threatening complication of allogeneic hematopoietic cell transplantation (allo-HCT), where allogeneic donor T cells recognize recipient tissues as foreign and damage them. Gut microbiota disruption following allo-HCT has been associated with increased aGvHD incidence, severity, and mortality. Growing clinical evidence suggests the efficacy of fecal microbiotherapy to restore a healthy and diverse gut microbiota and to treat aGvHD. MaaT013 (Xervyteg®), a standardized allogeneic fecal microbiotherapy derived from pooled healthy human fe

The Inflammatory Response in the Murine Ovalbumin-Induced Skin Inflammation Model Is Robust Despite Major Gut Microbiota Differences.

Valid pre-clinical animal models are essential for effective drug development in inflammatory skin diseases like atopic dermatitis. Most murine models exhibit low-to-moderate inflammatory responses with significant variability, undermining their validity and contributing to high drug attrition rates. Previous studies showed that microbiota transfer and early-life microbial exposure affect inflammatory phenotypes and allergy development. This concept might extend to other pre-clinical inflammatory models. We tested the impact of different microbiotas on ovalbumin (OVA)-induced skin inflammation

Transplant of cryopreserved mosquito gut microbiota reveals strain-specific patterns of microbial acquisition and assembly.

Mosquitoes are important vectors of human pathogens, and their gut microbiome influences traits such as survival, fecundity, pathogen resistance, insecticide resistance, and vector competence. Manipulating this microbiome offers a promising strategy for reducing mosquito-borne disease. While 16S rRNA gene amplicon sequencing has revealed global patterns of gut microbial diversity, the factors driving these patterns remain poorly understood. Here, we used a community-transplantation approach that allows the collection, cryopreservation, and transfer of whole adult microbial communities into axe
