Distinct laterality alterations distinguish mild cognitive impairment and Alzheimer's disease from healthy aging: statistical parametric mapping with high resolution MRI.

Laterality of human brain varies under healthy aging and diseased conditions. The alterations in hemispheric asymmetry may embed distinct biomarkers linked to the disease dynamics. Statistical parametric mapping based on high-resolution magnetic resonance imaging (MRI) and image processing techniques have allowed automated characterization of morphological features across the entire brain. In this study, 149 subjects grouped in healthy young, healthy elderly, mild cognitive impairment (MCI), and Alzheimer's disease (AD) were investigated using multivariate analysis for regional cerebral latera…

Aging and drug interactions. II. Effect of phenytoin and smoking on the oxidation of theophylline and cortisol in healthy men.

The effect of age on the induction of theophylline metabolism by phenytoin was examined in healthy young and old male cigarette smokers (greater than or equal to 20 cigarettes/day) and nonsmokers. Two single dose studies of theophylline pharmacokinetics were performed, one as a base-line control and another after a 2-week course of phenytoin. Phenytoin was administered as an i.v. loading dose followed by oral ingestion. The dose was adjusted to achieve total phenytoin plasma concentrations within a low therapeutic range (10-13 micrograms/ml). Free phenytoin concentrations in plasma were slight

A chronological age prediction model using clinical biochemistry and hematological data identifies features of biological age in healthy Labrador retrievers.

We hypothesised that clinical blood biochemistry and haematological data could predict the age of healthy dogs. Using a large veterinary electronic medical record (EMR) database, we investigated age-related relationships in Labrador Retrievers, a common large breed. Analysing over 3 million blood test values across 24 analytes and ~ 145,000 dogs, we modelled age and sex using a generalised additive model. Changes in multiple analytes were seen around 3-6 and 12 years of age. As these findings demonstrated age-related changes in blood analytes, we considered the data suitable for modelling chro

Coronary artery calcium as a marker of healthy and unhealthy aging in adults aged 75 and older: The Atherosclerosis Risk in Communities (ARIC) study.

BACKGROUND AND AIMS: Coronary artery calcium (CAC) is validated for risk prediction among middle-aged adults, but there is limited research exploring implications of CAC among older adults. We used data from the Atherosclerosis Risk in Communities (ARIC) study to evaluate the association of CAC with domains of healthy and unhealthy aging in adults aged ≥75 years. METHODS: We included 2,290 participants aged ≥75 years free of known coronary heart disease who underwent CAC scoring at study visit 7. We examined the cross-sectional association of CAC = 0, 1-999 (reference), and ≥1000 with seven d…

Concomitant administration of varicella vaccine with combined measles, mumps, and rubella vaccine in healthy children aged 12 to 24 months of age.

The reactogenicity and immunogenicity of three combined measles, mumps and rubella (MMR) vaccines and one administered with a varicella vaccine was studied in infants. The vaccines were Priorix (designated MeMuRu, Group 1), M-M-R II (Group 2), Triviraten (Group 3) and Priorix + a varicella vaccine, Varilrix (Group 4). Fever was greater in Group 2 (61.3%) compared to Group 1 (48.5%; p = 0.033) or Group 3 (37.1%; p = 0.009). Rash with fever was reported in Group 2 (4.8%) and Group 4 (3.3%), but not for Group 1. Anti-measles, -mumps and -rubella seroconversion was similar for Group 1 (96.1%, 96.1

Pain and self-rated health among middle-aged and older Canadians: an analysis of the Canadian community health survey-healthy aging.

BACKGROUND: Pain is an important health problem adversely affecting functionality and quality of life. Though self- rated health (SRH) is a major predictor of mortality, its relationship with pain is not well understood. We explore 1) how pain and age interact to influence SRH, and 2) provincial variations in SRH across Canada. METHODS: We analyzed cross-sectional data from Statistics Canada's Canadian Community Health Survey-Healthy Aging (n = 30,685), which targeted those 45 years and older and was conducted from 2008 to 12-01 to 2009-11-30. The response rate was 74.4%.The topics covered in…

Age and education effects and norms on a cognitive test battery from a population-based cohort: the Monongahela-Youghiogheny Healthy Aging Team.

OBJECTIVES: Performance on cognitive tests can be affected by age, education, and also selection bias. We examined the distribution of scores on several cognitive screening tests by age and educational levels in a population-based cohort. METHOD: An age-stratified random sample of individuals aged 65+ years was drawn from the electoral rolls of an urban US community. Those obtaining age and education-corrected scores > or = 21/30 on the Mini-Mental State Examination (MMSE) were designated as cognitively normal or only mildly impaired, and underwent a full assessment including a battery of neu

Impact of the SARS-CoV-2 pandemic on healthy aging and functionality in older Mexican adults: insights from the MHAS cohort.

BACKGROUND: Older adults were among the most affected by the COVID-19 pandemic, facing greater vulnerability to infection, hospitalization, and post-infection sequelae. However, evidence on its multidimensional impact on healthy aging remains limited, particularly in Latin America. This study examined the association of COVID-19 infection, hospitalization, and vaccination with healthy aging and functional impairment among older Mexican adults. METHODS: We analyzed longitudinal data from the Mexican Health and Aging Study (MHAS), comparing pre-pandemic (2018) and post-pandemic (2021) waves. Tw…

Parental socioeconomic position, childhood poverty and healthy aging: retrospective and prospective study of six cohorts across 32 wealthy and developing nations.

BACKGROUND AND AIM: Children who were poor in wealthy countries have reported worse cognitive, muscle, and mental functions, as well as greater frailty and multimorbidity, in later life. However, it remains unclear whether those who were poor in childhood worldwide fall short of healthy aging, as information on childhood conditions is often unreliable. Here, I present new evidence on the lifecourse shaping of healthy aging among older adults worldwide. METHODS: Some 80,000 older adults aged 50 and over in 32 wealthy and developing nations recalled their childhood conditions at ages 10-14 and …

Body mass index for the assessment of adiposity in healthy aging Caucasian women: a dual-energy X-ray absorptiometry based study.

Accurate classification of obesity levels is of clinical importance for public health and decision making. Body mass index (BMI) is the most broadly used measure of obesity on a population scale. But, as with other diagnostic indicators, BMI can have potential pitfalls. We attempted to explore the diagnostic performance of BMI for assessing obesity. 330 healthy Caucasian women (native Greeks) (ages, 20 to 85 years) volunteers participated in the study. Dual-energy X-ray absorptiometry (DXA) was used to measure percentage of fat mass (%FM). The diagnostic performance of BMI was assessed using t

A Defined Synbiotic Produces Immunomodulatory Metabolites, Engages Gut-Immune Pathways Relevant to Inflammaging, and Supports Healthy Aging in a Nematode Model.

Chronic low-grade aging-associated inflammation, or inflammaging, is a central pillar of age-related decline in quality of life. Inflammaging is partially mediated by impaired intestinal, immune, and microbiome function, and it has been hypothesized that probiotics could be used to promote healthy aging. SBD121, a defined synbiotic containing food-derived microbial strains and prebiotic fibers, has previously been shown to improve grip strength in male rats, an important indicator of healthspan, and is under evaluation in a clinical trial of 143 newly diagnosed rheumatoid arthritis patients (N…

Performance fatigability on a constant-load dynamic task is greater in females with moderate-severity Parkinson's disease than healthy-aging controls.

Parkinson's disease (PD) is a neurodegenerative disorder associated with self-reported fatigue. While fatigue is a disabling symptom, performance fatigability, a decline in strength or power during sustained tasks, remains poorly understood, particularly during isotonic (constant-load) contractions, which are encountered in everyday activities. We assessed performance fatigability and recovery in people with PD compared to healthy-aging controls. Fifteen females with PD and 14 healthy-aging controls underwent neuromuscular testing of knee extensors using dynamometry. Participants then performe

Impact of sensory organization tasks on prefrontal cortex activity in older women: a comparative fNIRS study of osteoarthritis and healthy aging.

INTRODUCTION: Osteoarthritis (OA), a prevalent musculoskeletal condition, is associated with an increased risk of falls. Maintaining posture relies on visual, vestibular, and proprioceptive inputs, but these systems can be compromised due to aging or disease, heightening fall risk. Such impairments may result from neuromuscular decline and reduced cognitive or visuospatial processing abilities. This study aimed to investigate prefrontal cortical (PFC) activation patterns during clinical sensory organization tasks (SOT) using functional near-infrared spectroscopy (fNIRS) in older women with OA

Multivariate Computational Analysis of Gamma Delta T Cell Inhibitory Receptor Signatures Reveals the Divergence of Healthy and ART-Suppressed HIV+ Aging.

Even with effective viral control, HIV-infected individuals are at a higher risk for morbidities associated with older age than the general population, and these serious non-AIDS events (SNAEs) track with plasma inflammatory and coagulation markers. The cell subsets driving inflammation in aviremic HIV infection are not yet elucidated. Also, whether ART-suppressed HIV infection causes premature induction of the inflammatory events found in uninfected elderly or if a novel inflammatory network ensues when HIV and older age co-exist is unclear. In this study we measured combinational expression
