Atlas selection for hippocampus segmentation: Relevance evaluation of three meta-information parameters.

Current state-of-the-art methods for whole and subfield hippocampus segmentation use pre-segmented templates, also known as atlases, in the pre-processing stages. Typically, the input image is registered to the template, which provides prior information for the segmentation process. Using a single standard atlas increases the difficulty in dealing with individuals who have a brain anatomy that is morphologically different from the atlas, especially in older brains. To increase the segmentation precision in these cases, without any manual intervention, multiple atlases can be used. However, reg…

Linking persistent negative symptoms to amygdala-hippocampus structure in first-episode psychosis.

Early persistent negative symptoms (PNS) following a first episode of psychosis (FEP) are linked to poor functional outcome. Reports of reduced amygdalar and hippocampal volumes in early psychosis have not accounted for heterogeneity of symptoms. Age is also seldom considered in this population, a factor that has the potential to uncover symptom-specific maturational biomarkers pertaining to volume and shape changes within the hippocampus and amygdala. T1-weighted volumes were acquired for early (N=21), secondary (N=30), non-(N=44) PNS patients with a FEP, and controls (N=44). Amygdalar-hippoc…

Selective Oestrogen Receptor Agonists Rescued Hippocampus Parameters in Male Spontaneously Hypertensive Rats.

Spontaneously hypertensive rats (SHR) show pronounced hippocampus alterations, including low brain-derived neurotrophic factor (BDNF) expression, reduced neurogenesis, astrogliosis and increased aromatase expression. These changes are reverted by treatment with 17β-oestradiol. To determine which oestradiol receptor (ER) type is involved in these neuroprotective effects, we used agonists of the ERα [propylpyrazole triol (PPT)] and the ERβ [diarylpropionitrite (DPN)] given over 2 weeks to 4-month-old male SHR. Wistar Kyoto normotensive rats served as controls. Using immunocytochemistry, we deter…

Neonatal binge alcohol exposure increases microglial activation in the developing rat hippocampus.

Aberrant activation of the developing immune system can have long-term negative consequences on cognition and behavior. Teratogens, such as … F-β compared to both SI and SCs. In summary, the number and activation of microglia in the neonatal hippocampus are both affected in a rat model of FASD, along with increased gene expression of pro- and anti-inflammatory cytokines. This study shows that alcohol exposure during development induces a neuroimmune response, potentially contributing to long-term a …

Proteomic changes in rat hippocampus and adrenals following short-term sleep deprivation.

BACKGROUND: To identify the biochemical changes induced by sleep deprivation at a proteomic level, we compared the hippocampal proteome of rats either after 4 hours of sleep or sleep deprivation obtained by gentle handling. Because sleep deprivation might induce some stress, we also analyzed proteomic changes in rat adrenals in the same conditions. After sleep deprivation, proteins from both tissues were extracted and subjected to 2D-DIGE analysis followed by protein identification through mass spectrometry and database search. RESULTS: In the hippocampus, 87 spots showed significant variatio…

Growth hormone enhances excitatory synaptic transmission in area CA1 of rat hippocampus.

The hippocampus produces growth hormone (GH) and contains GH receptors, suggesting a potential role for GH signaling in the regulation of hippocampal function. In agreement with this possibility, previous investigations have found altered hippocampal function and hippocampal-dependent learning and memory after chronic GH administration or deficiency. In this study we applied GH to in vitro rat hippocampal brain slices, to determine whether GH has short-term effects on hippocampal function in addition to previously documented chronic effects. We found that GH enhanced both AMPA- and NMDA-recept…

Disruption of MAP-2 immunostaining in rat hippocampus after traumatic brain injury.

The effects of diffuse brain injury on dendritic morphology in rat hippocampus and cortex were examined in this study using the recently described impact acceleration model of traumatic brain injury (Marmarou et al., 1994). Dendritic structure was visualized using immunostaining of microtubule associated protein-2 (MAP-2). Brains were studied 24, 48, and 72 h after brain injury. Results from immunohistochemistry and light microscopy indicated a time-dependent disruption of dendritic cytoarchitecture in the CA1 subregion and in the hilus of the hippocampus but not in the dentate gyrus or CA3 su…

Changes in IP3 3-kinase immunoreactivity following transient ischaemia in gerbil hippocampus.

The distribution of inositol 1,4,5-triphosphate 3-kinase (IP3 3-kinase) in the gerbil hippocampus was studied before and after transient ischaemia, by immunohistochemistry and immunoblot with monoclonal antibody to IP3 3-kinase. In control gerbils, intense IP3 3-kinase immunoreactivity was localized in the dendritic field of CA1 pyramidal neurones. However, after ischaemia for 5 min, the immunoreactivity decreased gradually, until after 24 h there was very little IP3 3-kinase detectable. Immunoblot study showed a similar decrease of IP3 3-kinase in ischaemic hippocampus. Infliction of 2 min is…

Agonists at metabotropic glutamate receptors presynaptically inhibit EPSCs in neonatal rat hippocampus.

1. The effects of metabotropic glutamate receptor agonists on excitatory synaptic transmission in the CA1 region of rat hippocampal slices (11-30 days) were studied using extracellular and whole-cell patch-clamp recording techniques. 2. Trans-1-amino-1,3-cyclopentanedicarboxylic acid (trans-ACPD; 25-100 microM) reversibly depressed excitatory postsynaptic currents (EPSCs) without affecting presynaptic fibre excitability or EPSC reversal potential. 3. DSP-4 treatment produces abnormal tyrosine hydroxylase immunoreactive fibers in rat hippocampus.

DSP-4 has been used as selective toxin for central norepinephrine (NE) systems. The depletion of NE by DSP-4 is though to result from neurotoxic destruction of locus coeruleus terminals and axons. We have used tyrosine hydroxylase immunocytochemistry (TH-IR), silver stains for neural degeneration, and electron microscopy to examine the morphological effects of DSP-4 treatment on the rat hippocampus. Animals survived for either 2 or 5 weeks after DSP-4 treatment. DSP-4 depleted hippocampal norepinephrine levels to 15% of control values. Abnormally enlarged TH-IR fibers were found in the hippoca…

The ventral hippocampus and fear conditioning in rats: different anterograde amnesias of fear after infusion of N-methyl-D-aspartate or its noncompetitive antagonist MK-801 into the ventral hippocampus.

Previous studies on hippocampal involvement in classical fear conditioning mainly focused on the dorsal hippocampus and conditioning to a context. However, in line with the strong interconnectivity of the ventral hippocampus with amygdala and nucleus accumbens, more recent studies indicated an even more global role for the ventral hippocampus in fear conditioning. The present study examined the formation of classical fear conditioning to explicit and contextual cues following stimulation or blockade of N-methyl-D-aspartate (NMDA) receptors in the ventral hippocampus. NMDA (0.5 microg/side) or …

Cell-state mapping reveals a reversible neuroblast accumulation in the aging mouse hippocampus.

Hippocampal neurogenesis supports learning and memory by generating new granule cells throughout life. However, both the rate and speed of this process decline with age. To elucidate the molecular determinants underlying the effects of aging on neuronal differentiation, we combined lineage tracing of adult-born granule cells (aGCs) with single-nucleus RNA sequencing. This approach produced a temporally resolved transcriptional atlas of aGC development. Aging led to a marked accumulation of postmitotic neuroblasts (NBs), revealing a stage-specific bottleneck in the neurogenic trajectory. Volunt

Neural geometry in the human hippocampus enables generalization across spatial position and gaze.

Hippocampal neurons track the positions of self, others, and gaze direction. However, it is unclear how their respective neural codes differ enough to avoid confusion while allowing for abstraction across spatial frames. We recorded from populations of hippocampal neurons while human participants performed a joystick-controlled virtual prey-pursuit task. We found that neurons have mixed selective responses that map the positions of self, prey, and predator, as well as gaze direction. Their codes occupied mostly orthogonal subspaces, but the geometric structures of these subspaces allowed them

Severity-dependent proteomic alterations in the rat hippocampus following pilocarpine-induced status epilepticus.

Status epilepticus (SE) is a prolonged seizure state that can induce lasting hippocampal damage and promote the development of spontaneous seizures and cognitive deficits. The severity and duration of SE affect long-term outcomes; however, many studies rely on behavioral assessments such as the Racine scale, which may miss subclinical or non-convulsive seizures and leave the molecular effects of mild SE unclear. Here, we examined whether behavioral seizure severity in the pilocarpine rat model of acquired epilepsy correlates with distinct hippocampal proteomic changes. Adult male rats were cla

FED-UNet++: An Improved Nested UNet for Hippocampus Segmentation in Alzheimer's Disease Diagnosis.

The hippocampus is a key structure involved in the early pathological progression of Alzheimer's disease. Accurate segmentation of this region is vital for the quantitative assessment of brain atrophy and the support of diagnostic decision-making. To address limitations in current MRI-based hippocampus segmentation methods-such as indistinct boundaries, small target size, and limited feature representation-this study proposes an enhanced segmentation framework called FED-UNet++. The residual feature reconstruction block (FRBlock) is introduced to strengthen the network's ability to capture bou…

Limited Microvascular Remodelling Occurs in the Aged Human Hippocampus in Obstructive Sleep Apnoea.

In mice, intermittent hypoxia is associated with an increase in microvessels in the hippocampus, whereas in humans with obstructive sleep apnoea (OSA), microvessels are lost from the heart and retina. The present study investigated microvascular changes in the hippocampus of patients with OSA, and whether patient age or use of continuous positive airway pressure (CPAP) influence microvascularisation. Using autopsy samples from 31 people with confirmed OSA, microvessels were immunolabelled and quantitatively analysed. Compared to the Low OSA group, the High OSA group had larger mean microvessel…
