Platelet agonists and subendothelial extracellular matrix (ECM) induce morphological and biochemical changes in animal megakaryocytes, reminiscent of the response of platelets to the same substances. We have examined the behavior of human megakaryocytes exposed for up to 36 hours to the ECM produced by cultured bovine corneal endothelial cells. By phase contrast and scanning electron microscopy these megakaryocytes demonstrated non-reversible adherence and flattening with formation of long filopodia, thus confirming that human megakaryocytes acquire platelet functional capacities. In addition,

BACKGROUND: Inflammation during pregnancy is an important contributor to maternal and offspring morbidity and mortality. Evidence from both nonpregnant human and animal studies suggests that dietary choline can attenuate inflammation, but this has not yet been explored in human pregnancy.
OBJECTIVES: This study explored the cross-sectional associations between maternal mid-pregnancy dietary choline intake and inflammation biomarkers, specifically IL-6, tumor necrosis factor- α (TNF-α), and C-reactive protein (CRP), while also examining the modifying effects of other methyl donor nutrients.

Contactin-associated protein-like 4 (Cntnap4) is a member of the neurexin superfamily of transmembrane molecules that have critical functions in neuronal cell communication. Cntnap4 knockout mice display decreased presynaptic gamma-aminobutyric acid (GABA) and increased dopamine release that is associated with severe, highly penetrant, repetitive, and perseverative movements commonly found in human autism spectrum disorder patients. However, no known function of Cntnap4 has been revealed besides the nervous system. Meanwhile, secretory protein neural EGFL-like 1 (Nell-1) is known to exert pote

Cancer-associated thrombosis (CAT) is the second leading cause of death among cancer patients. Its pathogenesis is multifactorial and involves dynamic interactions among tumor cells, platelets, coagulation factors, endothelial cells, immune cells, and blood flow. Traditional in vitro 2-dimensional models fail to accurately mimic the dynamic microenvironment of CAT in humans. Although animal models can mimic complex pathological conditions, significant differences in the composition and regulation of the coagulation system between animals and humans limit their ability to accurately model CAT i

Members of the alpha-proteobacteria display a broad range of interactions with higher eukaryotes. Some are pathogens of humans, such as Rickettsia and Bartonella that are associated with diseases like epidemic typhus, trench fever, cat scratch disease and bacillary angiomatosis. Others like the Brucella cause abortions in pregnant animals. Yet other species have evolved elaborate interactions with plants; in this group we find both plant symbionts and parasites. Despite radically different host preferences, extreme genome size variations and the absence of toxin genes, similarities in survival

The threat of disease transmission at the intersection of human-wildlife interfaces underscores the urgent need for detailed studies on the transmission of human-borne pathogens across species, especially among nonhuman primates in urban areas. This research focuses on the social and demographic determinants of Mycobacterium tuberculosis complex (MTBC) infection in free-ranging long-tailed macaques (Macaca fascicularis) in Thailand. Behavioral observations and noninvasive biological specimens (freshly defecated feces and rope-baited oral samples) were collected from 98 long-tailed macaques liv

The discontinuation of smallpox vaccination will lead to a gradual decrease or disappearance of immunity to poxviruses of the genus Orthopoxvirus. It is discussed whether orthopoxviruses of animals may then constitute a potential danger to man, with respect to their mutagenic and adaptation capabilities as well as their possible genetic interactions. Infection of man with these viruses is generally possible, but at present, not of acute importance. Monkeypox and cowpox viruses, as well as their variants occurring in carnivores and rodents, are of primary interest for individuals who have not b

Both generic and environmental factors are likely to contribute to the pathogenesis of neurodevelopmental disorders. Even in heritable disorders of high penetrance, variability in timing of onset or severity of disease indicate a role for modifying principles. Investigation in animal models of the consequences of interactions between host response genes and microbes, toxins, and other environmental agents in a temporal context may elucidate the pathophysiology of a wide spectrum of chronic diseases. Here we review the evidence that infectious and immune factors may contribute to the pathogenes

The growing comorbidity between cardiovascular and neuropsychiatric disorders presents a major global health challenge. This reciprocal interaction worsens patient prognosis and hinders the diagnosis and treatment of these comorbid conditions. In recent years, the concept of the "gut-heart-brain axis" has highlighted the central pathogenic and therapeutic role of the gut microbiota in linking these distal systems. This review therefore aims to clarify this axis and to explore how specific gut microbiota metabolites modulate this bidirectional physiological interaction. We have integrated recen

Mineral/mineral interactions at the intestinal level are important in animal nutrition and toxicology, but only limited understanding of their extent or importance in humans has been developed. An inhibitory interaction of dietary tin on zinc retention has been recently described from human metabolic studies. We have explored the tin/zinc interaction using the change-in-plasma-zinc-concentration method with a standard dosage of 12.5 mg of zinc as zinc sulfate in 100 ml of Coca-Cola. Sn/Zn ratios of 2:1, 4:1, and 8:1, constituted by addition of 25, 50, and 100 mg of tin as stannous chloride, ha

Individuals with a SHANK3-related neurodevelopmental disorder, also termed Phelan-McDermid syndrome or abbreviated as PMS, exhibit significant global developmental delay, language impairment, and muscular hypotonia. Also common are repetitive behaviors and altered social interactions, in line with a diagnosis of autism spectrum disorders. This study investigated the developmental aspect of autism-related behaviors and other phenotypes in a Shank3-transgenic mouse model. The animals underwent two sets of identical behavioral experiments, spanning motor skills, social and repetitive behavior, an

The experimental rodent models for the fungal disease are a handy tool for understanding host-fungus interactions. To Fonsecaea sp., one of the causative agents of chromoblastomycosis, there is an extra challenge because the animals preferably used show a spontaneous cure; so until now, there is no model to reproduce the long-term disease similar to human chronic disease. In this chapter, we described an experimental model using rats and mice with a subcutaneous route, with the checkpoints of acute-like and chronic-like lesion analysis comparable with human lesions, the fungal burden, and the

Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) is one of the fastest-growing hepatic disorders worldwide. During its natural course, MASLD tends to progress from isolated steatosis of the liver to Metabolic Dysfunction-Associated Alcoholic Steatohepatitis (MASH), advanced fibrosis, and finally cirrhosis, with the risk of developing hepatocellular carcinoma (HCC). Although frequently related to overweight or obesity and other components of the metabolic syndrome (MS), MASLD can also be present in individuals without such risk factors. The mechanisms leading to MASLD are incomp

Malaria is caused by apicomplexan parasites of the Plasmodium genus. Plasmodium chabaudi is an excellent animal model for the study of human malaria caused by P. falciparum. Merozoites invade erythrocytes but are also found in other host cells including macrophages from the spleen and liver. Methodologies for obtaining merozoites usually involve treatment with protease inhibitors. However, merozoites obtained in this way may have their enzymatic profile altered and, therefore, are not ideal for cell-interaction assays. We report the obtainment of P. chabaudi merozoites naturally egressed from

Sporotrichosis is a worldwide distributed subcutaneous mycosis that affects mammals, including human beings. The infection is caused by members of the Sporothrix pathogenic clade, which includes Sporothrix schenckii, Sporothrix brasiliensis, and Sporothrix globosa. The fungus can be acquired through traumatic inoculation of conidia growing in vegetal debris or by zoonotic transmission from sick animals. Although is not considered a life-threatening disease, it is an emergent health problem that affects mostly immunocompromised patients. The sporotrichosis causative agents differ in their virul

Leptospirosis is a widespread zoonotic disease caused by pathogenic Leptospira spp., affecting humans, domestic animals, and wildlife. Wild mammals are recognized as potential maintenance hosts for pathogenic Leptospira spp., and some species may act as asymptomatic reservoirs, representing a potential public health risk, especially in areas with increasing human-wildlife interaction. This cross-sectional study investigated the occurrence of pathogenic Leptospira spp. in wild mammals from Paraíba, Brazil, sampled between June 2022 and June 2023 using serological and molecular approaches. To determine the state of the research on ototoxic properties of Pb, evaluate possible synergistic effects with concurrent noise exposure, and identify opportunities to improve future research, we performed a review of the peer-reviewed literature to identify studies examining auditory damage due to Pb over the past 50 years. Thirty-eight studies (14 animal and 24 human) were reviewed. Of these, 24 suggested potential ototoxicity due to Pb exposure, while 14 found no evidence of ototoxicity. More animal studies are needed, especially those investigating Pb exposure levels that are occupational

With more than 71 million chronically infected people, the hepatitis C virus (HCV) is a major global health concern. Although new direct acting antivirals have significantly improved the rate of HCV cure, high therapy cost, potential emergence of drug-resistant viral variants, and unavailability of a protective vaccine represent challenges for complete HCV eradication. Relevant animal models are required, and additional development remains necessary, to effectively study HCV biology, virus-host interactions and for the evaluation of new antiviral approaches and prophylactic vaccines. INTRODUCTION: Narcolepsy is a debilitating sleep disorder characterized by excessive daytime sleepiness, cataplexy and intrusive REM sleep. Deficits in endogenous orexins are a major pathogenic component of the disease. This disorder is also associated with the gene marker, HLADQB1*0602. An increased prevalence of sleep apnea in narcolepsy suggested interactions among ventilatory chemosensitivity, narcolepsy-cataplexy, and sleep apnea.
RESULTS: Evidence from animal studies using orexin knockout mice and focal microdialysis of an orexin receptor antagonist demostrated that orexins are also con

Vector-borne pathogens cause enormous suffering to humans and animals. Many are expanding their range into new areas. Dengue, West Nile and Chikungunya have recently caused substantial human epidemics. Arthropod-borne animal diseases like Bluetongue, Rift Valley fever and African horse sickness pose substantial threats to livestock economies around the world. Climate change can impact the vector-borne disease epidemiology. Changes in climate will influence arthropod vectors, their life cycles and life histories, resulting in changes in both vector and pathogen distribution and changes in the a
