Type I interferons and IL-12: convergence and cross-regulation among mediators of cellular immunity.

Therapeutic use of type I IFN (IFN-alpha/beta) has become common. Many of the diverse diseases targeted are marked by pathogenetic abnormalities in cell-mediated immunity (CMI), these cellular immune responses either causing injury to the host, lacking sufficient vigor for virus or tumor clearance, or both. In general, therapeutic efficacy is limited. It is thus notable that the pleiotropic effects of type I IFN on CMI remain poorly understood. We characterized the effects of type I IFN on the production of IL-12, the central immunoregulatory cytokine of the CD4(+) T cell arm of CMI. We show t

5'-triphosphate-siRNA activates RIG-I-dependent type I interferon production and enhances inhibition of hepatitis B virus replication in HepG2.2.15 cells.

Hepatitis B virus (HBV) infection often results in acute or chronic viral hepatitis and other liver diseases including cirrhosis and hepatocellular carcinoma. Current therapies for HBV usually have severe side effects and can cause development of drug-resistant mutants. An alternative and safe immunotherapeutic approach for HBV infection is urgently needed for effective anti-HBV therapy. In this study, we propose a new strategy for anti-HBV therapy that activates type-I interferon (IFN) antiviral innate immunity through stimulating pattern-recognition receptors with RNA interference (RNAi) usi

Suppression of Type I Interferon Production by Human T-Cell Leukemia Virus Type 1 Oncoprotein Tax through Inhibition of IRF3 Phosphorylation.

UNLABELLED: Infection with human T-cell leukemia virus type 1 (HTLV-1) is associated with adult T-cell leukemia (ATL) and tropical spastic paraparesis. Type I interferons (IFNs) are key effectors of the innate antiviral response, and IFN-α combined with the nucleoside reverse transcriptase inhibitor zidovudine is considered the standard first-line therapy for ATL. HTLV-1 oncoprotein Tax is known to suppress innate IFN production and response but the underlying mechanisms remain to be fully established. In this study, we report on the suppression of type I IFN production by HTLV-1 Tax through i

The Lyme disease spirochete Borrelia burgdorferi utilizes multiple ligands, including RNA, for interferon regulatory factor 3-dependent induction of type I interferon-responsive genes.

We recently discovered a critical role for type I interferon (IFN) in the development of murine Lyme arthritis. Borrelia burgdorferi-mediated induction of IFN-responsive genes by bone marrow-derived macrophages (BMDMs) was dependent upon a functional type I IFN receptor but independent of Toll-like receptor 2 (TLR2), TLR4, TLR9, and the adapter molecule MyD88. We now demonstrate that induction of the IFN transcriptional profile in B. burgdorferi-stimulated BMDMs occurs independently of the adapter TRIF and of the cytoplasmic sensor NOD2. In contrast, B. burgdorferi-induced transcription of the

Immune horses rapidly increase antileukoproteinase and lack type I interferon secretion during mucosal innate immune responses against equine herpesvirus type 1.

UNLABELLED: Equine herpesvirus type 1 (EHV-1) is one of the most prevalent respiratory pathogens in horses with a high impact on animal health worldwide. Entry of the virus into epithelial cells of the upper respiratory tract and rapid local viral replication is followed by infection of local lymphoid tissues leading to cell-associated viremia and disease progression. Pre-existing mucosal immunity has previously been shown to reduce viral shedding and prevent viremia, consequently limiting severe disease manifestations. Here, nasopharyngeal transcriptomic profiling was used to identify differe

PARP inhibitor YCH1899 activates type I interferon signaling and synergizes with STING agonists in BRCA-proficient tumors.

Poly(ADP-ribose) polymerase (PARP) inhibitors are highly effective in BRCA-deficient tumors, yet their therapeutic benefit in BRCA-proficient cancers remains limited. Here, we demonstrate that YCH1899, a next-generation, resistance-overcoming PARP inhibitor, elicits significantly stronger type I interferon signaling than currently approved PARP inhibitors in BRCA-proficient tumor cells. Mechanistically, YCH1899 facilitates the accumulation of cytosolic DNA, thereby robustly activating the cGAS-STING-IRF3 pathway. Consequently, YCH1899 exhibits potent inhibitory effects on the growth of various

Increased Type I Interferon Activity with Concurrent Plasmablast Expansion Identifies Systemic Lupus Erythematosus Patients with Poor Outcomes.

Recent evidence suggests that type I interferon (IFN) activity has prognostic relevance in systemic lupus erythematosus (SLE). This study investigated whether combining IFN activity with elevated peripheral blood plasmablast (PB) levels-another key feature of lupus pathophysiology-improves risk stratification for poor clinical outcomes. Clinical data were prospectively collected at a single lupus center. Flow cytometry was performed on freshly isolated peripheral blood mononuclear cells to investigate Sialic acid-binding Immunoglobulin-like Lectin 1 (SIGLEC-1) as a surrogate marker of IFN acti

Identification of Histone Deacetylase Inhibitor Targeting Type I Interferon and B Cell Abnormalities in Systemic Lupus Erythematosus.

OBJECTIVE: Systemic lupus erythematosus (SLE) is characterized by increased Type I interferon (IFN-I) and autoantibody production. This study aimed to identify drugs that can inhibit both IFN-I and autoantibody production. METHODS: We identified an inhibitor of IFN-I production from a chemical library. Subsequently, we examined its efficacy and underlying mechanisms in suppressing the expression and phosphorylation of upstream signaling molecules for IFN-I production and the differentiation of B cells into plasma cells. Additionally, we examined whether it could alleviate disease severity in

SIRT1-mediated deacetylation of TRIM21 promotes IRF5 degradation and restrains type I interferon signatures in Sjögren's disease.

INTRODUCTION: Persistently activated type I interferon (IFN) signaling is a key factor in glandular dysfunction of Sjögren's disease(SjD), with elevated "type I interferon signatures" observed in both peripheral blood and salivary gland tissues of affected patients. However, the intrinsic mechanisms suppressing this response remain incompletely understood. This study aims to investigate whether the NAD⁺-dependent deacetylase SIRT1 regulates the type I IFN signatures in salivary gland tissue by enhancing the ubiquitin-mediated degradation of IRF5 through modulation of TRIM21's E3 ubiquitin liga

DHX34 deficiency triggers tumor-intrinsic immunity via a dsRNA-mediated type I interferon pathway activation in HCC.

Tumors often evade immune surveillance by crippling their immunogenicity in the microenvironment. DHX34, an RNA helicase involved in nonsense-mediated mRNA decay pathway, is critical for aberrant RNA degradation. However, the effect of DHX34 in regulating the immunogenicity in hepatocellular carcinoma (HCC) is still unclear. Here, a surprising function of DHX34 in inhibited HCC immunogenicity is identified. DHX34-deficient tumors were infiltrated by activated T cells that impaired tumor growth and enhanced survival in mice. Mechanistically, DHX34 depletion triggered dsRNA accumulation which ma

Type I interferon protects against bone loss in periodontitis by mitigating an interleukin (IL)-17-neutrophil axis.

Type I interferons (IFNs-I), a group of pleiotropic cytokines, critically modulate host response in various inflammatory diseases. However, the role of the IFN-I pathway in periodontitis remains largely unknown. In this report, we describe that the IFN-β levels in the gingival crevicular fluid of human subjects were negatively associated with periodontitis and clinical gingival inflammation. Disruption of IFN-I signaling worsened alveolar bone resorption in a ligature-induced periodontitis murine model. Deficiency of the IFN-I pathway resulted in an exaggerated inflammatory response in myeloid

Myeloid cell-specific type I interferon signaling mediates age-dependent inflammation and protection in Bordetella pertussis infection.

Type I interferons (IFNs) play complex roles during bacterial infections. We previously found that type I IFNs were induced in Bordetella pertussis-infected adult mice but not in infant mice, a potentially relevant clinical dichotomy, since pertussis can be fatal in human infants. We investigated the role of type I IFNs and their cross-regulation with type III IFNs (IFN-λ) in B. pertussis infection across developmental stages. In contrast to global IFNAR1 knockout adult mice, in which lung inflammation was equivalent to that in wild-type mice, myeloid cell-specific deficiency of the type I IFN

Implications of serum calcitonin gene-related peptide and type I interferon in systemic lupus erythematosus-associated headaches.

ObjectiveHeadache is a common neurological symptom in patients with systemic lupus erythematosus (SLE). Calcitonin gene-related peptide (CGRP) is a potential chemical mediator of headaches, while type I interferon (IFN) plays a pivotal role in the immune system of SLE. This study investigated the implications of CGRP and type I IFNs as headache biomarkers in patients with SLE.MethodsWe used clinical information and serum samples from 144 patients with SLE from a Japanese multicenter cohort and a biobank. Serum CGRP, IFN-α, and IFN-β levels, which were measured using enzyme-linked immunoassay,

The ubiquitin E3 ligase TRIM21 suppresses type I interferon signaling via STING degradation and ameliorates systemic autoimmunity.

Tripartite motif-containing 21 (TRIM21) is a cytoplasmic protein with E3 ubiquitin ligase activity. Although autoantibodies against TRIM21 are frequently detected in patients with systemic lupus erythematosus (SLE), its role in disease pathogenesis remains unclear. Here we demonstrate that TRIM21 directly interacts with the stimulator of interferon genes (STING) to regulate type I interferon (IFN) production. In both induced and spontaneous murine models of lupus, TRIM21 deficiency exacerbated lupus-like pathology and heightened IFN production after STING activation. By contrast, TRIM21 overex

Type I interferon as a pre-lupus marker in anti-nuclear antibody-associated childhood immune thrombocytopenic purpura.

Paediatric-onset anti-nuclear antibody-associated immune thrombocytopenic purpura (ITP-ANA+) is a pre-lupus condition. Interferon signature (IS) is a reliable method to measure interferon-stimulated gene expression which is commonly raised in systemic lupus erythematosus (SLE) and in adult pre-lupus cases. Between 2022 and 2024, IS analysis was performed on 61 children, 17 with ITP-ANA+, 15 with ITP-ANA-, 15 with SLE and 14 with juvenile idiopathic arthritis. IS was positive in 14/17 children (82%), with a median score of 17 (1.2-143). This median IS was significantly higher than in the ITP-AN

Working in negative space: Type I interferon mediated immuno-modulation through transcriptional suppression in disease and homeostasis.

The type I interferon family of cytokines are rapidly produced following innate pattern recognition receptor engagement and establish a critical early state of host defense. Type I interferons act in antiviral immunity as transcriptional activators and the binding of any type I interferon to the common IFNAR receptor triggers the transcription of Interferon Stimulated Genes (ISGs). A defined set of ISGs have been described through exhaustive studies and the protein products of these ISGs function to increase cell intrinsic resistance to viral growth and to promote viral clearance. Unleashed monocytic engagement in Sézary syndrome during the combination of anti-CCR4 antibody with type I interferon.

Sézary syndrome (SS) is an aggressive leukemic expansion of skin-derived malignant CD4+ T cells. Drug monotherapy often results in disease relapse because of the heterogenous nature of malignant CD4+ T cells, but how therapies can be optimally combined remains unclear because of limitations in understanding the disease pathogenesis. We identified immunologic transitions that interlink mycosis fungoides with SS using single-cell transcriptome analysis in parallel with high-throughput T-cell receptor sequencing. Nascent peripheral CD4+ T cells acquired a distinct profile of transcription factors

The antiviral activity of myricetin against pseudorabies virus through regulation of the type I interferon signaling pathway.

UNLABELLED: The type I interferon signaling pathway constitutes a pivotal component of the innate immune response, encompassing the cGAS/STING and JAK/STAT pathways. Drugs that affect the body's innate immune response could potentially be used as broad-spectrum antivirals. In this study, the antiviral activities of 25 flavonoids against pseudorabies virus (PRV) were tested in PK-15 cells. Eight active flavonoids were identified, with IC50 values ranging from 23.24 to 323.09 µM. Subsequently, the regulatory effects of these flavonoids on the cGAS/STING pathway in PRV-infected cells were investi

African swine fever virus B175L inhibits the type I interferon pathway by targeting STING and 2'3'-cGAMP.

African swine fever virus (ASFV), the only known DNA arbovirus, is the causative agent of African swine fever (ASF), an acutely contagious disease in pigs. ASF has recently become a crisis in the pig industry in recent years, but there are no commercially available vaccines. Studying the immune evasion mechanisms of ASFV proteins is important for the understanding the pathogenesis of ASFV and essential information for the development of an effective live-attenuated ASFV vaccines. Here, we identified ASFV B175L, previously uncharacterized proteins that inhibit type I interferon signaling by tar
