We report on patients with STAT3 loss-of-function (LOF) mutations who developed lupus-like autoimmunity. Immune dysregulation seen in STAT3 LOF patients suggests a susceptibility to systemic autoimmunity with important implications in monitoring and management of these patients.

We demonstrate that autosomal-dominant STAT3-deficient hyper-IgE syndrome confers late-onset susceptibility to molds, especially Aspergillus, despite preserved myeloid functions, in association with previous parenchymal lung damage (bronchiectasis/pneumatoceles). This suggests a critical role for non-hematopoietic processes in innate antifungal immunity in AD-HIES.

A 4-month-old baby with a family history of hyper-IgE syndrome acquired Pneumocystis jirovecii pneumonia. The patient's hyper-IgE syndrome score was low, but a genetic study yielded a STAT3 mutation. P. jirovecii pneumonia can be added to the infections associated with hyper-IgE syndrome. In some cases, it may be the presenting manifestation of this immunodeficiency.

Data from several recent studies is presented showing that Fetal Alcohol Syndrome (FAS) has long term consequences. Effects on the motor function and social life of the patients after Guillain-Barré syndrome (GBS) were studied. A letter of inquiry was sent to the patients in November 2001; 98 patients responded validly. The questionnaire had questions with 3 possible answers related to situations at work, at home, and about leisure activities. The Hughes functional grade, which evaluates physical conditions, and arm disability scale scores were assessed.
