RXRα suppression drives hepatic metabolic and immune dysfunction in sepsis.

Sepsis is a life-threatening condition in which a dysregulated host response to infection leads to organ dysfunction and metabolic and immune failure. We identify hepatocyte retinoid X receptor α (RXRα) as a key integrator of host resilience during polymicrobial sepsis. RXRα is transcriptionally regulated by hepatocyte nuclear factor 4α (HNF4α), and sepsis rapidly decreases RXRα mRNA and protein levels. Transcriptomic analyses show that the septic liver becomes partially resistant to pharmacological activation of RXRα with bexarotene. Prophylactic- but not therapeutic- bexarotene improves surv

Light- and electron microscopic studies of the liver in "bronze baby" syndrome.

The light- and electron-microscopic changes of the hepatic tissue of a case with the "bronze baby" syndrome were described. The light-microscopic examinations revealed intrahepatic cholestasis of the hepatocanalicular type associated with portal inflammation. The electron-microscopic examinations showed dense deposits in the hepatic cells, Kupffer cells and in the bile canaliculi. The bile canaliculi were distended, the canalicular membrane severely destructed. The patient included in the study has been free of symptoms for 2 years.

Tissue-resident macrophage and dendritic cells drive type I IFN immunity to enteroviruses in the liver.

Enteroviruses are major causes of neonatal morbidity and mortality, with echovirus infections commonly associated with severe disease, inclu … iruses and to identify the specific cells producing IFNs in response. We found that hepatocytes and Kupffer cells were the most heavily infected cell types. In contrast, early and robust type I IFN responses were primarily driven by Kupffer cells and a subset of dendritic cells. To determine whether type I IFNs act directly on hepatocytes to mediate protection, …

Spatial and Single-Cell Transcriptomics Reveals the Regional Division of the Spatial Structure of MASH Fibrosis.

OBJECTIVE: To elucidate the regional distribution of metabolic dysfunction-associated steatohepatitis (MASH) fibrosis within the liver and t … the single-cell-sequencing data set (GSE189175) showed a greater proportion of inflammatory cells (Kupffer cells and T cells) and myofibroblasts in MASH. Six genes, showing high- or low-specific expression in Cluster 9, namely, ADAMTSL2, PTGDS, S100A6, PPP1R1A, ASS1 and G6PC, were identified in combination with pathology. The average expression levels of ADAMT …

Low-level laser therapy ameliorates CCl4-induced liver cirrhosis in rats.

This study investigated the effects of low-level laser therapy (LLLT) in the liver function, structure and inflammation in a experimental mo … umulation and the hepatic inflammatory infiltrate. Of note, LLLT reduced CCl(4)-increased number of Kupffer cells (P < 0.05) and hepatic stellate cells (P < 0.05). We conclude that LLLT presents beneficial effects on liver function and structure in an experimental model of CCl(4)-induced cirrhosis.

Warm liver ischemia in experiment and lysosomal markers.

BACKGROUND: The aim of the study is to perform a morphological analysis of certain lysosomal enzymes and parenchymal alterations during warm ischemia in the pig liver. METHODS: Standard hepatectomies were performed in a set of 24 pigs. Intra-operative intravenous (portal vein) Pentoxiphylline and hydroxyl radical scavenger Stobadine was administered. Tissue specimens were removed from the margo acutum in 10 minutes interval. RESULT: In normal pig liver, the acid phosphatase (ACP) activity is in not numerous Kupffer cells and on the biliary pole of hepatocytes, diffusely in the whole parenchy…

Necroinflammatory changes in hepatic lobules in primary biliary cirrhosis with less well-defined cholestatic changes.

Primary biliary cirrhosis (PBC) is characterized by nonsuppurative inflammation and destruction of interlobular bile ducts. In this study we … wed perivenular zonal necrosis and central to central bridging necrosis. Focal necroses and swollen Kupffer cells were associated with lymphocytes with plump eosinophilic cytoplasm, resembling epithelioid cells. Well-developed and less-defined epithelioid cell granuloma also was occasionally found. The size, number, and distribution of these intralobular necroi …

Balancing immune evasion and hepatocyte delivery: a hybrid liposomal platform for enhanced nanotherapy in non-alcoholic fatty liver disease.

The clinical translation of nanomedicine for non-alcoholic fatty liver disease (NAFLD) is frequently hindered by rapid hepatic clearance mediated by Kupffer cells, which are the predominant phagocytic macrophages in the liver. Conventional stealth strategies, such as PEGylation, protect host cells from phagocytosis; however, their exploitation in drug delivery is limited by the prevailing assumption that immune evasion impairs cellular uptake. Our previous study demonstrated that liposomes functionalized with a CD47-derived self-peptide (SLip) effectively evaded macrophage clearance while reta…

High Kupffer cells interaction reduces the nanomedicine efficacy. During fibrosis, the extracellular matrix accumulation in the perisinusoidal space restricts nanoparticle delivery to hepatocytes. Long-term retention of gold nanoparticles in the liver is not affected by their physicochemical characteristics.

Gold nanoparticles (GNPs) are considered promising candidates for healthcare applications, however, their toxicity after long-term exposure to the material remains uncertain. Since the liver is the main filter organ for nanomaterials, this work was aimed at evaluating hepatic accumulation, internalisation and overall safety of well-characterised and endotoxin-free GNPs in healthy mice from 15 minutes to 7 weeks after a single administration. Our data demonstrate that GNPs were rapidly segregated into lysosomes of endothelial cells (LSEC) or Kupffer cells regardless of coating or shape but with…

Hepatic F4/80+ CD11b+ CD68- cells influence the antibacterial response in irradiated mice with sepsis by Enterococcus faecalis.

Gut-associated sepsis is a major problem in patients undergoing abdominal radiation therapy; the majority of pathogens causing this type of sepsis are translocated from the gut microbiota. While treating sepsis, bacterial clearance must be achieved to ensure patient survival, and the hepatic immune response is responsible for this process. In particular, Kupffer cells play a crucial role in the hepatic immune response against infectious agents. Recently, two populations of Kupffer cells have been described: liver-resident macrophages (Mϕ) (F4/80+ CD11b- CD68+ cells) and hepatic Mϕ derived from…

The Histopathologic Features of Sickle Cell Hepatopathy: A Multi-Institutional Study.

OBJECTIVES: Recent data on hepatic histopathology in patients with sickle cell disease (SCD) are lacking. METHODS: A total of 39 liver biopsies from SCD patients from 4 medical institutes were systematically evaluated. RESULTS: The average age of patients was 27 years; 23 were female. The majority of the patients had hemoglobin SS (33), 3 had hemoglobin SC, and 3 sickle cell trait. Elevated liver functional tests and evaluation for cirrhosis were the main indications for biopsy. At the time of biopsy, most had elevated liver transaminases or hepatomegaly. The most common histopathologic abno

Endotoxemia, pentose cycle, and the oxidant/antioxidant balance in the hepatic sinusoid.

During the innate immune response, excessive release of reactive oxygen species (ROS) from sequestered phagocytes and activated resident mac … ell type depends on the accompanying adaptive changes in ROS-metabolizing enzymes. In LPS-activated Kupffer cells, the elevated expression of glucose transporter GLUT1 and G6PD mainly serves primed production of superoxide anion, hydrogen peroxide, and nitric oxide.
