[Serum selectin E level in lead-exposed workers].

Lead is recognised as a potential atherogenic factor. One of the earliest events in the development of atherosclerosis is monicyte attachment to the endothelial surface. This is followed by recruitment of monocytes into the subendothelial space and ingestion of modified LDL by these cells. In turn, modified LDL stimulates endothelial cells to induce expression of proinflammatory adhesion molecules, such as selectins, which further promote monocyte migration. It was observed that atherosclerotic vascular damage is associated with increased level of circulating selectin E. The aim of this study …

Dietary exposure of children in lead-laden environments.

Children are the most susceptible population to lead exposure because of three interacting factors; they have more opportunity for contact with lead sources due to their activities, lead absorption occurs more readily in a child compared to an adult, and the child's development is more vulnerable to lead than adults. Low levels of lead in the blood have been shown to cause adverse health effects; the level of concern for children is currently 10 microg/dl. The contribution of dietary exposure of lead to increased blood lead levels (PbB) is not well characterized. This study was conducted to me…

State of pregnancy modifies lead toxicity in mice.

Toxicity of lead acetate after administration through the oral route at 0-50 mg/kg body weight of animal has been assessed in the liver of pregnant mice and compared with the effect in the liver of nonpregnant dams. Analysis showed that the basal level of hepatic lead is considerably reduced during pregnancy as compared to that in nonpregnant state. After administration of Pb-acetate, deposited lead in liver of nonpregnant mice was 3- to 4-fold while in pregnant mice was, it was 1.8- to 3.0-fold over their respective control values. Although hepatic Fe, Cu, and Zn levels had a tendency to be l…

Subclavian venogram as a guide to lead implantation.

Recent reports have raised doubts regarding the safety and efficacy of the blind subclavian venipuncture technique for intracardiac lead implantation. To permit a more lateral entry, we used a simple subclavian venogram performed through the brachial vein of the ipsilateral arm of 22 consecutive unselected patients undergoing lead implantation (19 permanent pacemakers and 3 intracardiac defibrillators). A total of 35 leads were implanted (31 left pectoral and 4 right pectoral). Lead insertion by venogram technique was used successfully in all patients. Two inconsequential arterial punctures oc…

Lead--the toxic metal to stay with human.

Lead has been known to be toxic to most living things at high dose. It is found naturally in earth and present in almost all parts of the environment, such as foods, air, water, dust, soil, paint, and tissues of living organisms including human. This metal is being used in various aspects including the manufacturing of storage batteries, production of chemicals, paints and gasoline additives. It is also used to make various metal products, e.g. sheet lead, solder, and pipes. Human exposure to lead is mainly from foods and other environments. However, it is expected that exposure to environment…

Update: blood lead levels--United States, 1991-1994.

Lead is an environmental toxicant that may deleteriously affect the nervous, hematopoietic, endocrine, renal, and reproductive systems. Lead exposure in young children is a particular hazard because children absorb lead more readily than do adults and because the developing nervous systems of children are more susceptible to the effects of lead. Blood lead levels (BLLs) at least as low as 10 micrograms/dL can adversely affect the behavior and development of children. CDC's National Health and Nutrition Examination surveys (NHANES), an ongoing series of national examinations of the health and n…

Renal effects of environmental and occupational lead exposure.

Environmental and industrial lead exposures continue to pose major public health problems in children and in adults. Acute exposure to high concentrations of lead can result in proximal tubular damage with characteristic histologic features and manifested by glycosuria and aminoaciduria. Chronic occupational exposure to lead, or consumption of illicit alcohol adulterated with lead, has also been linked to a high incidence of renal dysfunction, which is characterized by glomerular and tubulointerstitial changes resulting in chronic renal failure, hypertension, hyperuricemia, and gout. A high in…

Umbilical cord blood lead levels in Shanghai, China.

This study was designed to determine the cord blood lead (BPb) levels of babies born in one urban area of Shanghai, and to preliminarily identify the demographic, social environment and prenatal factors which have an effect on the cord BPb concentrations. From August to November 1993, umbilical cord blood samples were obtained from 605 live newborns in the Yangpu Maternal and Child Hospital. 257 samples were excluded from measurement because of clotting. In 348 cord samples, the geometric mean of cord BPb levels was 9.2 micrograms/dl, with a 95% confidence interval of the mean 8.86-9.54 (micro…

Parental occupational lead exposure and low birth weight.

This study suggests that paternal occupational lead exposure may be associated with low birth weight in the offspring. The odds of low birth weight rose fivefold among infants of fathers who were potentially exposed to high levels of lead during the period 6 months before pregnancy to the end of pregnancy (adjusted odds ratio (aOR) = 4.7, 95% confidence interval (CI) = 1.1-20). This effect was most prominent for low-birth-weight infants who were both preterm and small for gestational age. There was a suggestion of a gradual increase of the odds of low birth weight at medium levels of exposure,…

Pharmacological prevention of acute lead poisoning in Paramecium.

To understand how lead (Pb2+) and other metals and chelating agents effect living cells, behavioral experiments in the marine ciliate Paramecium calkinsi were carried out. The duration of Backward Swimming Behavior (BSB) of Paramecium was partially reduced when cells were exposed to 100 microM of Ni2+, CD2+ and Co2+. In contrast, Pb2+ increased Paramecium BSB in a dose-dependent manner. Thus, 1, 10, 20, 50 and 100 microM of Pb2+ increased the duration of BSB by 20.4, 83.9, 143.2, 163.2 and 185.2%, respectively. The naphthalenesulphonamide W-7, a calcium channel blocker in lower organisms, abol…

Lead and the kidney: nephropathy, hypertension, and gout.

Lead intoxication in human beings has been documented since the second century B.C. Renal disease, hypertension, and gout have all been linked to lead by strong circumstantial evidence. Both acute and chronic nephropathy can occur as a result of lead poisoning. Acute renal failure develops following acute lead intoxication and is often associated with gastrointestinal, neurologic, and hematologic disorders. Both blood and urinary laboratory abnormalities are associated with acute intoxication and are often diagnostic. Chronic lead nephropathy, a chronic tubulointerstitial nephritis on biopsy, …

Lead increases aldosterone production by rat adrenal cells.

Exposure to lead has been postulated to contribute to elevated blood pressure in humans and has been shown to raise blood pressure in animals. The mechanism of action of lead on blood pressure is unknown. We fed lead to rats in their drinking water and then examined the production of aldosterone by their adrenal cells in vitro. We also measured excretion of aldosterone and corticosterone by intact rats stimulated with corticotropin, with and without lead treatment. At a dose (273 ppm) that raised blood levels to 30 to 40 micrograms/dL, comparable to blood levels in exposed humans, lead induced…

Spinal cord uptake of lead injected into muscle.

It has been hypothesised that some motor neuron diseases may result from lead (Pb) being retrogradely transported in axons from muscle to spinal cord. To test this hypothesis, we injected mice in the right hindlimb with 0.1 ml of 0.25% or 2.5% Pb. In some animals the sciatic nerve was cut before injection. Pb levels were measured in the lumbar and thoracic spinal cord and the kidney with atomic absorption spectrophotometry (AAS). Four weeks after injection, Pb was found in the spinal cord on AAS, with higher levels in the lumbar than the thoracic cord only in the high-Pb group. Sciatic nerve c…

Role of lead-binding proteins in renal cancer.

High-dose lead exposure in rodents has been shown to produce pathognomonic lead intranuclear inclusion bodies and to result in an increased incidence of renal adenocarcinomas. Studies from this laboratory and others have demonstrated the presence of high-affinity renal lead-binding proteins in rat kidneys which act as tissue sinks for lead at low dose levels. Cell-free nuclear translocation studies have shown that these molecules are capable of facilitating the intranuclear movement of lead and that they are associated with chromatin. These data suggest that renal lead-binding proteins may pla…

Anatomical and morphological evaluation of pacemaker lead compression.

In recent years, pacemaker lead failure due to compressive damage has been reported with increasing frequency. To document the mechanism of this failure, we evaluated explanted mechanically damaged leads with electrical testing, optical microscopy, and in some cases, scanning electron microscopy (SEM). In addition, we performed an autopsy study to measure the compressive loads on catheters placed percutaneously through the costoclavicular angle, as well as by cephalic cutdown. Of the 49 explanted compression damaged leads with enough clinical data for analysis, all had been placed by percutane…
