[Effects of melatonin on circadian time keeping system].

This paper is intended to provide an overview of a physiological role of melatonin (MLT) in sleep-wake regulation. In human MLT studies, MLT has significantly increased sleepiness and fatigue during the daytime but not at night, which indicates time-dependent sleep-promoting effect of MLT. Op the other hand, MLT is known to have phase-shifting effect of biological clock. Phase response curve (PRC) for MLT administration was introduced in human studies. Successful melatonin treatments for circadian rhythm sleep-disorders based on PRC have been reported. MLT is known to decrease core body temper…

Modulation of plasma melatonin concentrations by changes in posture.

Posture change from a lying position to a standing position results in a decrease in plasma volume, which leads to an increase in plasma constituents, especially that of proteins and blood constituents bound to them. The aim of the present study was to investigate the physiological effects of postural changes on plasma nocturnal melatonin concentrations in healthy human volunteers. The study was divided into four stages. During stage one, subjects were seated from 21.00 hr to 01.00 hr. In stage two, subjects were lying at ground level from 21.00 hr to 01.00 hr. In stage three, subjects were is…

[Development of animal models for the chronobiotics of melatonin analogs].

Melatonin is a neurohormone secreted by the pineal gland (or epiphysis). It is secreted only during the night whether in man or in diurnal or nocturnal animals. Melatonin secretion is regulated by light, which inhibits or synthesizes it. By a 'feedback' mechanism, melatonin can regulate the suprachiasmatic nuclei which function like internal clocks at the level of melatonin receptors. Specific melatonin agonists are active in different animal models representative of sleep disturbances due to alterations of circadian rhythms.

Pharmacological actions of melatonin in oxygen radical pathophysiology.

Melatonin, the chief secretory product of the pineal gland, was recently found to be a free radical scavenger and antioxidant. This review briefly summarizes the published reports supporting this conclusion. Melatonin is believed to work via electron donation to directly detoxify free radicals such as the highly toxic hydroxyl radical. Additionally, in both in vitro and in vivo experiments, melatonin has been found to protect cells, tissues and organs against oxidative damage induced by a variety of free radical generating agents and processes, e.g., the carcinogen safrole, lipopolysaccharide,…

Control of the circannual rhythm of reproduction by melatonin in the ewe.

Annual variations in day length are responsible for seasonal changes in reproductive activity in sheep. However, in constant photoperiodic c … d through neural processing into an endocrine signal by the pineal gland: the nocturnal increase in melatonin release. Recent studies strongly suggest that melatonin has a hypothalamic target to modulate the reproductive neuroendocrine axis. Most LHRH perikarya are located in the preoptic area, but this region is devoid of melatonin receptors, and microimpl …

Melatonin: generation and modulation of avian circadian rhythms.

The pineal organ and its hormone melatonin are significant components of avian circadian pacemaking systems. In songbirds, pinealectomy results in the abolition or destabilization of overt circadian rhythms such as the rhythm of locomotor activity, feeding, or body temperature. A stable rhythmicity can be restored either by reimplanting a pineal organ, by periodic injections or infusions of melatonin, or by applying melatonin rhythmically through the drinking water. Several results suggest that the pineal melatonin rhythm acts on at least one other oscillator within the circadian pacemaking sy…

Ocular melatonin rhythms in the goldfish, Carassius auratus.

Ocular melatonin rhythms in the goldfish were studied and compared to those in the pineal organ and plasma. Under light:dark (LD) of 12 h light:12 h dark, melatonin contents in the eye as well as the pineal organ and plasma exhibited clear day-night changes with higher levels at mid-dark than at mid-light. However, melatonin contents in the eye at mid-light and mid-dark were approximately 100 and 9 times greater than those in the pineal organ, respectively. Day-night changes of ocular melatonin persisted after pinealectomy, which abolished those in plasma melatonin under LD 12:12. Ocular melat…

Melatonin site and mechanism of action: single or multiple?

By affecting the entrainment pathways of the biologic clock, melatonin has a major influence on the circadian and seasonal organization of vertebrates. In addition, a number of versatile functions that far transcend melatonin actions on photoperiodic time measurement and circadian entrainment have emerged. Melatonin is a free radical scavenger and antioxidant and it has a significant immunomodulatory activity, being presumably a major factor in an organism's defense toxic agents and invading organisms. Besides affecting specific receptors in cell membranes to exert its effects, the interaction…

Photic entrainment of the mammalian rhythm in melatonin production.

This review summarizes studies on the photic entrainment of the circadian rhythm in the rat pineal melatonin production, namely of the rhythm in N-acetyltransferase (NAT) activity, and compares the NAT rhythm resetting with preliminary results on the resetting of an intrinsic rhythmicity in the suprachiasmatic nucleus (SCN) of the hypothalamus, namely with the entrainment of the rhythm in the light-induced c-fos gene expression. Phase delaying of the NAT rhythm after various light stimuli proceeds within 1 day with almost no transients, whereas during phase advancing of the rhythm only the mor…

Melatonin in the rat testis: evidence for local synthesis.

The vertebrate pineal gland rhythmically produces melatonin, a hormone involved in regulation of several physiological and behavioral processes. Melatonin is synthesized from serotonin essentially by two enzymatic steps involving N-acetyltransferase (NAT) and hydroxyindole-O-methyltransferase activities. We have previously demonstrated the presence of melatonin binding sites in the rat testes, and an inhibitory effect of melatonin on testicular gonadotrophin-stimulated androgen production. It is unknown whether these effects are mediated by melatonin synthesized locally or by melatonin from pi…

Melatonin: a master hormone and a candidate for universal panacea.

The molecule of melatonin seems to have been evolutionarily conserved. Its presence has been demonstrated in almost all groups of organisms, from plants, protozoa to people. During evolution, melatonin is claimed to have mediated dark adaptation. The universal presence of melatonin may be because it is lipophilic in nature which enables it to cross all biological (lipid membrane) barriers and to diffuse into every compartment of the cell, and because it serves as an antioxidant and is used as a free radical scavenger. In vertebrates, the pineal gland is the single largest source of melatonin p…

Melatonin binding sites and their role in seasonal reproduction.

The pineal gland has a major role in the translation of scotophase duration into a hormonal signal by the hormone melatonin. Animals such as sheep, goats and deer use the seasonal variation of this signal to coordinate reproductive behaviour with the environment. Despite intensive research over more than 30 years the site(s) of action of melatonin and the resultant intracellular responses are still not clear. This review discusses recent work that has localized the site of action of melatonin in sheep using administration into the hypothalamus in vivo as well as studies on putative melatonin r…

Distinct modes of melatonin secretion in normal men.

As for many hormones, melatonin levels in the blood suggest that it is discharged from the pineal gland in a pulsatile manner. Recently, the existence of short-term episodes, superimposed on the circadian pattern of circulating melatonin, has been questioned. Because plasma melatonin levels reflect not only the secretory process, but also the effects of distribution and degradation, secretory rates were estimated from peripheral levels, using a deconvolution procedure. Fourteen healthy volunteers were studied during the night, while sleeping in the dark (2300-0700), and seven of them subsequen…

The effect of estradiol on the production of melatonin in postmenopausal women.

The influence of two routes of estradiol administration on pineal melatonin production in postmenopausal women was investigated. Both transdermal and oral estradiol treatment led to an increase as well as decrease of melatonin production in different patients. The reason why individuals respond either in a stimulatory or inhibitory manner is unknown and requires to be evaluated in further more extensive studies.

[The renal effects of melatonin in intact and epiphysectomized rats].

Epiphysectomy in adult pubescent Wistar rats causes polyuria and Na uresis at the expense of the increased rate of the glomerular filtration and sodium filtration charge. Melatonin in a dose of 4.31 mkM/100 g of the body weight increased the glomerular filtration of both sodium and water and stimulated the sodium tubular transport in rats with intact epiphysis and did not normalize changes of the nephron excretory activity after epiphysectomy.

Thermotrophic effect of melatonin in adrenalectomized and thyroidectomized rats.

A single injection of 25 micrograms melatonin induced hyperthermia in rats. The mechanism(s) by which melatonin induces hyperthermia is not clear. In this study the thermotrophic effects of melatonin were studied in adrenalectomized (ADX) and thyroidectomized (TX) rats to ascertain if melatonin-induced hyperthermia is mediated through these glands, as both these glands play a significant role in mammalian thermoregulation. Melatonin (25 micrograms) was administered as a single subcutaneous injection at 06.00, 12.00, 18.00, and 24.00 h. Body temperatures were recorded, using an anal thermometer…

[Uterine contraction induced by carbachol is inhibited by melatonin].

It has been suggested that the pineal gland has a specific role in the regulation of reproductive functions. Melatonin, secreted by pineal gland, is involved in the control of mammalian reproduction. Previous investigations have show that melatonin reduced the smooth muscle contraction. The purpose of the present study was to investigate the effect of melatonin on the uterine contraction provoked by carbachol. This effect was studied in isolated uterus were taken from Wistar rats pretreated with diethylstilboestrol. Here, we describe the effects of various concentrations of melatonin was found…

The pattern of melatonin in amenorrheic women affected by sterility.

The authors analyzed the pattern of melatonin in the menstrual cycles of six amenorrheic women affected by sterility. Only in one ovulatory cycle did melatonin respect its typical secretory behavior, i.e., nadir in the preovulatory period followed by an increase in the luteal phase, even though melatonin values were higher than normal over the entire cycle. The melatonin pattern in the five cycles showing no evidence of follicular recruitment nor regular formation of a main follicle was different from that seen in an ovulatory cycle. The authors support the concept of a correlation between the…

[The role of melatonin in growth of malignant choroid melanoma].

Increasing evidence of the immunomodulatory and tumor-depressing effects of melatonin prompted us to study altered concentrations of this multifunctional pineal hormone in the aqueous humor of eyes with melanoma. Eyes undergoing cataract surgery served as controls. First, RIA was used for the detection of melatonin (5 melanoma and 8 control eyes). In a second series we used ELISA with a newly developed antibody (6 melanoma and 11 control eyes). Moreover, the supernatant media of cultured melanomas were investigated for melatonin. We cautiously interpret the results as follows: (1) in the morni…
