The X-linked Nsdhl gene encodes a sterol dehydrogenase involved in cholesterol biosynthesis. Mutations in this gene cause the male lethal ph … the gene that encodes the major hedgehog receptor. We demonstrate expression of Ptch1 in allantoic mesoderm of the placenta from wild-type mid-gestation embryos. The evidence suggests that the signaling is induced by Indian hedgehog that is produced by distal (ectoplacental) visceral endoderm cells that migrate into the allantoic mesoderm before embryonic …

OBJECTIVES: The layered structure of the human vocal fold is unique to humans and absent in animals. Previous studies suggested that the mac … eneity observed. Cells in the gorilla macula flavae expressed vimentin, an intermediate filament of mesoderm-derived mesenchymal cells, but not desmin (mesoderm-derived muscle cells) or cytokeratin (ectoderm-derived epithelial cells). SOX17, a marker of endoderm-derived cells, was not expressed. These findings suggest that cells in the gorilla macula flava …

Domestication leads to phenotypic characteristics that have been described to be similar across species. However, this "domestication syndro … more variation in the neural crest-derived parts of the cranium compared with those derived of the mesoderm, in accordance with the "neural crest hypothesis." Our own data are consistent with older literature records, suggesting that although there is evidence for some cranial characteristics of the "domestication syndrome" in rabbits, facial length is no …

Twist transcription factors, members of the basic helix-loop-helix family, play crucial roles in mesoderm development in all animals. Humans have two paralogous genes, TWIST1 and TWIST2, and mutations in each gene have been identified in specific craniofacial disorders. Here, we describe a new clinical entity, Sweeney-Cox syndrome, associated with distinct de novo amino acid substitutions (p.Glu117Val and p.Glu117Gly) at a highly conserved glutamic acid residue located in the basic DNA binding domain of TWIST1, in two subjects with frontonasal dysplasia and additional malformations. Although a…

Neural crest cells, delaminating from the neural tube during migration, undergo an epithelial-mesenchymal transition and differentiate into several cell types strongly reinforcing the mesoderm of the craniofacial body area - giving rise to bone, cartilage and other tissues and cells of this human body area. Recent studies on craniomaxillofacial neural crest-derived cells have provided evidence for the tremendous plasticity of these cells. Actually, neural crest cells can respond and adapt to the environment in which they migrate and the cranial mesoderm plays an important role toward patternin…

Branchial apparatus consists of six pairs of mesodermal arches. Invagination of endoderm from inside and ectoderm from outside are known as pharyngeal pouch and branchial clefts respectively. Incomplete fusion of these structure lead to formation of branchial sinus/ fistula. Anomoly of second arch is commonest and manifests as Branchial sinus. We present such a case in a 18 years old boy.

Ectrodactyly-ectodermal dysplasia-clefting syndrome is a rare congenital anomaly that affects tissues of mesodermal and ectodermal origin. Musculoskeletal involvement frequently requires orthopedic intervention. The authors present a review of the literature pertaining to this rare syndrome as well as a case report of a female patient who exhibited the complete clinical triad. A description of the surgical management of her condition is also presented.

Oculocutaneous albinism and anterior mesodermal dysgenesis are well-known heritable conditions, but their occurrence in association has only been rarely reported. We present cases of three siblings of a family with identical presentation suggesting that this association may be more than just a coincidence. This association is worth noting, as this could be one of the causes of ocular morbidity and poor vision in oculocutaneous albinism.

This case report describes a rare instance of primary orbital teratoma with anophthalmia in a neonate. A 6-day-old female presented with a congenital right orbital swelling and absence of visible ocular structures. MRI revealed a large, well-vascularized orbital mass without intracranial extension, accompanied by malformations in the right cerebral hemisphere. Histopathological examination confirmed a benign, mature/mixed teratoma comprising elements from all three germ layers, including neuroectoderm, mesoderm, and endoderm, with no evidence of malignancy. The patient underwent successful orb…

Odontodysplasia is a rare, non-hereditary developmental anomaly affecting dental tissues derived from both the mesoderm and ectoderm which results in a bizarre clinical and radiographic appearance. Regional odontodysplasia describes the segmental and localized nature of the condition. Odontodysplasia occurring in a single tooth is a rare occurrence. A case of Solitary odontodysplasia in an eleven year and half old male whose chief complaint was the absence of eruption of permanent maxillary right central incisor teeth is presented. Clinical, radiographic and histological findings of a single t…

The vertebrate brain develops in association with neighboring tissues: neural crest, placodes, mesoderm and endoderm. The molecular and evolutionary relationships between the forming nervous system and the other craniofacial structures were at the focus of a recent meeting at the Fondation des Treilles in France. Entitled 'Relationships between Craniofacial and Neural Development', the meeting brought together researchers working on diverse species, the findings of whom provide clues as to the origin and diversity of the brain and facial regions that are involved in forming the 'new head' of v…

Congenital ptosis of the upper lid is classified as a developmental anomaly of the levator muscle. We report the histochemical findings in 38 specimens of levator muscle removed in patients with congenital ptosis and operated on by a resection of the levator muscle. Results demonstrated a close relationship between the presence or the absence of muscular fibres in the specimen and the degree of ptosis and the levator function. We suggest that congenital ptosis may result from a failure of differentiation and/or fusion of the anterior portion of the superior orbital mesoderm complex.

We report the case of a 14-year-old girl with a left atrial myxoma associated with atrial septal defect. Histopathologically, glandular structures were found at the base of the tumor. Immunohistochemical examination of the tumor revealed positive staining of the glands with cytokeratin, epithelial membrane antigen, and carcinoembryonic antigen. The positive staining for epithelial markers suggests that these structures are epithelial and could represent either endodermal heterotopia or multipotential mesoderm retaining the capacity for epithelial expression. The glands should not be confused w…

Teratomas originate from pluripotent germ cells and differentiate into the 3 germ cell layers: endoderm, mesoderm, and ectoderm. Hence, these tumors arise most often in the gonads. Extragonadal teratomas are rare in veterinary medicine. Congenital oropharyngeal teratoma, also known as epignathus, is a neoplasm that has been reported in humans and a few veterinary species. We describe the clinical, gross, cytologic, and histopathologic features of an oropharyngeal teratoma in a neonatal Boer × Nigerian Dwarf goat that died within 4 h of birth, and briefly review extragonadal teratomas in veteri…

Dominant deafness-onychodystrophy (DDOD) syndrome is a rare autosomal dominant disorder caused by mutations in ATP6V1B2 gene. We previously generated an induced pluripotent stem cell (iPSC) line (CPGHi002-A) from a DDOD patient with a heterozygous c.1516 C>T mutation in the ATP6V1B2 gene. Here we genetically corrected the c.1516 C>T mutation in the ATP6V1B2 gene using CRISPR/Cas9 technology to generate an isogenic control, CPGHi002-A-1. The characterization of CPGHi002-A-1 demonstrates normal karyotype, pluripotent state, and potential to differentiate in vitro towards endoderm, mesoderm, and …
