BACKGROUND: Chromosomal aberrations play an important role as prognostic factors in chronic lymphocytic leukemia (CLL). These aberrations are mostly detected by fluorescent in situ hybridization (FISH), as chromosomal banding analysis has been scarce due to low proliferative activity of malignant B-lymphocytes in vitro. In 2006, a new method using stimulation with IL-2 and CpG oligonucleotide DSP30 for metaphase generation in CLL was published [1]. During the last few years, various technologies and applications of fluorescence in situ hybridization (FISH) have been developed. Hybridization on nuclei allows an increase in the resolution of the technique. It also permits the characterization of some chromosomal abnormalities such as trisomies, monosomies or translocations in pathological cells when it is difficult to obtain metaphases. Recently, several methods which extend the chromatin or the DNA molecules have been developed. Such a support increases the FISH resolution to the molecular level.

A cytogenetic and histological study of nine XO/XY or XO/XY/XYY mosaic mice revealed that XO germ cells were selectively eliminated from the spermatogenic epithelium. Although the XO contribution to the bone marrow in seven mice exceeded 50%, in only two cases were significant numbers of dividing XO spermatogonia present. These XO germ cells only occasionally progressed to meiosis and then degenerated prior to first meiotic metaphase. It was concluded that the mouse Y chromosome carries a "spermatogenesis gene" (or genes) which acts autonomously in the germ cells.
