Clinical and MRI correlates in 27 patients with acquired pendular nystagmus.

Clinical and MRI investigations were carried out on 27 patients with acquired pendular nystagmus in an attempt to delineate possible sites of lesions responsible for pendular nystagmus and mechanisms underlying the frequent ocular disconjugacy of this nystagmus. The aetiologies were multiple sclerosis (n = 21), brainstem stroke (n = 3) and other neurological conditions. In at least 59% of the patients, pendular nystagmus appeared > 1 year after the first symptom of the disease. Patients MRIs were characterized by multiple areas of abnormal signal and were analysed statistically to identify are

Development of choline acetyltransferase-immunoreactive neurons in normal and intracranially transplanted retinas in rats.

Retinas from embryonic day 14 (E14) Sprague-Dawley rats were transplanted to the tectum of newborn (P0) recipient rats, and the distribution pattern of choline acetyltransferase immunoreactivity (ChAT-I) in developing transplants was studied and compared with those observed in the retinas of normal developing rats. In normal retinas, ChAT-I cells were first identified in restricted regions in the ganglion cell layer (GCL) at P4, but were found to cover the entire GCL by P6. A second population of ChAT-I cells was detected in the inner nuclear layer (INL) at P8, and they were observed in most p

RBFOX1 Dysfunction Unlocks APOE4-Associated Microglial Genesis and Exacerbates Alzheimer's Pathology in Human Cerebral Organoids.

Alzheimer's disease (AD) pathogenesis is strongly influenced by APOE4, though how cooperative genetic factors modulate this relationship remains unclear. While genomic studies have tentatively linked RBFOX1 to AD susceptibility, its functional synergy with APOE4 has never been experimentally defined. We engineered APOE3 or APOE4 isogenic human cerebral organoids with CRISPR/Cas9-mediated RBFOX1 knockout. Remarkably, RBFOX1 depletion selectively triggered robust microglial generation exclusively in APOE4 organoids. Time-course gene expression revealed that this APOE4-specific effect correlated

Beyond the nerve: Understanding cortical contributions to post-paralytic facial synkinesis.

Patients with post-paralytic facial synkinesis (PPFS) experience a diverse array of neuronal changes from aberrant peripheral neurogenesis to cortical changes. The aim of this review is to utilize a comprehensive search strategy to (1) identify studies using functional magnetic resonance imaging (fMRI) to evaluate PPFS, and (2) synthesize the available evidence for any unifying structural and functional cortical changes which hold promise for future investigation. A search of Pubmed and Embase databases was conducted with a research librarian. Articles were then screened and reviewed independe

A Knock-In Igfn1iCre transgenic mouse line provides partial developmental access to type-7 bipolar cells.

Functional neuronal circuits in the vertebrate retina emerge through coordinated developmental events, yet the timeline by which bipolar cells acquire visual feature selectivity remains unclear. A major barrier is the limited genetic access to bipolar subtypes during early postnatal stages. Recent comprehensive transcriptomic study points to Igfn1 as a molecular marker for type-7 bipolar cells (BC 7), a subtype that exhibits direction-selective glutamate releases in adult. Here, we generated an Igfn1iCre knock-in mouse line and characterized Igfn1-positive cell morphology from postnatal day 4(

Pathogenic variants in BORCS5 cause a spectrum of neurodevelopmental and neurodegenerative disorders with lysosomal dysfunction.

BORCS5 encodes a subunit of the BLOC-One-Related Complex (BORC), which is known to promote anterograde movement and fusion of lysosomes. We identified 16 individuals from 9 families with bi-allelic BORCS5 variants, revealing a spectrum of neurodevelopmental and neurodegenerative phenotypes. Carriers of homozygous protein-truncating variants (PTVs), resulting in complete loss of BORCS5, presented with prenatally lethal arthrogryposis multiplex congenita, brain malformations, and neuropathological evidence of neuroaxonal dystrophy. Individuals with missense or splice-site variants presented diff

Using cortical organoids to understand the pathogenesis of malformations of cortical development.

Malformations of cortical development encompass a broad range of disorders associated with abnormalities in corticogenesis. Widespread abnormalities in neuronal formation or migration can lead to small head size or microcephaly with disorganized placement of cell types. Specific, localized malformations are termed focal cortical dysplasias (FCD). Neurodevelopmental disorders are common in all types of malformations of cortical development with the most prominent being refractory epilepsy, behavioral disorders such as autism spectrum disorder (ASD), and learning disorders. Several genetic pathw

Neural mechanisms distinguishing two types of cooperative problem-solving approaches: An fNIRS hyperscanning study.

Collaborative cooperation (CC) and division of labor cooperation (DLC) are two prevalent forms of cooperative problem-solving approaches in daily life. Despite extensive research on the neural mechanisms underlying cooperative problem-solving approaches, a notable gap exists between the neural processes that support CC and DLC. The present study utilized a functional near-infrared spectroscopy (fNIRS) hyperscanning technique along with a classic cooperative tangram puzzle task to investigate the neural mechanisms engaged by both friends and stranger dyads during CC versus DLC. The key findings

Increasing histone acetylation improves sociability and restores learning and memory in KAT6B-haploinsufficient mice.

Mutations in genes encoding chromatin modifiers are enriched among mutations causing intellectual disability. The continuing development of the brain postnatally, coupled with the inherent reversibility of chromatin modifications, may afford an opportunity for therapeutic intervention following a genetic diagnosis. Development of treatments requires an understanding of protein function and models of the disease. Here, we provide a mouse model of Say-Barber-Biesecker-Young-Simpson syndrome (SBBYSS) (OMIM 603736) and demonstrate proof-of-principle efficacy of postnatal treatment. SBBYSS results

Parietal Gamma Band Oscillation Induced by Self-Hand Recognition.

Physiological studies have shown that self-body images receive unique recognition processing in a wide range of brain areas, from the frontal lobe to the parietal-occipital cortex. Event-related potential (ERP) studies have shown that the self-referential effect on the image of a hand increases P300 components, but such studies do not evaluate brain oscillatory activity. In this study, we aimed to discover the self-specific brain electrophysiological activity in relation to hand images. ERPs on the fronto-parietal midline were elicited by a three-stimulus visual oddball task using hand images:

Spatially informed Bayesian neural network for neurodegenerative diseases classification.

Magnetic resonance imaging (MRI) plays an increasingly important role in the diagnosis and prognosis of neurodegenerative diseases. One field of extensive clinical use of MRI is the accurate and automated classification of degenerative disorders. Most of current classification studies either do not mirror medical practice where patients may exhibit early stages of the disease, comorbidities, or atypical variants, or they are not able to produce probabilistic predictions nor account for uncertainty. Also, the spatial heterogeneity of the brain alterations caused by neurodegenerative processes i

Mapping TRPM7 Function by NS8593.

The transient receptor potential cation channel, subfamily M, member 7 (TRPM7) is a ubiquitously expressed membrane protein, which forms a channel linked to a cytosolic protein kinase. Genetic inactivation of TRPM7 in animal models uncovered the critical role of TRPM7 in early embryonic development, immune responses, and the organismal balance of Zn2+, Mg2+, and Ca2+. TRPM7 emerged as a new therapeutic target because malfunctions of TRPM7 have been associated with anoxic neuronal death, tissue fibrosis, tumour progression, and giant platelet disorder. Recently, several laboratories have identi

A Neural Basis for Contagious Yawning.

Contagious yawning, in which yawning is triggered involuntarily when we observe another person yawn, is a common form of echophenomena-the automatic imitation of another's words (echolalia) or actions (echopraxia) [1]. The neural basis for echophenomena is unknown; however, it has been proposed that it is linked to disinhibition of the human mirror-neuron system [1-4] and hyper-excitability of cortical motor areas [1]. We investigated the neural basis for contagious yawning using transcranial magnetic stimulation (TMS). Thirty-six adults viewed video clips that showed another individual yawnin

Disembodied Mind: Cortical Changes Following Brainstem Injury in Patients with Locked-in Syndrome.

Locked-in syndrome (LIS) following ventral brainstem damage is the most severe form of motor disability. Patients are completely entrapped in an unresponsive body despite consciousness is preserved. Although the main feature of LIS is this extreme motor impairment, minor non-motor dysfunctions such as motor imagery defects and impaired emotional recognition have been reported suggesting an alteration of embodied cognition, defined as the effects that the body and its performances may have on cognitive domains. We investigated the presence of structural cortical changes in LIS, which may accoun

Cntnap4 differentially contributes to GABAergic and dopaminergic synaptic transmission.

Although considerable evidence suggests that the chemical synapse is a lynchpin underlying affective disorders, how molecular insults differentially affect specific synaptic connections remains poorly understood. For instance, Neurexin 1a and 2 (NRXN1 and NRXN2) and CNTNAP2 (also known as CASPR2), all members of the neurexin superfamily of transmembrane molecules, have been implicated in neuropsychiatric disorders. However, their loss leads to deficits that have been best characterized with regard to their effect on excitatory cells. Notably, other disease-associated genes such as BDNF and ERB

Decreased resting-state interhemispheric coordination in first-episode, drug-naive paranoid schizophrenia.

BACKGROUND: Dysconnectivity hypothesis posits that schizophrenia relates to abnormalities in neuronal connectivity. However, little is known about the alterations of the interhemispheric resting-state functional connectivity (FC) in patients with paranoid schizophrenia. In the present study, we used a newly developed voxel-mirrored homotopic connectivity (VMHC) method to investigate the interhemispheric FC of the whole brain in patients with paranoid schizophrenia at rest. METHODS: Forty-nine first-episode, drug-naive patients with paranoid schizophrenia and 50 age-, gender-, and education-ma

Frontal operculum temporal difference signals and social motor response learning.

Substantial experimental evidence supports the theory that the dopaminergic system codes a phasic (short duration) signal predicting the delivery of primary reinforcers, such as water when thirsty, during Pavlovian learning. This signal is described by the temporal difference (TD) model. Recently, it has been suggested that the human dopaminergic system also codes more complex cognitive goal states, including those required for human social interaction. Using functional magnetic resonance imaging (fMRI) with 18 healthy subjects, we tested the hypothesis that TD signals would be present during

Empathy and identification in Von Donnersmarck's The Lives of Others.

Florian Henckel von Donnersmarck's The Lives of Others, set in the German Democratic Republic in 1984, five years before the fall of the Berlin Wall, has been called the first accurate depiction of the psychological terror wielded by the Stasi, the East German secret police, who safeguarded the dictatorship of the proletariat. The film is about the psychological and political transformation of a Stasi officer, Wiesler, who undertakes the surveillance of a prominent playwright and his actress lover. The mechanisms through which Wiesler comes to empathize and identify with the subjects of his in
