Synthesis, Characterization, Biological Evaluation and In Silico Studies of Some New Thiazole Derivatives.

Background/Objectives: The thiazole ring is one of the core structures frequently favored in the design of new active compounds. Methods: A series of thiazole derivatives bearing a benzodioxole scaffold (1-9) were synthesized and evaluated for their antioxidant, anticholinesterase, and antidiabetic activities. Results: Antioxidant assays (ABTS and DPPH) revealed that compounds bearing 4-trifluoromethyl (compound 8) and phenyl (compound 9) moieties exhibited the strongest radical scavenging activities, attributed to efficient hydrogen atom transfer and extended π-conjugation, respectively. Enzy

Single-atom Pt-doped ceria nanozymes mitigate myocardial ischemia reperfusion injury via cardiomyocyte-targeted uptake and suppression of reactive oxygen species.

The primary treatment for myocardial infarction (MI) is restoring blood flow to the obstructed coronary artery. However, this approach can paradoxically generate reactive oxygen species (ROS), leading to secondary ischemia-reperfusion (IR) injury. Multifunctional nanomaterials present a promising alternative for managing IR injury, offering benefits including cost-effectiveness, robust catalytic stability, and customizable properties that surpass traditional antioxidants. This study explores single-atom Pt-doped ceria nanozymes (Pt@CeNZ) with multi-enzyme mimetic functions facilitated by atomi

Microwave Irradiation Pre-Treatment as a Sustainable Method to Obtain Bioactive Hydrolysates from Chicken Feathers.

Chicken feathers constitute a major by-product from the poultry industry, with a potential environmental impact and significant difficulties in their management. This study aimed to develop a sustainable method to hydrolyse chicken feathers and evaluate the effects of microwave (MW) irradiation pre-treatment in the generation of bioactive hydrolysates by simple or sequential hydrolysis with Alcalase. The hydrolysate with MW irradiation pre-treatment and Alcalase (2%, 2 h) (MWA) showed the highest overall antioxidant activity and neprilysin-inhibitory activity (55%), whereas samples without MW

Oral multifunctional gel beads for the targeted treatment of inflammatory bowel disease.

Current IBD therapies face limitations such as systemic toxicity, off-target effects, and microbiota dysbiosis. We developed an oral multifunctional gel bead integrating three innovations: (1) a butyrate-apigenin synergy that inhibits NF-κB via CK2/p65 phosphorylation suppression, activates a PPAR-γ-mediated metabolic shift to β-oxidation, and modulates the microbiota by enhancing Bacteroidetes/suppressing Enterobacteriaceae; (2) an apigenin-butyrate ester prodrug overcoming organoleptic issues while enabling CK2 targeting; and (3) a pH/enzyme-responsive delivery system using the FDA-approved

Tailorable bimetallic nanozyme mitigates intervertebral disc degeneration by inhibiting oxidative stress and inflammageing.

Rationale: Scavenging reactive oxygen species (ROS), modulating extracellular matrix (ECM) anabolism, and preventing senescence of nucleus pulposus cells (NPCs) are crucial factors of treatment approaches for intervertebral disc degeneration (IDD). However, addressing these issues simultaneously has been challenging due to the interactions among the various pathological factors in the disc microenvironment. Methods: Herein, we utilize self-assembly technology and the excellent drug-carrying potential of mesoporous Prussian blue to design a tailorable bimetallic nanozyme platform of a Mn-modifi

Oligomerisation of Ku from Mycobacterium tuberculosis promotes DNA synapsis.

Mycobacterium tuberculosis (Mtb), the causative agent of tuberculosis (TB), is estimated to infect nearly one-quarter of the global population. A key factor in its resilience and persistence is its robust DNA repair capacity. Non-homologous end joining (NHEJ) is the primary pathway for repairing DNA double-strand breaks (DSBs) in many organisms, including Mtb, where it is mediated by the Ku protein and the multifunctional LigD enzyme. In this study, we demonstrate that Ku is essential for mycobacterial survival under DNA-damaging conditions. Using cryogenic electron microscopy (cryo-EM), we so

DNA Gene's Basic Structure as a Nonperturbative Circuit Quantum Electrodynamics: Is RNA Polymerase II the Quantum Bus of Transcription?

Previously, we described that Adenine, Thymine, Cytosine, and Guanine nucleobases were superconductors in a quantum superposition of phases on each side of the central hydrogen bond acting as a Josephson Junction. Genomic DNA has two strands wrapped helically around one another, but during transcription, they are separated by the RNA polymerase II to form a molecular condensate called the transcription bubble. Successive steps involve the bubble translocation along the gene body. This work aims to modulate DNA as a combination of n-nonperturbative circuits quantum electrodynamics with nine Rad

Study of pulsed electrostatic field (PESF) in the perfusion of peripheral tissues: Microangiopathy, nutrition and quality of perceiver life.

Microangiopathy compromises the structural and functional integrity of organs and tissues in patients with type II diabetes mellitus (T2DM) negatively affecting the perceived quality of life. Nitric oxide (NO) is a multifunctional signalling molecule, acting as a vasodilator, neurotransmitter, and modulator of inflammatory processes. Patients with type II diabetes mellitus and chronic kidney disease, controlled from glycaemic status, were treated or not with pulsed electrostatic field (PESF) cycles to evaluate effect on the perfusion of peripheral tissues. Everyone was monitored for the metabo

Upregulation of α-enolase protects cardiomyocytes from phenylephrine-induced hypertrophy.

Cardiac hypertrophy often refers to the abnormal growth of heart muscle through a variety of factors. The mechanisms of cardiomyocyte hypertrophy have been extensively investigated using neonatal rat cardiomyocytes treated with phenylephrine. α-Enolase is a glycolytic enzyme with "multifunctional jobs" beyond its catalytic activity. Its possible contribution to cardiac dysfunction remains to be determined. The present study aimed to investigate the change of α-enolase during cardiac hypertrophy and explore its role in this pathological process. We revealed that mRNA and protein levels of α-eno

ACE2/Ang-(1-7) signaling and vascular remodeling.

The renin-angiotensin system (RAS) regulates vascular tone and plays a critical role in vascular remodeling, which is the result of a complex interplay of alterations in vascular tone and structure. Inhibition of the RAS has led to important pharmacological tools to prevent and treat vascular diseases such as hypertension, diabetic vasculopathy and atherosclerosis. Angiotensin converting enzyme 2 (ACE2) was recently identified as a multifunctional monocarboxypeptidase responsible for the conversion of angiotensin (Ang) II to Ang-(1-7). The ACE2/Ang-(1-7) signaling has been shown to prevent cel

Identification of potent inhibitors against snake venom metalloproteinase (SVMP) using molecular docking and molecular dynamics studies.

Snake venom metalloproteinase (SVMP) (Echis coloratus (Carpet viper) is a multifunctional enzyme that is involved in producing several symptoms that follow a snakebite, such as severe local hemorrhage, nervous system effects and tissue necrosis. Because the three-dimensional (3D) structure of SVMP is not known, models were constructed, and the best model was selected based on its stereo-chemical quality. The stability of the modeled protein was analyzed through molecular dynamics (MD) simulation studies. Structure-based virtual screening was performed, and 15 potential molecules with the highe

Rabies-specific antibodies: measuring surrogates of protection against a fatal disease.

Antibodies play a central role in prophylaxis against many infectious agents. While neutralization is a primary function of antibodies, the Fc- and complement-dependent activities of these multifunctional proteins may also be critical in their ability to provide protection against most viruses. Protection against viral pathogens in vivo is complex, and while virus neutralization--the ability of antibody to inactivate virus infectivity, often measured in vitro--is important, it is often only a partial contributor in protection. The rapid fluorescent focus inhibition test (RFFIT) remains the "go

Oxygen-sensitive mechanisms and protective strategies of metalloenzymes.

The highly reactive metal cofactors enable metalloenzymes to catalyze a wide range of biologically important reactions, including carbon dioxide fixation, nitrogen fixation, and hydrogen evolution, but also render many of them vulnerable to oxygen (O2)-induced damage. O2 sensitivity has long been a major bottleneck limiting the mechanistic study and industrial applications of metalloenzymes. In this review, we systematically analyze the molecular basis of O2 lability and the naturally evolved O2 resistance across five representative O2-sensitive metalloenzyme systems. Our analysis reveals that

Multi-spectroscopic and in silico approaches reveal the α-amylase and α-glucosidase inhibitory mechanism of anacardic acids and their effects on advanced glycation end products.

Anacardic acids (AAs) are a class of alkyl phenols widely distributed in the Anacardiaceae family and known for their diverse biological properties. However, their potential against type 2 diabetes mellitus (T2DM) has never been thoroughly investigated. In this study, a fraction isolated from a Pistacia vera shell extract, enriched in AAs, exhibited potent inhibitory activity against α-amylase (α-Amy) and α-glucosidase (α-Glu). Pure (15:0)-AA and (17:1)-AA also acted as strong inhibitors of both enzymes, displaying IC₅₀ values significantly lower than those of the marketed anti-diabetic drug a

Urtica dioica L. Phytochemistry, Green Extraction Techniques, Molecular Mechanisms, and Gene Expression Modulation: A Comprehensive Review.

Urtica dioica L. (stinging nettle) is a perennial herb with a long ethnomedicinal history and diverse pharmacological potential. This comprehensive review consolidates current knowledge on its phytochemistry, extraction technologies, bioactivities, and molecular mechanisms. However, recent reviews have generally addressed these aspects separately, and an integrated assessment linking green extraction technologies and phytochemical profiles to molecular mechanisms and gene expression modulation is still lacking. Cysteine-rich peptide for chiral self-assembly: a multi-functional antioxidant with novel SH-mediated ROS-responsive disassembly from natural medicine.

Peptide self-assembly is a promising strategy for developing bioactive nanomaterials with improved drug delivery and therapeutic performance. However, artificial design approaches may not fully incorporate natural paradigms and typically avoid cysteine-rich sequences due to their inherent instability. Challenging this convention, we established a natural medicine-inspired discovery pipeline to identify self-assembling thiol-rich peptides. From pangolin scales, we identified a novel natural peptide, Ac-KCSQLNVNCKG, containing two free cysteine thiols that spontaneously self-assembles into chira
