Impaired energy metabolism in skeletal muscle during phosphate depletion.

The effects of phosphate depletion (PD) of 4, 8, and 12 weeks on skeletal muscle energy metabolism were studied in rats fed a phosphate deficiency diet and compared with rats pairfed with a normal phosphate diet. Skeletal muscle biopsy specimens were examined for energy production, transport, and utilization. The results show that already by 4 weeks of PD, the concentration of inorganic phosphorus of the skeletal muscle was significantly reduced and remained low thereafter. There was significant (P less than 0.01) and direct correlation between the cellular inorganic phosphorus and that of ser

Endothelial alterations of skeletal muscle capillaries in childhood myopathies.

Intramuscular blood capillaries in thirteen biopsied specimens obtained from eleven patients with various childhood myopathies were investigated ultrastructurally. The patients were classified into 4 different groups according to the percent distribution of various stages of endothelial alteration; normal endothelial type (E0 Type), slightly abnormal type (E1 Type), moderately abnormal type (E2 Type) and markedly abnormal type (E3 Type). Endothelial types in Duchenne dystrophy progressed from E0 to E1 and further to E2 Type, which kept pace with muscle degeneration. The endothelial types in We

Pathogenesis of skeletal muscle necrosis induced by tarantula venom.

The pathogenesis of myonecrosis induced by venoms of the Arkansas tarantula (Dugesiella hentzi, Girard) and the Honduran tarantula (Aphonopelma spp.) was studied using light and electron microscopy, scanning electron microscopy and x-ray microprobe analysis, and histochemistry. White mice were injected intraperitoneally with a sublethal dose of tarantula venom. Gross examination 24 hr after injection revealed white areas of apparent calcification in the diaphragm muscle. Light microscopic examination at 15 min revealed hypercontracted muscle cells, and necrotic masses containing areas of conde

Nucleotide sequence of the rat skeletal muscle actin gene.

The actins constitute a family of highly conserved proteins found in all eukaryotic cells. Their conservation through a very wide range of taxonomic groups and the existence of tissue-specific isoforms make the actin genes very interesting for the study of the evolution of genes and their controlling elements. On the basis of amino acid sequence data, at least six different mammalian actins have been identified (skeletal muscle, cardiac muscle, two smooth muscle actins and the cytoplasmic beta- and gamma-actins). Rat spleen DNA digested by the EcoRI restriction enzyme contains at least 12 diff

The early effects of ischemia upon skeletal muscle mitochondria.

The effects of early ischemia were studied in the anterior tibial muscle of Sprague-Dawley rats after 2--24 hr of tourniquet compression at the thigh. Ragged-red fibers, moth-eaten fibers, cores and targets were seen in tissue examined by enzyme histochemistry and electron microscopy. Giant mitochondria, abnormalities of cristal arrangement, crystalloids, osmiophilic inclusion bodies and myeloid figures were dominant features of the mitochondrial reaction. The results of this experiment indicate that early ischemia induces a variety of changes described in other neuromuscular conditions such a

Studies of fragmented sarcoplasmic reticulum from human skeletal muscle.

Sarcoplasmic reticulum (SR) vesicles were isolated from muscle biopsies of 4 normal volunteers, a patient with McArdle disease (before and during contracture), and a patient with normokalemic periodic paralysis. Fractions were analyzed for purity by electron microscopy and biochemical analysis of specific marker enzymes. Adenosine triphosphate (ATP)-dependent calcium ion uptake was measured kinetically by the absorbence changes of murexide, a metallochromic indicator of ionized Ca++ concentrations, in the absence of oxalate or other calcium-complexing anions. In these experiments, time resolut

Alteration of skeletal muscle cellular structures by potassium depletion.

After rats had been fed a low-potassium diet for 4 to 8 weeks, skeletal muscle showed ultrastructural changes involving membranous organelles. Mitochondria were often swollen, condensed, or disintegrated. The transverse tubules were disoriented, focally dilated, and tortuous. The sarcoplasmic reticulum showed various degrees of dilatation. Vacuoles of different sizes occurred frequently. Whirls of membranes were closely associated with any of the membranous organelles, especially near an orifice of a transverse tubule. Supplementation of potassium reversed these changes. These findings are ver

Pathology of skeletal muscle in mixed connective tissue disease.

To characterize the pathology of muscle involvement in mixed connective tissue disease (MCTD), skeletal muscle biopsies from 13 patients with MCTD were examined by routine light microscopy, histochemistry, and direct immunofluorescence. The histologic and histochemical changes observed corresponded closely to changes seen in idiopathic polymyositis and the myopathy associated with systemic lupus erythematosus. Eight of 13 cases examined by direct immunofluorescence demonstrated immunoglobulin deposition either within normal appearing vessels, within normal fibers, around or on the sarcoplasmic

Intracellular bicarbonate of skeletal muscle under different metabolic states.

Intracellular bicarbonate of single muscle fibers in vivo was measured by a direct electrometric method simultaneously with the membrane PD in rats under seven different metabolic states. From the measured intracellular bicarbonate values and the PCO2, the bicarbonate equilibrium potential and the intracellular pH were calculated. The mean intracellular [HCO3-] under normal control conditions was 10.3 +/- 0.7 mM (SE). The intracellular bicarbonate fell significantly in both chronic metabolic acidosis and chronic K+ depletion. In contrast, intracellular bicarbonate was elevated in chronic metab

Skeletal muscle necrosis following membrane-active drugs plus serotonin.

Administration of imipramine plus serotonin (5-HT) to rats has been proposed as an animal model of Duchenne muscular dystrophy. We studied the skeletal muscle necrosis produced in male rats given 5-HT after pretreatment with imipramine, other tricyclic antidepressants, or antihistamines, which like the tricyclic antidepressants, can block neuronal reuptake of 5-HT. Following one of these agents plus 5-HT, 20 mg/kg subcutaneously (s.c.), necrosis was more severe in the soleus muscle than the quadriceps. There was no significant difference in the incidence of necrosis in the soleus and quadricep

Effects of thyroid hormone administration on skeletal muscle mitochondria.

The effects of thyroid hormone administration on the levels of a number of mitochondrial markers were measured in skeletal muscle and liver of normal rats. Injection of 18 mug of L-thyroxine (T4) per 100 g body wt every 4th day for 3 wk had no effect on the concentrations of cytochrome c, on citrate synthase activity, or on respiratory capacity of skeletal muscle. Injection of 200 mug of L-triiodothyronine (T3) daily for 5 days, or feeding 23 mg T4 and 7 mg T3/kg of diet for 2 wk, resulted in thyrotoxicosis and large increases in the activity of hepatic alpha-glycerophosphate dehydrogenase and

Txnrd2 loss in skeletal muscle causes muscle atrophy and drives leanness and obesity resistance.

The mitochondrial selenoenzyme thioredoxin reductase 2 (TXNRD2) plays a critical role in redox homeostasis and reactive oxygen species (ROS) scavenging. While heart-specific deletion of Txnrd2 in mice resulted in cardiac dysfunction, TXNRD2 function in skeletal muscle, the major component of lean body mass, remains unclear. In human GWAS the TXNRD2 locus is associated with total lean mass. Here, we show that Txnrd2 muscle-specific knockout (mTKO) induces a lean phenotype characterized by muscle atrophy and diminished adipose tissues. mTKO mice were resistant to weight gain on standard and high

Skeletal muscle Rac1 mediates exercise training adaptations towards muscle glycogen resynthesis and protein synthesis.

Long-term exercise training elicits tremendous health benefits; however, the molecular understanding is incomplete and identifying therapeutic targets has been challenging. Rho GTPases are among the most regulated groups of proteins after exercise in human skeletal muscle, yet, unexplored candidates for mediating the effects of exercise training. We found that the Rho GTPase Rac1 was activated acutely after multiple exercise modalities in human skeletal muscle. Loss of Rac1 specifically in muscle attenuated contraction-induced muscle protein synthesis, diminished improvements in running capaci

Repairing Volumetric Muscle Loss with Skeletal Muscle Units and Hyaluronic Acid Hydrogel in Rats.

Volumetric muscle loss (VML) is characterized as the loss of muscle tissue that exceeds the muscle's self-repair mechanism, resulting in incomplete restoration of muscle mass and function. Existing treatment modalities, including muscle grafts or autologous muscle transfers, are limited by constraints such as tissue availability and donor site morbidity. Moreover, the inadequate recovery of muscle may lead to fibrosis within the VML site, impeding the process of muscle regeneration and resulting in permanent deficits. Emerging therapeutics, such as hydrogels, show promise in addressing the lim

Muscle-homing peptides modified biomimetic curcumin nanoparticles ameliorate skeletal muscle dysfunction in aging mice.

With increasing age, skeletal muscle gradually loses mass and strength, and the risk of falls and fractures escalates among elderly individuals. Inflammation is closely related to age-related muscle atrophy and is the potential target for treating muscular atrophy. Here, biomimetic curcumin nanoparticles (M12MNCs) are prepared via encapsulating curcumin in the skeletal muscle cell membranes modified via muscle-homing peptides (M12) for the treatment of aging related skeletal muscle atrophy. The M12MNCs have good biocompatibility and can be enriched in aging skeletal muscle. After treatment wit

Skeletal muscle-focused guideline development: hierarchical model incorporating muscle form, function, and clinical outcomes.

Sarcopenia, sarcopenic obesity, malnutrition, and cachexia clinical guidelines were created by expert consensus over the past decade. These pathological states all share in common deficits in skeletal muscle mass, and in some cases muscle function, which adversely impact patient outcomes. Early identification is key as some detrimental outcomes are potentially preventable with available treatments. The four guidelines share common design features: patients suspected of having the condition are first screened with a focused clinical history; if positive, the next step is evaluation with either

Skeletal muscle-derived extracellular vesicles transport glycolytic enzymes to mediate muscle-to-bone crosstalk.

Identification of cues originating from skeletal muscle that govern bone formation is essential for understanding the crosstalk between muscle and bone and for developing therapies for degenerative bone diseases. Here, we identified that skeletal muscle secreted multiple extracellular vesicles (Mu-EVs). These Mu-EVs traveled through the bloodstream to reach bone, where they were phagocytized by bone marrow mesenchymal stem/stromal cells (BMSCs). Mu-EVs promoted osteogenic differentiation of BMSCs and protected against disuse osteoporosis in mice. The quantity and bioactivity of Mu-EVs were tig
